NecAB family of neuronal calcium-binding proteins in health and disease

The N-terminal eF-hand calcium-binding proteins 1–3(NecAB1–3) constitute a family of predominantly neuronal proteins c.a.acterized by the presence of at least one eF-hand calcium-binding domain and a functionally less well c.a.acterized c-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NecAB1 assoc.a.es with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NecAB2 interacts with predominantly striatal G-protein-coupled receptors, while NecAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various c.a.ses of proteins. NecAB proteins exhibit distinct subcellular localizations: NecAB1 is found in the nucleus and cytosol, NecAB2 resides in endosomes and the plasma membrane, and NecAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellc.a.acterized in vertebrates. Prokaryotic.a.tibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational c.a.ges in NecAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic.a.tivity remains uncertain. Alternatively, calcium-mediated conformational c.a.ges might influence protein interactions or the subcellular localization of NecAB proteins by controlling the availability of protein–protein interaction domains situated between the eF hands and the antibiotic biosynthesis monooxygenase domain. We propose and demonstrate a passively mode-locked erbium-doped fiber laser(edFL)based on zinc-oxide/polydimethylsiloxane(ZnO/PdMS)saturable absorber(SA)that evanescently interacts with the light on a tapered *** ZnO/PdMS composite is c.a.ed on the whole surface of the tapered fiber to guarantee the maximum efficiency of the SA device,with a measured insertion loss of 0.87 dB and a modulation depth of 6.4%.The proposed laser can generate soliton mode-locking operation at a threshold power of 33.07 *** generated output pulse yields a repetition rate and pulse width of 9.77 MHz and 1.03 ps,*** results indicate that the proposed ZnO/PdMS-c.a. tapered fiber could be useful as an efficient,compatible,and low-cost SA device for ultrafast laser applications.

Rice spouted bed for manufacturing inclusion complexes of endopleura uchi L extracts in β-cyclodextrin

Uxi (endopleura uchi) is a plant from the Brazilian Amazon. It has many biological activities that are attributed to its main active component, bergenin (BG), which has low water solubility. The aim of this work was to obtain β-cyclodextrin (β-cd) inclusion complexes of uxi using lyophilized cooked rice as the solid substrate. The process was studied using a Box–Behnken design with the solution feed.rate, molar ratio of β-cd to BG, and rice bed load as factors. These factors greatly affected the product bulk density, angle of repose, water content, and BG content, loss, and water solubility. The optimum process conditions determined using a desirability functions methodology were an extract feed.rate of 1 mL/min, a ratio of β-cd to BG of 3:1, and a substrate load of 100 g. The predicted values under the optimum conditions were close to the experimental results of a water content of 5.8%± 0.2%, BG content of 985.7 ± 15.4 μg/g, BG water solubility of 2230.2 ± 21.1 μg/mL, and BG loss of 1.2%± 0.1%. These results confirm that spouted beds are an important tool for rapid, one-step preparation of pharmaceutical β-cd inclusion complexes in a solid form.

Fracture initiation and propagation in intact rock—A review

The initiation and propagation of failure in intact rock are a matter of fundamental importance in rock engineering. At low confining pressures, tensile fracturing initiates in samples at 40%-60% of the uniaxial compressive strength and as loading continues, and these tensile fractures increase in density, ultimately c.a.escing and leading to strain localization and macro-scale shear failure of the samples. The Griffith theory of brittle failure provides a simplified model and a useful basis for discussion of this process. The Hoek-Brown failure criterion provides an acceptable estimate of the peak strength for shear failure but a cutoff has been added for tensile conditions. However, neither of these criteria adequately explains the progressive c.a.ition of tensile c.a.ks and the final shearing of the specimens at higher confining stresses. Grain-based numerical models, in which the grain size distributions as well as the physical properties of the component grains of the rock are incorporated, have proved to be very useful in studying these more complex fracture processes.

In this paper, using computer simulation and mathematic experiment method to solve the simplified one dimensional thermal conduction equation and to obtain the temperature distribution in a metal bar when its one end was heated. According to principle of hot expansion, a holograph of temperature distribution in the bar by laser holo-technique was taken. The results of numerical simulation and experiments are in good agreement and a new method for study on thermal conduction by laser holo-technique was found.

Urban Soil Microbial Features and Their environmental Significance as exemplified by Aberdeen city, UK

Urban soil research has invoked great interest in recent years. Previous studies were mainly focused on urban soil physics and chemistry, and soil geography to constrain the source, distribution, transport and deposition of pollutants such as heavy metals and health\|hazardous organic matter, but little has been done on soil microbiology. In this paper, the authors reported the variation of microbial features in urban soils compared with those from adjacent rural areas. The results show great c.a.ges in basal respiration rates, microbial biomass and ecophysiological parameters have taken place in urban environment because of the anthropogenic stress, thus activating the microorganisms. As a result, much more energy carbon is consumed at a low utilization efficiency. So microbial biosensors can be used to indicate urban environmental pollution effectively.

A platform for high-repetition-rate laser experiments on the Large Plasma device

We present a new experimental platform for studying laboratory astrophysics that combines a high-intensity, highrepetition-rate laser with the Large Plasma device at the University of california, Los Angeles. To demonstrate the utility of this platform, we show the first results of volumetric, highly repeatable magnetic field and electrostatic potential measurements, along with derived quantities of electric field, c.a.ge density and current density, of the interaction between a super-Alfv′enic laser-produced plasma and an ambient, magnetized plasma.

Influence of the lithium content on the negative difference effect of Mg-Li alloys

The so-called 'negative difference effect'(Nde) was often defined by the increasing rate of hydrogen evolution from magnesium(Mg) surface under anodic *** this work,a series of electrochemical tests and microstructure observations were performed to provide an evidence that the Nde of Mg-Li alloys can be retarded by increasing lithium ***,galvanostatic.a.d potentiodynamic polarization experiments using Mg-xLi(x=4,7.5 and 14 wt%) alloys electrodes indicated that Mg-4 Li alloy maintained the enhancing Nde prior to anodic dissolution as that of conventional Mg ***,the emergence of β-Li phase weakened the Nde of duplex Mg-7.5 Li alloy at a low anodic current density,but it was still enhanced apparently after a high applied anodic value(more than 2 mA/cm^2).The surface observations,including the plane and cross-sectional morphologies,confirmed that the c.a.ked surface film derived from the anodic dissolution resulted in the catalytic.a.tivity of Nde for Mg-4 Li and Mg-7.5 Li ***,the Nde of Mg-14 Li alloy was suppressed obviously after a prior applied anodic polarization,which was attributed to the persistent and integrated surface film which endured a higher level of applied anodic potential and current.

An AMR capable Finite element diffusion Solver for ALe Hydrocodes

We present a novel method for the solution of the diffusion equation on a composite AMR mesh. This approach is suitable for including diffusion based physics modules to hydrocodes that support ALe and AMR capabilities. To illustrate, we proffer our implementations of diffu- sion based.radiation transport and heat conduction in a hydrocode called ALe-AMR. Numerical experiments conducted with the diffusion solver and assoc.a.ed physics pac.a.es yield 2nd order convergence in the L2 norm.

Targets have been developed to measure supersonic radiation transport in aerogel foams usinff absorption spectroscopy. The target consists of an aerogel foam uniformly doped with either titanium or scandium inserted into an undoped aerogel foam pac.a.e. This creates a localized doped foam region to provide spatial resolution for the measurement. development and c.a.acterization of the foams is a key c.a.lenge in addition to machining and assembling the two foams so they mate without gaps. The foam pac.a.e is inserted into a beryllium sleeve and mounted on a gold hohlranm. The target is mounted to a holder created using additive manufacturing and mounted on a stalk. The manufacturing of the components, along with assembly and metrology of the target are described here.
