Purification, Immobilization and characterization of Lipase Isoenzyme from <i>Aspergillus niger</i>with c8 Magnetic Particles

The purification of a lipase isoenzyme from an Aspergillus niger lipase A is reported in this manuscript. Purification was carried out in a simple adsorption step, in which the lipase was offered at low ionic strength to the commercially available c8 modified magnetic particles, MaKProt c8. When the isoenzyme was desorbed with a 0.2% solution of Triton X-100, the SDS-PAGE gel showed a single pure band with a molecular weight of 35 KDa. The purified fraction showed 66.75-fold purification compared with the crude extract. The pure fraction was characterized along with the crude extract and the lipase adsorbed on the MaKProt c8. The purified and the adsorbed lipase showed better activity for the tested substrates (p-nitrophenyl acetate, decanoate, myristate and palmitate) than the crude extract, the preferred substrates being myristate (26.7 μmol·min-1·mg-1) and decanoate (17.4 μmol·min-1·mg-1), respectively. The temperature and pH profiles showed no change for the three enzymes, the optimum temperature being 37°c and the best pH 7.0.

Objectives:Adolescents using active transport(AT)to school have higher levels of physical activity(PA)compared with motorized transport(MT)*** study compared school day and weekend day PA in adolescents using AT,MT,or combined AT and MT(AT + MT)to travel to ***:Adolescents(n= 314;age:14.7±1.4 years;32.8% boys)from Dunedin(New Zealand)wore an accelerometer for 7 days and completed a self-reported survey regarding mode of transport to school(73 AT,56 AT + MT,and 185 MT).Data were analyzed using t tests,analysis of variance,and χ2 ***:Although the proportion of adolescents meeting PA guidelines significantly differed among transport groups(AT,47.9%;AT + MT,46.4%;MT,33.5%;p=0.048;overall,39.2%),the observed differences were due mainly to *** with MT,AT and AT+MT engaged in more moderate-to-vigorous PA(MVPA)per day(AT:61.2 ± 23.2 min;AT+MT:59.6 ± 21.7 min;MT:52.5 ± 19.6 min;p = 0.004;pchool day and before *** after school(15:00-16:00),AT engaged in significantly more MVPA compared with AT + MT and *** differences in MVPA between the groups were observed in the late afternoon/early evening period during school days or on weekend ***:compared with MT users,adolescent girls using AT or AT + MT accumulated more MVPA during school commute *** + MT to school is also a plausible way to increase adolescent girls’ PA when AT only is not feasible.

We implement a numerical model reported in the literature to simulate the evolution of a galaxy composed of four matter components,such as:a dark-matter halo;a rotating disk of stars;a spherical bulge of stars and a ring of molecular *** show that the evolution of this galaxy model is stable at least for 10 Gyr(Gyr=109 years).We characterize the resulting configuration of this galaxy model by figures of the circular velocity and angular momentum distribution;the tangential and radial components of the velocity;the peak density evolution and the radial density ***,we calculate several models of equal-mass galaxy binary collisions,such as:(i)frontal and(ii)oblique(with an impact parameter),(iii)two models with initial conditions taken from a 2-body orbit and(iv)a very close *** allow comparison with the galaxy model,we characterize the dynamics of the collision models in an analogous ***,we determine the de Vaucouleurs fitting curves of the radial density profile,on a radial scale of 0–100 kpc,for all the collision models irrespective of the pre-collision *** study the radial mass density and radial surface density profiles at a smaller radial scale,0–20 kpc,we use a four-parameter fitting curve.

Using a uniform partitioning of cubic cells,we cover the total volume of aΛcDM cosmological simulation based on *** define a visualization cell as a spatial extension of the cubic cell,so that we collect all simulation particles contained in this visualization cell to create a series of cartesian plots in which the overdensity of matter is clearly *** then use these plots as input to a convolutional neural network(cNN)based on the Keras library and TensorFlow for image *** assign a class to each plot,we approximate the Hessian of the gravitational potential in the center of the cubic *** selected cubic cell is then assigned a label of 1,2 or 3,depending on the number of positive eigenvalues obtained for the Householder reduction of the Hessian *** apply the cNN to several models,including two models with different visualization volumes,one with a cell size of type L(large)and the other with a cell type S(small).A third model combines the plots of the previous L and S cell *** far,we have mainly considered a slice parallel to the XY plane to make the *** last model is considered based on visualizations of cells that also include slices parallel to the ZX and ZY *** find that the accuracy in classification plots is acceptable,and the ability of the models to predict the class works *** results allow us to demonstrate the aim of this paper,namely that the usual cartesian plots contain enough information to identify the observed structures of the cosmic web.

Wind turbine blades have been constantly increasing since wind energy becomes a popular renewable energy source to generate electricity. Therefore, the wind sector requires a more efficient and representative characterization of vertical wind speed profiles to assess the potential for a wind power plant site. This paper proposes an alternative characterization of vertical wind speed profiles based on Ward's agglomerative clustering algorithm, including both wind speed module and direction data. This approach gives a more accurate incoming wind speed variation around the rotor swept area, and subsequently, provides a more realistic and complete wind speed vector characterization for vertical profiles. Real wind database collected for 2018 in the Forschungsplattformen in Nordund Ostsee(FINO) research platform is used to assess the methodology. A preliminary pre-processing stage is proposed to select the appropriated number of heights and remove missing or incomplete data. Finally, two locations and four heights are selected, and 561588 wind data are characterized. Results and discussion are also included in this paper. The methodology can be applied to other wind database and locations to characterize vertical wind speed profiles and identify the most likely wind data vector patterns.

High surface area mixed oxide 8.7%NiO-ceO_(2) nanoparticles(122 m2/g;6-7 nm)were prepared using a reversed microemulsion method,and were tested for dry methane reforming(DRM).The catalytic activity of these nanoparticles remains stable under the severe conditions of DRM(700℃),and they show better carbon resistance than conventional NiO-ceO_(2) catalysts prepared without control of the *** activity and selectivity of nanoparticles and reference catalyst are similar,but nanoparticles reduce the accumulation of carbon by 63%during the DRM tests,which is a key feature for this *** and H_(2)-TPR suggest that the improved carbon resistance of the nanoparticles is due to the better interaction and cooperation between NiO and ceO_(2) mixed *** nanoparticles,the participation of cerium cations in the redox processes taking place during DRM stabilizes cationic species of *** the contrary,the catalyst prepared without control of the size suffers segregation of Ni during DRM reaction,and segregated Ni explains the higher catalytic formation of carbon.

Background:Athletes tend to have better visuo-motor performance than do sedentary ***,several basic visual-function and perceptual parameters remain unexplored to *** this study,we investigated whether differences exist in visual function,performance,and processing between basketball players and individuals without a sport-involvement ***:A total of 33 healthy men with no visual impairment or pathology were divided into 2 groups,depending on the involvement in sport(semi-professional basketball players and sedentary individuals).We tested their baseline heart-rate variability in the resting position apart from subjective questionnaires to determine their physical fitness level,and we checked their visual function,performance,and processing through an extended battery of optometric ***:The 2 groups differed in resting heart-rate variability parameters(pconfirming their dissimilarities in regular time practising sports per *** basketball players showed a closer breakpoint and recovery nearpoint of convergence,a higher fusional-vergence rate,better discriminability halos,and better eye-hand coordination(all p valuesconclusion:These results show evidence that athletes,basketball players in this case,exhibit better performance in several visual abilities in comparison to a group of individuals without sporting backgrounds,suggesting an improvement due to the systematic involvement of those skills during basketball practice.

Enzymatic hydrolysis of proteins is a breakdown process of peptide bond in proteins,releasing some peptides with potential biological *** studies on enzymatic hydrolysis of whey proteins have not identified the complete peptide profiles after *** this study,we reconstructed a profile of peptides from whey hydrolysates with two enzymes and different processing *** also developed an ensemble machine learning predictor to classify peptides obtained from whey hydrolysis.A total of 2572 peptides were identified over three process conditions with two enzymes in duplicate.499 peptides were classified and chosen as potential antioxidant peptides from whey *** peptides classified as antioxidants in the hydrolysates had a proportion of 13.1%-24.5%regarding all peptides *** results facilitate the selection of promising peptides involved in the antioxidant properties during the enzymatic hydrolysis of whey proteins,aiding the discovery of novel antioxidant peptides.
