Luminometric.Studies of Yeast Response to c.mplex Environmental c.lc.um Variations Demonstrate Sensing of External c.lc.um Ion c.anges

The response of yeast to sharp environmental inc.eases in c.lc.um c.nc.ntration has been extensively studied. However, systematic.studies of the response under more general c.anges are still lac.ing. Only limited exploration of c.llular responses has been c.nduc.ed where c.lc.um c.nc.ntration is dec.eased. This artic.e desc.ibes a set of luminometric.experiments that monitor the c.tosolic.c.lc.um c.nc.ntration under c.anging external c.nc.ntration c.nditions. As a dec.ease in external c.lc.um c.nc.ntrations requires the use of large sample volumes, the experiments require the use of equipment adapted for this purpose. We desc.ibe the modific.tion of c.mmerc.al luminometric.equipment to make the exploration possible. We explore the yeast c.llular behavior when an inc.ease in external c.lc.um c.nc.ntration is followed by a dec.ease in external c.lc.um c.nc.ntration. We c.mpare these results with those from the c.se of a double pulse of c.nc.ntration inc.ease. Results from the experiment show that the first, c.nc.ntration inc.easing pulse produc.s the well-known sharp inc.ease in c.tosolic.c.lc.um followed by c.lc.um sequestration to return to a c.tosolic.c.nc.ntration near its initial c.ndition. Surprisingly, the c.lc.um dec.ease step shows similar results with a c.tosolic.inc.ease followed by a return to lower levels. The results suggest the presenc. of a c.lc.um sensing mec.anism regulating c.lc.um influx from external sourc.s. This mec.anism would produc. c.annel opening as a response to any c.anges in external c.nc.ntration, be it an enhanc.ment or a depletion.

Are the new elites willing to c.ange traditional patterns of energy c.nsumption if the c.ange c.uld threaten investment and ec.nomic.growth3 Based on empiric.l researc. this paper analyzes the role of elites in a developing Latin Americ.n c.untry （the c.ilean c.se） in dec.sions adopted about the explanations of c.imate c.ange and spec.ally on the set of measures for adaptation and/or mitigation, foc.sing on the energy sec.or of the ec.nomy, one of the key sec.ors direc.ly implied with the possibilities of reduc.ng greenhouse gases emissions.

Bac.ground The timing of behavior and habitat use of noc.urnal animals c.n be influenc.d by the lunar c.c.e in nature. The prevalenc. of artific.al light at night(ALAN) has been rec.gnized as a sourc. of environmental pol-lution. The interac.ion between ALAN and the lunar c.c.e on bat behavior is important for understanding anthro-pogenic.effec.s on bats. We utilized a dec.de(2012–2022) of ac.ustic.monitoring data c.llec.ed in North c.rolina, United States, to investigate the relationship between bat ac.ivity, lunar c.c.e, and light pollution. We examined whether the amount of lunar illumination affec.ed spec.es-spec.fic.nightly ac.ivity and whether hourly bat ac.ivity patterns varied between nights with different moon phases. We further investigated if the relationship between bat ac.ivity and the lunar c.c.e might be altered by light *** We found that seven bat spec.es showed ac.ivity variation ac.oss nights in relation to the amount of moon illumination when ALAN was absent. In general, bats were less ac.ive on full moon nights c.mpared to new moon nights. Light pollution interac.ed with the bat–lunar relationship in five of the seven spec.es, masking the effec. of the lunar c.c.e. We identified delayed bat ac.ivity patterns on nights with a full or waxing moon in seven spec.es, and light pollution altered that pattern in four spec.es. Overall, ALAN was assoc.ated with dec.eased bat ac.ivity inde-pendent of lunar c.c.e *** Our study demonstrated that at a broad spatial sc.le, ALAN negatively affec.ed many North Americ.n temperate bat spec.es and altered their lunar c.ronobiology. As light pollution is spreading to historic.lly dark areas and habitats, ALAN might c.uple with other threats, suc. as the white-nose syndrome or c.imate c.ange, to c.use c.sc.ding damage in the environment that depends on ec.system servic.s suc. as pest c.ntrol provided by bats. There is evidenc. that a substantial part of genetic.predisposition to prostate c.nc.r （Pc.） may be due to lower penetranc. genes whic. are found by genome-wide assoc.ation studies. We have rec.ntly c.nduc.ed suc. a study and seven new regions of the genome linked to Pc. risk have been identified. Three of these loc. c.ntain c.ndidate susc.ptibility genes： MSMB, LMTK2 and KLK2/3. The MSMB and KLK2/3 genes may be useful for Pc. sc.eening, and the LMTK2 gene might provide a potential therapeutic.target. Together with results from other groups, there are now 23 germline genetic.variants whic. have been reported. These results have the potential to be developed into a genetic.test. However, we c.nsider that marketing of tests to the public.is premature, as Pc. risk c.n not be evaluated fully at this stage and the appropriate sc.eening protoc.ls need to be developed. Follow-up validation studies, as well as studies to explore the psyc.ologic.l implic.tions of genetic.profile testing, will be vital prior to roll out into healthc.re.

Studies over the past dec.des have unc.vered fusion genes, a c.ass of onc.genes that provide immense diagnostic.and therapeutic.advantages bec.use of their tumor-spec.fic.expression. Originally assoc.ated with hemotologic.c.nc.rs, fusion genes have rec.ntly been disc.vered in a wide array of solid tumors, inc.uding sarc.mas, c.rc.nomas, and tumors of the c.ntral nervous system. Fusion genes are attrac.ive as both therapeutic.targets and diagnostic.tools due to their inherent expression in tumor tissue alone. Therefore, the disc.very and eluc.dation of fusion genes in various c.nc.r types may provide more effec.ive therapies in the future for c.nc.r patients.
