The mountains of Southwest c.ina c.mprise a signific.nt large mountain range and biodiversity hotspot imperiled by global c.imate c.ange.The high spec.es diversity in this mountain system has long been attributed to a c.mplex set of fac.ors,and rec.nt large-sc.le mac.oevolutionary investigations have plac.d a broad timeline on plant diversific.tion that stretc.es from 10 million years ago(Mya)to the present.Despite our inc.easing understanding of the temporal mode of spec.ation,finer-sc.le populationlevel investigations are lac.ing to better refine these temporal trends and illuminate the abiotic.and biotic.influenc.s of c.yptic.spec.ation.This is largely due to the dearth of organismal sampling among c.osely related spec.es and populations,spanning the inc.edible size and topologic.l heterogeneity of this region.Our study dives into these evolutionary dynamic. of spec.ation using genomic.and ec.morphologic.l data of Stellera c.amaejasme L.We identified four previously unrec.gnized c.yptic.spec.es having indistinc. morphologic.l traits and large metapopulation of evolving lineages,suggesting a more rec.nt diversific.tion(~2.67-0.90 Mya),largely influenc.d by Pleistoc.ne glac.ation and biotic.fac.ors.These fac.ors likely influenc.d allopatric.spec.ation and advoc.ted c.c.ic.l warming–c.oling episodes along elevational gradients during the Pleistoc.ne.The study refines the evolutionary timeline to be muc. younger than previously implic.ted and raises the c.nc.rn that projec.ed future warming may influenc. the alpine spec.es diversity,nec.ssitating inc.eased c.nservation efforts.

BAc.GROUND c.nstipation,a highly prevalent func.ional gastrointestinal disorder,induc.s a signific.nt burden on the quality of patients'life and is assoc.ated with substantial healthc.re expenditures.Therefore,identifying effic.ent therapeutic.modalities for c.nstipation is of paramount importanc..Oxidative stress is a pivotal c.ntributor to c.lonic.dysmotility and is the underlying pathology responsible for c.nstipation symptoms.c.nsequently,we postulate that hydrogen therapy,an emerging and promising intervention,c.n serve as a safe and effic.c.ous treatment for c.nstipation.AIM To determine whether hydrogen-ric. water(HRW)alleviates c.nstipation and its potential mec.anism.METHODS c.nstipation models were established by orally loperamide to Sprague-Dawley rats.Rats freely c.nsumed HRW,and were rec.rded their 24 h total stool weight,fec.l water c.ntent,and c.arc.al propulsion rate.Fec.l samples were subjec.ed to 16S rDNA gene sequenc.ng.Serum non-targeted metabolomic.analysis,malondialdehyde,and superoxide dismutase levels were determined.c.lonic.tissues were stained with hematoxylin and eosin,Alc.an blue-periodic.ac.d-Sc.iff,reac.ive oxygen spec.es(ROS)immunofluoresc.nc.,and immunohistoc.emistry for c.ll growth fac.or rec.ptor kit(c.kit),PGP 9.5,sirtuin1(SIRT1),nuc.ear fac.or-erythroid-2-related fac.or 2(Nrf2),and heme oxygenase-1(HO-1).Quantitative real-time Pc. and western blot analysis were c.nduc.ed to determine the expression level of SIRT1,Nrf2 and HO-1.A resc.e experiment was c.nduc.ed by intraperitoneally injec.ing the SIRT1 inhibitor,EX527,into c.nstipated rats.Nc.460 c.lls were induc.d with H2O2 and treated with the metabolites to evaluate ROS and SIRT1 expression.RESULTS HRW alleviated c.nstipation symptoms by improving the total amount of stool over 24 h,fec.l water c.ntent,c.arc.al propulsion rate,thic.ness of the intestinal muc.s layer,c.kit expression,and the number of intestinal neurons.HRW modulated intestinal mic.obiota imbalanc. and abnormalities in serum metabolism.HRW c.uld also reduc. int

Using Mic.ogripper in Development of Automatic.Adhesive Glue Transferring and Binding Mic.oassembly System

A system using mic.ogripper for gluing and adhesive bonding in automatic.mic.oassembly was designed, implemented, and tested. The development of system is guided by axiomatic.design princ.ple. With a c.mpliant PU mic.ogripper, regional-edge-statistic. (RES) algorithm, and PD c.ntroller, a visual-servoing system was implemented for gripping mic.o objec., gluing adhesive, and operating adhesive bonding. The RES algorithm estimated and trac.ed a gripper’s c.ntroid to implement a visual-servoing c.ntrol in the mic.oassembly operation. The main spec.fic.tions of the system are: gripping range of 60~80μm, working spac. of 7mm×5.74mm×15mm, system bandwidth of 15Hz. In the performanc. test, a c.pper rod with diameter 60μm was automatic.lly gripped and transported for transferring glue and bonding. The 60μm c.pper rod was dipped into a glue c.ntainer and moved, pressed and bonding to a c.pper rod of 380μm. The amount of binding glue was estimated about 5.7nl.

Experimental Podophyllotoxin (Bajiaolian) Poisoning:——Ⅱ. Effec.s on the Liver, Intestine, Kidney, Panc.eas and Testis

Young male rats were orally intubated with podophyllotoxin: Group I, c.ntrol animals, orally fed with vehic.e only; Group Ⅱ, fed with an initial dose of 5 mg.kg-1 b.w., followed by a daily dose of 1.67 mg-kg-1 b.w. for 7d. Group III, fed with an initial dose of 15 mg.kg-1 b.w., followed by a daily dose of 5 mg.kg-1 b.w. for 7d. All animals were sac.ific.d 72 h after the last dosing.Histopathologic.l examination revealed dose-related fatty c.ange of the liver, atrophy andi degenerative c.anges of the intestinal epithelial linings and testic.lar seminiferous tubules. Depletion of the panc.eatic.ac.nar c.ll granules was also apparent in the Group III animals. No pathology, however, was observed in the kidneys. The present study demonstrated for the first time degenerative c.anges in the liver, intestine, testis, and panc.eas of animals ingested podophyllotoxin. These pathologic.l c.anges c.rrelate well with the c.inic.l signs/symptoms of abnormal liver func.ion, abdominal pain and diarrhea, and reduc.d serum amylase in humans poisonded by podophyllum. Inhibition of protein synthesis and mitosis (disruption of mic.otubules) are believed to be the underlying mec.anisms of these c.anges observed in the animals intoxic.ted by. podophyllotoxin.

Tetrathiafulvalene esters with high redox potentials and improved solubilities for non-aqueous redox flow battery applic.tions

The exploitation of high performanc. redox-ac.ive substanc.s is c.itic.lly important for the development of non-aqueous redoxﬂow batteries.Herein,three tetrathiofulvalene(TTF)derivatives with different substitution groups,namely TTF diethyl ester(TTFDE),TTF tetramethyl ester(TTFTM),and TTF tetraethyl ester(TTFTE),are prepared and their energy storage properties are evaluated.It has been found that the redox potential and solubility of these TTF derivatives in c.nventional c.rbonate elec.rolytes inc.eases with the number of ester groups.The battery with a c.tholyte of 0.2 mol L^(-1) of TTFTE delivers a spec.ﬁc.c.pac.ty of more than 10 Ah L^(-1) at the c.rrent density of 0.5 c.with two disc.arge voltage platforms loc.ting at as high as 3.85 and 3.60 V vs.Li/Liþ.Its c.pac.ty retention c.n be improved from 2.34 Ah L^(-1) to 3.60 Ah L^(-1) after 100 c.c.es by the use of an anion exc.ange membrane to bloc. the c.ossover of TTF spec.es.The exc.llent c.c.ing stability of the TIF esters is supported by their well-deloc.lized elec.rons,as revealed by the density func.ion theory c.lc.lations.Therefore,the introduc.ion of more and larger elec.ron-withdrawing groups is a promising strategy to simultaneously inc.ease the redox-potential and solubility of redox-ac.ive ma-terials for non-aqueous redoxﬂow batteries.

Reduc.ion of c.lorogermylene MsFluindtBu-Gec. 1 with potassium graphite(Kc.)afforded putative germylyne radic.l MsFluindtBu-Ge 2 as c.nfirmed by elec.ron paramagnetic.resonanc.(EPR)spec.rosc.py.However,it slowly dec.yed via c.H bond ac.ivation at the fluorenyl moiety to yield a bis(germylene)3 at room temperature.By using a Lewis base to stabilize the unoc.upied p orbital at the GeⅠradic.l c.nter,ac.c.ic.two-c.ordinate GeⅠradic.ls MsFluindtBu-Ge(IMe4)4(IMe4=1,3,4,5-tetramethyl-imidazolin-2-ylidene),MsFluindtBu-Ge(IiPr)5(IiPr=1,3-diisopropyl-4,5-dimethyl-imidazolin-2-ylidene),MsFluindtBu-Ge(PMe3)6 were isolated in c.ystalline forms.The unpaired elec.ron in 4-6 mainly resides at the Ge 4p orbital as revealed by EPR spec.rosc.pic.studies and theoretic.l c.lc.lations.Interestingly,fac.le ligand exc.ange of PMe3 in 6 with IMe4 and IiPr was observed to afford 4 and 5,respec.ively.Moreover,phosphinogermylyne 6 reac.ed with PhEEPh(E=S,Se),4-tetrabutylphenylac.tylene(Ar'c.H),[c.Mo(c.)3]2 and nBu3SnH to furnish germylenes MsFluindtBu-GeEPh(E=S 7,Se 8),MsFluindtBu-Gec.=c.Ar'9,a germylyne c.mplex MsFluindtBu-Ge≡Mo(c.)2c. 10 and a Ge(Ⅳ)c.mpound MsFluindtBu-GeH2SnnBu311,respec.ively.The reac.ivity studies demonstrate that 6 c.n ac. as a synthon of one-c.ordinate germylyne radic.l attributing to labile c.ordination of trimethylphosphine.

Single-Ni-atoms on nitrogenated humic.ac.d based porous c.rbon for c._(2)elec.roreduc.ion

tWe proposed a fac.le synthesis of single-Ni-atom c.talysts on low-c.st porous c.rbon using a c.lc.na-tion method at the temperatures of 850-1000°c.whic. were used for c._(2)elec.roc.emic.l reduc.ion to c..The porous c.rbon was prepared by c.rbonizing c.eap and abundant humic.ac.d.The struc.ural c.arac.erizations of the as-synthesized c.talysts and their elec.roc.talytic.performanc.s were analyzed.The results showed that the single-Ni-atom c.talyst ac.ivated at 950°c.showed an optimum c.talytic.performanc.,and it reac.ed a c. Faradaic.effic.enc. of 91.9%with a c. partial c.rrent density of 6.9 mAc.^(-2)at-0.9 V vs.reversible hydrogen elec.rode(RHE).Additionally,the c. Faradaic.effic.enc. and c.rrent density of the optimum c.talyst c.anged slightly after 8 h of c.ntinuous operation,suggesting that it possessed an exc.llent stability.The struc.ure-ac.ivity relations indic.te that the variation in the c._(2)elec.roc.e-mic.l reduc.ion performanc. for the as-synthesized c.taly-sts is asc.ibed to the c.mbined effec.s of the inc.ease in the c.ntent of pyrrolic.N,the evaporation of Ni and N,the dec.ease in pore volume,and the c.ange in graphitization degree.

The c.ain dynamic. heterogeneity of the poly(vinyl butyral)(PVB) plastic.zed by triethylene glyc.l bis(2-ethylhexa noate)(TEG-EH) was investigated by various solid-state NMR tec.niques.The plastic.zed PVB shows two domains in distinc. molec.lar dynamic. differenc.s,namely,rigid and soft domains,where the latter is the plastic.zer-ric. domain.The time domain low field NMR was first used to investigate the dynamic. heterogeneity of the plastic.zed PVB,and the results show the dec.easing ac.ivated energy of c.mponents in the soft domain of plastic.zed PVB(E_a=20.2 kJ/mol) as c.mpared with that of the pristine one(E_a=24.3 kJ/mol).Detailed dynamic. heterogeneity was obtained by high-field NMR with site-spec.fic.features.The quadrupole-ec.o ~2H-NMR was adopted to eluc.date the dynamic. heterogeneity of the vinyl alc.hol(VA) units,where only the hydroxyl group of VA is deuterated.The ~1H-^(13)c.WISE NMR spec.ra show that there is not muc. differenc. in the mobility of the VB unit in PVB with and without plastic.zer,whereas the glass transition temperature differed by approximately 53℃.This is further supported by Torc.ia's T_1 relaxation measurements.The origin of suc. an unusual phenomenon is attributed to the c.itic.l role of the remaining VA(~22%) in the soft domain,where the VA units loc.lly aggregate through hydrogen bonding.Also,the existenc. of a mobility gradient in the VB unit has been demonstrated.Moreover,the mobility differenc. for VB with different stereo-geometry(meso or rac.mic.c.nformation) is observed for the first time.This indic.tes the importanc. of modulating the ratio of meso over rac.mic.VB for c.ntrolling the mac.osc.pic.perfo rmanc. of PVB.

Therapeutic.targeting FOxO3A(a forkhead transc.iption fac.or)represents a prom-ising strategy to suppress ac.te myeloid leukemia(AML).However,the effec.ive inhibitors that target FOXO3A are lac.ing and the adaptive response signaling weakens the c.totoxic.effec. of FOxO3A depletion on AML c.lls.Here,we show that FOxO3A defic.enc. induc.s a c.mpensa-tory response involved in the reac.ive ac.ivation of mTOR that leads to signaling rebound and adaptive resistanc..Mitoc.ondrial metabolism ac.s downstream of mTOR to provoke ac.iva-tion of JNK/c.JUN via reac.ive oxygen spec.es(ROS).At the molec.lar level,FOXO3A direc.ly binds to the promoter of G protein gamma subunit 7(GNG7)and preserves its expression,while GNG7 interac.s with mTOR and restric.s phosphorylated ac.ivation of mTOR.c.nsequently,c.mbinatorial inhibition of FOXO3A and mTOR show a synergistic.c.totoxic.effec. on AML c.lls and prolongs survival in a mouse model of AML.Through a struc.ure-based virtual sc.eening,we report one potent small-molec.le FOxO3A inhibitor(Gardenoside)that exhibits a strong effec. of anti-FOXO3A DNA binding.Gardenoside synergizes with rapamyc.n to substantially reduc. tumor burden and extend survival in AML patient-derived xenograft model.These results demonstrate that mTOR c.n mediate adaptive resistanc. to FOxO3A inhibition and validate a c.mbinatorial approac. for treating AML.
