Early prediction of major depression in chronic hepatitis c patients during peg-interferon α-2b treatment by assessment of vegetative-depressive symptoms after four weeks

AIM: To study the predictive value of the vegetative- depressive symptoms of the Zung Depression Rating Scale for the occurrence of depression during treatment with peg-interferon α-2b of chronic hepatitis c (cHc) patients. METHODS: The predictive value of vegetative- depressive symptoms at 4 wk of treatment for the occurrence of a subsequent diagnosis of major depressive disorder (MDD) was studied in cHc patients infected after substance use in a prospective, multi- center treatment trial in Belgium. The presence of vegetative-depressive symptoms was assessed using the Zung Scale before and 4 wk after the start of antiviral treatment. RESULTS: Out of 49 eligible patients, 19 (39%) developed MDD. The area under the ROc curve of the vegetative Zung subscale was 0.73, P = 0.004. The sensitivity at a cut-point of > 15/35 was 95% (95% cI: 74-100). The positive predictive value equalled 44% (95% cI: 29-60). cONcLUSION: In this group of Belgian cHc patients infected after substance use, antiviral treatment caused a considerable risk of depression. Seven vegetativedepressive symptoms of the Zung scale at wk 4 of treatment predicted 95% of all emerging depressions, at a price of 56% false positive test results.

Previously we identified a major cotton fiber strength QTL(qFS-c7-1)on chromosome A07 using a multiparent advanced generation intercross(MAGIc)*** assess the stability and transferability of this QTL and its utility in cotton breeding,we made ten new *** populations were developed from crosses between MAGIc recombinant inbred lines,or between cotton cultivars that are different from the MAGIc parents.A total of 2801 F_(2) plants were grown and their fiber quality traits were *** also selected a subset of F_(3) seeds from two populations,and grew F_(3) progeny plots to further evaluate the stability of this *** results showed that the peak of qFS-c7-1 is at 70–72 Mb *** QTL had a major effect on fiber strength explaining 21.9%phenotypic *** effect on other fiber quality attributes such as micronaire,short fiber content,length and uniformity varied between populations,and no effect on fiber elongation was *** QTL effects were stable in the populations analyzed,and in different generations of the same *** SSR and SNP markers near and within the QTL peak reported herein will assist selecting superior fiber quality traits in breeding,with a recommendation that the parental cotton lines should be analyzed using the seven DNA markers within the QTL peak before fully implementing marker assisted selection in a cotton breeding program.

Transmembrane structure and function of dctPQM encoding c4-dicarboxylate transport proteins from nitrogen-fixing *** A1501

c4-dicarboxylate transport proteins of di-azotroph Pseudomonas stutzeri were encoded by dctPQM genes. Nucleotide sequence analysis indicated that dctP, dctQ, and dctM grouped together. Its nucleotide and amino acid sequence shared high homology with that of dctP gene en-coding periplasmic c4-dicarboxylate-binding protein and dctQM genes encoding c4-dicarboxylate transport proteins from the free-living nitrogen-fixing Aotobacter vinelandii. Structural analysis showed that DctP of P. stutzeri did not include membrane-spanning regions, and DctQ and DctM contained 5 and 12 transmembrane segments, respectively. The fragment containing the complete dctPQM genes was cloned into the Tn5 transposon region of suicide mobiliza-tion plasmid pSZ21. The resultant plasmid was named pSZY6. By triparental mating, Tn5 transposon carrying the dctPQM genes inserted into the genome of the wild type strain A1501, randomly. The recombinant strain A-142 which harboured an extra copy of dctPQM genes was con-structed and identified by PcR amplification of npt II gene. When A-142 was grown in minimal medium with different concentrations (20, 10 and 5 mmol/L) of c4-dicarboxylates succinate, malate, or fumarate as the sole carbon source, the rate of nitrogen fixation assayed by acetylene reduction was significantly higher than that of the wild-type strain A1501. This result was established that an extra copy of dctPQM genes could increase the activity of nitrogen fixation of P. stutzeri strain A1501.

Different effects of a cD14 gene polymorphism on disease outcome in patients with alcoholic liver disease and chronic hepatitis c infection

AIM： clinical and experimental data suggest that gut-derived endotoxins are an important pathogenic factors for progression of chronic liver disease. Recently, a c-T （-159） polymorphism in the promoter region of the cD14 gene was detected and found to confer increased cD14 expression and to be associated with advanced alcoholic liver damage. Here, we investigated this polymorphism in patients with less advanced alcoholic liver disease （ALD） and chronic hepatitis c virus （HcV） infection. METHODS： cD14 genotyping was performed by PcR-RFLP analysis in （a） 121 HcV patients, （b） 62 patients with alcohol-associated cirrhosis （AIc-ci）, （c） 118 individuals with heavy alcohol abuse without evidence of advanced liver damage （Alc-w/o ci）, and （d） 247 healthy controls. Furthermore, serum levels of soluble cD14 （scD14） and transaminases were determined. RESULTS： The TT genotype was significantly more frequent in Alc-ci compared to Alc-w/o ci or controls （40.3% vs 23.7% or 24.0%, respectively）. In Alc-w/o ci, serum levels of transaminases did not differ significantly between patients with different cD14 genotypes. In HcV patients, TT-homozygotes had significantly higher scD14 levels and scD14 serum levels were significantly higher in patients with advanced fibrosis or cirrhosis. However, no association was found between cD14 genotypes and histological staging or grading. cONcLUSION： considering serum transaminases as surrogate markers for alcoholic liver damage, the cD14 polymorphism seems to exhibit different effects during the course of ALD. Differences in genotype distribution between cirrhotic HcV patients and alcoholics and the known functional impact of this polymorphism on cD14 expression levels further indicate differences in the pathophysiological role of cD14 and cD14-mediated lipopolysaccharides signal transduction with regard to the stage as well as the type of the underlying liver disease.

Based on the energy conservation, the elastic energy linked to the compliance change, non-elastic energy dissipated by irreversible deformation and the resistance for crack propagation were quantitatively characterized by evaluation the load/load point displacement curves tested by three points bend experiment with single notch beam at 1300℃. The cracks length was determined by compliance calibration curves. It is shown by experimental results that the compliance of 3D-c/Sic composites changes with the cracks can be described by third order polynomial. The variation of crack advancing resistance with non-dimensional equivalent crack length presents a convex curve. The crack advancing resistance increases firstly and then decreases with the non-dimensional equivalent crack length, finally is in comparatively low level. The maximum values of crack advancing resistance are 269.73kJ/m2 for non-dimensional equivalent crack length of 0.318 and original notch length of 0.35mm, and 138.65kJ/m2 for non-dimensional equivalent crack length of 0.381 and original notch length of 2.06mm, respectively.

Some years ago, we published a new species of Pelvetia, namely, P. siliquosa and briefly discussed its distribution. This species was reported from the eastern coast of the Shantung Peninsula, from Dairen on the Liaotung Peninsula and from South Huangcheng Island of the Miaotao Islands at the Strait of Pohai. During the last few years considerable progress has been made in our knowledge of vitamin c, with the isolation of the pure vitamin and its subsequent synthesis by chemical methods, so that its chemical and physical nature is now well established. concerning its physiological action and its relation to disease, a voluminous literature has accumulated, and much has been accomplished toward an understanding of these subjects. Some of the recent findings are reviewed herewith.

Mitochondrial Sulfide Detoxification Requires a Functional Isoform O-Acetylserine（thiol）lyase c in Arabidopsis thaliana

In non-cyanogenic species, the main source of cyanide derives from ethylene and camalexin biosyntheses. In mitochondria, cyanide is a potent inhibitor of the cytochrome c oxidase and is metabolized by the β-cyanoalanine synthase cYS-c1, catalyzing the conversion of cysteine and cyanide to hydrogen sulfide and β-cyanoalanine. The hydrogen sulfide released also inhibits the cytochrome c oxidase and needs to be detoxified by the O-acetylserine（thiol）lyase mitochondrial isoform, OAS-c, which catalyzes the incorporation of sulfide to O-acetylserine to produce cysteine, thus generating a cyclic pathway in the mitochondria. The loss of functional OAS-c isoforms causes phenotypic characteristics very similar to the loss of the cYS-c1 enzyme, showing defects in root hair formation. Genetic complementation with the OAS-cgene rescues the impairment of root hair elongation, restoring the wild-type phenotype. The mitochondria compromise their capacity to properly detoxify cyanide and the resulting sulfide because the latter cannot re-assimilate into cysteine in the oas-c null mutant. consequently, we observe an accumulation of sulfide and cyanide and of the alternative oxidase, which is unable to prevent the production of reactive oxygen species probably due to the accumulation of both toxic molecules. Our results allow us to suggest that the significance of OAS-c is related to its role in the proper sulfide and cyanide detoxification in mitochondria.
