Involvement of Lipid r.fts and c.llular.Ac.in in Ac.NPV GP64 Distr.bution and Vir.s Budding

GP64 is the major.envelope glyc.pr.tein assoc.ated with the budded vir.s (BV) of Autogr.pha c.lifor.ic. nuc.eopolyhedr.vir.s (Ac.NPV) and is essential for.attac.ment and budding of BV par.ic.es. c.nfoc.l mic.osc.py and flotation assays established the pr.senc. of lipid r.ft domains within the plasma membr.nes of Ac.NPV-infec.ed Sf9 c.lls and suggested the assoc.ation of GP64 with lipid r.fts dur.ng infec.ion. GP64 and filamentous ac.in (F-ac.in) wer. found to c.-loc.lise at the c.ll c.r.ex at 24 and 48 hpi and an additional r.str.c.ur.ng of F-ac.in dur.ng infec.ion was visualised, r.sulting in a str.ngly polar.sed distr.bution of both F-ac.in and GP64 at the c.ll c.r.ex. Depletion of F-ac.in, ac.ieved by tr.atment of Sf9 c.lls with latr.nc.lin B (LB), r.sulted in the r.distr.bution of GP64 with signific.nt c.toplasmic.aggr.gation and r.duc.d pr.senc. at the plasma membr.ne. Tr.atment with LB also r.sulted in r.duc.d pr.duc.ion of BV in Sf9 c.lls. Analysis of vir.s gene tr.nsc.iption c.nfir.ed this r.duc.ion was not due to dec.eased tr.ffic.ing of nuc.eoc.psids to the nuc.eus or.to dec.eased pr.duc.ion of infec.ious pr.geny nuc.eoc.psids. r.duc.d BV pr.duc.ion due to a lac. of GP64 at the plasma membr.ne of Ac.NPV-infec.ed Sf9 c.lls tr.ated with LB, suggests a key r.le for.F-ac.in in the egr.ss of BV.

AIM： To test the hypothesis that c.lc.um sensing r.c.ptor.（c.Sr. polymor.hisms ar. assoc.ated with c.r.nic.panc.eatitis （c.）, and to deter.ine whether.ser.ne pr.tease inhibitor.Kazal 1type （SPINK1） N34S or.alc.hol ar. nec.ssar. c.-fac.or. in its etiology. METHODS： Initially, 115 subjec.s with panc.eatitis and 66 c.ntr.ls wer. evaluated, of whom 57 patients and 21 c.ntr.ls wer. pr.deter.ined to c.r.y the high-r.sk SPINK1 N34S polymor.hism. We sequenc.d c.Sr.gene exons 2, 3, 4, 5 and 7, ar.as c.ntaining the major.ty of r.por.ed polymor.hisms and novel mutations. Based on the initial r.sults, we added 223 patients and 239 c.ntr.ls to analyze thr.e c.mmon nonsynonymous single nuc.eotide polymor.hisms （SNPs） in exon 7 （A986S, r.90G, and Q1011E）. r.SULTS： The c.Sr.exon 7 r.90G polyr.or.hism was signific.ntly assoc.ated with c. （Or. 2.01; 95% c., 1.12-3.59; P = 0.015）. The assoc.ation between c.Sr.r.90G and c. was str.nger.in subjec.s who r.por.ed moder.te or.heavy alc.hol c.nsumption （Or. 3.12; 95% c., 1.14-9.13; P = 0.018）. Ther. was no assoc.ation between the var.ous c.Sr.genotypes and SPINK1 N34S in panc.eatitis. None of the novel c.Sr.polymor.hisms r.por.ed fr.m Ger.any and India was detec.ed. c.Nc.USION： Our.United States-based study c.nfir.ed an assoc.ation of c.Sr.and c. and for.the fir.t time demonstr.ted that c.Sr.r.90G is a signific.nt r.sk fac.or.for.c..
