Leukoc.te adhesion defic.enc. （LAD） I is a disorder c.used due to mutations in a gene （ITGB2） loc.ted on c.romosome 21 and enc.des the β2 subunit of the leukoc.te integrin molec.les. This leads to defec.s in the adhesion of leukoc.tes on endothelial c.lls whic. further leads to rec.rrent mic.obial infec.ions due to a dec.ease in the immune response. Base Exc.sion Repair Mec.anism （BER） is instrumental in repairing damaged DNA by removing mutated/ damaged bases. We have proposed a hypothesis for the treatment of LAD I by making use of the proteins/enzyme c.mplexes responsible for base exc.sion repair mec.anism be introduc.d into the leukoc.tes via liposomes. This will target the mutated gene in the leukoc.tes （mostly neutrophils） and DNA repair will oc.ur. The liposomes c.n be introduc.d into the patients via intravenous methods.

BAc.GROUND： Marine ac.inomyc.tes are effic.ent produc.rs of new sec.ndary metabolites that show different biologic.l ac.ivities, inc.uding antibac.erial, antifungal, antic.nc.r, insec.ic.dal, and enzyme inhibition ac.ivities. METHODS： The morphologic.l, physiologic.l, and bioc.emic.l properties of the strain Streptomyees sp. VITPSA were c.nfirmed by c.nventional methods. Antibac.erial, anti-oxidant, anti-inflammatory, anti-diabetic. and c.totoxic.ac.ivities of Streptomyc.s sp. VITPSA extrac. were determined. The media were optimized for the produc.ion of sec.ndary metabolites. c.arac.erization and identific.tion of sec.ndary metabolites were c.nduc.ed by high-performanc. liquid c.romatography, gas c.romatography-mass spec.rosc.py, and Fourier transform infrared spec.rosc.py analysis. RESULTS： The strain showed signific.nt antibac.erial, anti-oxidant, and c.totoxic.ac.ivities, moderate anti-inflammatory ac.ivity, and no satisfac.ory anti-diabetic.ac.ivity. The ethyl ac.tate extrac. of Streptomyc.s sp. VITPSA showed maximum antibac.erial ac.ivity against two gram-positive and gram-negative bac.eria at 0.5 mg/mL. The antioxidant potential of the c.ude extrac. exhibited strong reduc.ng power ac.ivity at 0.5 mg/mL with 95.1% inhibition. The c.totoxic.effec. was found to be an Ic.o of 50 gg/mL on Mc.-7 c.ll lines. Experimental design of optimization by one-fac.or analysis revealed the most favorable suc.ose, yeast extrac., pH （7.25）, and temperature （28~c. c.nditions for the effec.ive produc.ion of sec.ndary metabolites. c.Nc.USION： This study revealed that Streptomyc.s sp. VITPSA is an exc.llent sourc. of sec.ndary metabolites with various bioac.ivities.

The c.rrent work was attempted to isolate and c.arac.erize the serratiopeptidase produc.ng Serratia sp. Among the 10 bac.erial isolates 7 strains were identified as Serratia sp. Out of 7 strains one showed potent proteolytic.ac.ivity and selec.ed for further studies. Based on the morphologic.l, bioc.emic.l and molec.lar c.arac.erization, the potent isolate （RH03） was identified as Serratia marc.sc.ns （GenBank ac.ession number： Kc.61637） and the strain was designated as Serratia marc.sc.ns VITSD2. The produc.ion of serratiopeptidase was c.rried out in tryptic.se soya broth and the enzyme was partially purified using ammonium sulfate prec.pitation and dialysis. The spec.fic.ac.ivity was determined by c.sein hydrolysis assay and was found to be 12.00, 21.33, and 25.40 units/rag for c.ude, prec.pitated and dialysed samples. The molec.lar weight of the protease was determined by SDS-PAGE and it was found to be 50 kDa. The antibac.erial ac.ivity of the produc.d serratiopeptidase showed moderate ac.ivity against Pseudomonas aeruginosa MTc. No. 4676 （12 mm） and Esc.eric.ia c.li MTc. No. 1588 （15 mm）.

BAc.GROUND： Serratia is one of the most important groups of bac.eria whic. produc.s proteolytic.enzymes effec.ively and known to possess anti-inflammatory properties. The main foc.s of the c.rrent study was to extrac. the enzyme serratiopeptidase and pigment prodigiosin from Serratia masc.sc.ns. Prodigiosin is a red c.lored pigment produc.d by the bac.erium Serratia marc.sc.ns. It is emerging as a valuable molec.le bec.use of its large applic.tions. It has already been proved that pigmented strain of Serratia marc.sc.ns is less virulent than non-pigmented strains. Moreover the strain we have obtained is from farm soil whic. indic.tes that prodigiosin produc.ion c.n be c.rded safely using this strain. METHODS： In the present study, the isolate VITASP strain was c.nfirmed by morphologic.l, bioc.emic.l and molec.lar studies. The enzyme and pigment were analyzed for anti-oxidant, anti-inflammatory and c.totoxic.properties. RESULTS： The isolate was further c.nfirmed and identified as Serratia marc.sc.ns with 99% similarity. The extrac.ed pigment showed potent radic.l sc.venging effec. with 86% and the enzyme was found to inhibit 83%, whic. was signific.nt in c.mparison to asc.rbic.ac.d standard. The in vitro anti-inflammatory effec. of pigment in c.ntrolled experimental c.nditions revealed its protec.ion at 88% and the enzyme with 90%. Aspirin was used as the referenc. drug. The present findings exhibited a c.nc.ntration dependent inhibition. The c.totoxic.bioassay of pigment showed the Ic.o value as （50） ~tg/mL with 63% c.totoxic.ty whic. was statistic.lly signific.nt c.mpared to positive c.ntrol. c.Nc.USIONS： Therefore, it appears to be an essential remedial and applic.tion researc.. It may turn out to be highly benefic.al to mankind in solving many problems assoc.ated with human health.

BAc.GROUND： Nattokinase （NK） is a serine protease enzyme of the subtilisin family. It exhibits a strong fibrinolytic.ac.ivity. The fibrinolytic.enzymes from Bac.llus sp. have attrac.ed interest as thrombolytic.agents bec.use of their effic.enc. in the fibrinolytic.proc.ss inc.uding plasmin ac.ivation. METHODS： In the present study, VIT garden soil was c.llec.ed and subjec.ed to isolation proc.ss in order to sc.een for the NK produc.ion. Sc.eening for NK enzyme was performed by radial c.seinolytic.assay. The produc.ion of NK enzyme was done in two different produc.ion medium for c.mparative studies. The NK enzyme was purified by gel permeation c.romatography. The ac.ivity of the purified NK was c.ec.ed by c.ot lysis and c.sein digestion assay. To investigate the struc.ural basis of NK and fibrinogen interac.ion and also to identify the best binding mode, molec.lar dynamic. and doc.ing studies were performed. RESULTS： Based on the morphologic.l and bioc.emic.l c.arac.erization, the isolate was identified as Bac.llus sp. The overall purific.tion fold of NK was about 3 with the spec.fic.ac.ivity of 664U/mg and 9.9% yield. Homogeneity of the purified enzyme was analyzed and c.nfirmed by the single band obtained in SDS-PAGE. Molec.lar weight of the purified protease was estimated as 25 kDa. Purified NK enzyme exhibited 97% of effec.ive c.ot lysis ac.ivity. The NK was doc.ed in to the knob region of the fibrinogen at its binding site using Doc. server. A total of 26 residues of fibrinogen and 29 residues of NK c.nstitute the interfac. region. However, 9 residues offibrinogen （THR238, MET264, LYS266, ARG275, THR277, ALA279, ASN308, MET310, and LYS321） and 8 residues ofNK （GLY61, SER63, THR99, PHE189, LEU209, TYR217, ASN218, and MET222） are involved in intac. binding. c.Nc.USIONS： A signific.nt amount of NK enzyme was obtained from Bac.llus sp. This study investigates the relationship between c.rc.lation patterns and austral summer temperature anomalies in southern Afric.. The results show that the formation of c.ntinental lows tends to inc.ease the thic.ness of the lower atmosphere. Further, the distinc. variabilities of high and low pressure under the c.rc.lation types, influenc. air mass advec.ion from the adjac.nt oc.ans, as well as atmospheric.stability over land. Stronger antic.c.onic.c.rc.lation at the western branc. of the Masc.rene high-pressure system enhanc.s the low-level c.ld air advec.ion by southeast winds,dec.eases the thic.ness, and lowers the temperature over a majority of the land in southern Afric.. c.nversely, a weaker Masc.rene High, c.upled with enhanc.d c.c.onic.ac.ivity in the southwest Indian Oc.an inc.eases low-level warm air advec.ion and inc.eases temperature anomalies over vast regions in southern Afric.. The ridging of a c.osed South Atlantic.antic.c.one at the southern c.ast of southern Afric. results in c.lder temperatures near the tip of southern Afric. due to enhanc.d low-level c.ld air advec.ion by southeast winds. However, when the ridge is weak and westerly winds dominate the southern c.ast of southern Afric., these areas experienc. temperature inc.eases. The northward trac. of the Southern Hemisphere mid-latitude c.c.one, whic. c.n be linked to the negative Southern Annular Mode, reduc.s the temperature in the southwestern part of southern Afric.. Also, during the analysis period, El Ni?o was assoc.ated with temperature inc.eases over the c.ntral parts of southern Afric.;while the positive Indian Oc.an dipole was linked to a temperature inc.ease over the northeastern, northwestern, and southwestern parts of southern Afric..

BAc.GROUND： The c.rrent study demonstrated the possibility of statistic.l design tools c.mbination with c.mputational tools for optimization of fermentation c.nditions for enhanc.d fibrinolytic.protease produc.ion. METHODS： The effec.s of using different c.rbon and nitrogen sourc.s for protease produc.ion by Streptomyc.s radiopugnans_VITSD8 were examined by a full fac.orial design method. The inc.bation time, temperature, pH of the medium, and RPM were assessed by the predic.able one fac.or at a time （OFAT） method. Optimization was c.rried out using starc. and oat meal as c.rbon sourc., nitrogen sourc. as peptic.and malt extrac. using Frac.ional Fac.orial Design （FFD）. The analysis was further c.ntinued for medium volume, temperature, initial medium pH, inoc.lum c.nc.ntration, high determination c.-effic.ent as （R＇-0.965）, and lower determination c.-effic.ent of variation （c.-8.19%）, whic. defines a reliable and ac.urate experimental value. RESULTS： Analysis of varianc. by the fixed slope effec. by temperature and starc.; temperature and L-aspargine, temperature and oat meal, temperature and peptic.extrac.s, temperature and pH, temperature and duration of inc.bation were more vital for protease produc.ion at an interac.ive level. Response surfac. plots revealed that temperature, starc., and peptic.extrac.s affix c.itic.l c.nc.rning in temperature. Programming estimated a 28% inc.ease in protease produc.ion. Inc.bation temperature and medium volume portrayed extreme impac. among all fac.or. Starc., peptic.and temperature play an important regulatory role in protease produc.ion. Optimium temperature for protease produc.ion was 33~c. The ratio of c.rbon and nitrogen sourc.s and pH were the major regulatory fac.ors in protease produc.ion by Streptomyc.s radiopugnans_VITSD8. It demonstrated a 4% noteworthy c.ange in c.ndition. c.Nc.USION： Among all the selec.ed parameters, temperature was the most intuitive fac.or, demonstrating a notable c.nnec.ion with the type of medi
