In a previous transcriptomic study of human bone marrow stromal cells(BMSCs,also known as bone marrow-derived^mesenchymal stem cells#,)/SFRP2 was highly over-represented in a subset of multipotent BMSCs(skeletal stem cells,SSCs),which recreate a bone/marrow organ in an in vivo ectopic bone formation assay.SFRPs modulate WNT signaling,which is essential to maintain skeletal homeostasis,but the specific role of SFRP2 in BMSCs/SSCs is unclear.Here,we evaluated Sfrp2 deficiency on BMSC/SSC function in models of skeletal organogenesis and regeneration.The skeleton of Sfrp2-def\c\ent(KO)mice is overtly normal;but their BMSCs/SSCs exhibit reduced colony-forming efficiency,reflecting low SSC self-renewal/abundancy.Sfrp2 KO BMSCs/SSCs formed less trabecular bone than those from WT littermates in the ectopic bone formation assay.Moreover,regeneration of a cortical drilled hole defect was dramatically impaired in Sfrp2 KO mice.Sfrp2-deficient BMSCs/SSCs exhibited poor in vitro osteogenic differentiation as measured by Runx2 and Osterix expression and calcium accumulation.Interestingly,activation of the Wnt co-receptor;Lrp6,and expression of Wnt target genes,Axin2,C-myc and Cyclin Dl,were reduced in Sfrp2-deficient BMSCs/SSCs.Addition of recombinant Sfrp2 restored most of these activities,suggesting that Sfrp2 acts as a Wnt agonist.We demonstrate that Sfrp2 plays a role in self-renewal of SSCs and in the recruitment and differentiation of adult SSCs during bone healing.SFRP2 is also a useful marker of BMSC/SSC multipotency,and a factor to potentially improve the quality of ex vivo expanded BMSC/SSC products.

Quietly, with little apparent notice from even the strongest advocates for global mental health, China is undertaking the world's largest – and arguably most important – mental health services demonstration project, a project focused on providing comprehensive care for persons with severe mental illnesses.As Professor Ma indicates in her short report,[1] the ‘686 Project' was launched as part of China's commitment to rebuild its public health infrastructure following the SARS epidemic, and has now moved beyond the initial pilot phase into a process of scaling up community mental health services throughout the country.China is currently moving toward passage of its first national mental health law, so the project has profound implications for mental health policy in the country.It will also provide useful models for the development of mental health policies in other countries with limited mental health personnel.

Cost Analysis of Diffusion Tensor Imaging and MR Tractography of the Brain

Purpose: To determine the total direct costs (fixed and variable costs) of diffusion tensor imaging (DTI) and MR tractography reconstruction of the brain. Materials and Methods: The direct fixed and variable costs of DTI with MR tractography were determined prospectively with time and motion analysis in a 1.5-Tesla MR scanner using 15 encoding directions. Seventeen patients with seizure disorders, 9 males & 8 females, with mean age of 13 years (age range 2 - 33 years) were studied. Total direct costs were calculated from all direct fixed and variable costs. Sensitivity analyses between 1.5 versus a 3-Tesla MR system, and 15 versus 32 encoding directions were done. Results: The total direct costs of DTI and MR tractography for a 1.5-T system with 15 encoding directions were US $97. Variable cost was $76.80 and fixed cost was $20.20. Total direct costs for a 3-T system with 15 directions decreased to US $94.5 because of the shorter scan time despite the higher cost of the 3-T system. The most costly component of the direct cost was post-processing analysis at US $46.00. Conclusion: DTI with MR tractography has important total direct costs with variable costs higher than the fixed costs. The post processing variable cost is the most expensive component. Developing more accurate automated post-processing software for DTI and MR tractography is important to decrease this variable labor cost. Given the added value of DTI-MR tractography and the costs involved reimbursement codes should be considered.

Seeds are a source of organic (carbon, C) and mineral (nitrogen, N and phosphorus, P) nutrients for the growing seedling. There is much information on seed mass and N and P contents, and the relationship between these and seedling mass. Within the world’s temperate regions, these collectively show that N and P concentrations remain constant or rise with increase in seed mass and that seeds are larger and more nutrient-enriched in poorer soils. Seed N and P were more important than seed C in accounting for seedling mass in 85% of studies we assessed. In nutrient- and water-limited environments that are not light-limited, large seeds routinely provision the seedling with N and P that enhance C-fixation and thus general growth in the first wet season. This system is so efficient that growth response to soil nutrients may be negligible in first-year seedlings arising from seeds > 15 mg mass, N content > 5 mg and P content > 1.6 mg. The elongating taproot system absorbs nutrients and maintains water uptake as soil water retreats, enhancing the chances of survival in the first dry season. We outline an interpretative scenario for the special role of large seeds (>15 mg) in nutrient- and water-limited environments that recognizes the critical role of N and P for photosynthesis in ensuring sufficient C-supply to the rapidly descending roots for effective drought-avoidance by the young plant.

Objectives: Non-adherence to tuberculosis (TB) treatment causes development of multi-drug resistance (MDR). In Armenia, about 47% of previously-treated TB patients develop MDR-TB. This pilot intervention intended to explore the feasibility and effectiveness of a family-based-counseling (FBC) that included a psychological component in terms of improved adherence of TB patients to treatment and reduced stigma of TB. Methods: Overall, 136 regular TB patients (55) and their family members (81) participated in a single 90-minute interactive counseling session conducted in each household by the team of professional psychologist and TB nurse. To evaluate FBC effectiveness, we administered baseline and follow-up surveys to 52 TB patients and their 57 family members in 2012 and compared treatment outcomes of the study participants with the national data for 2011 and 2013. Results: We found that the intervention substantially improved the mean knowledge score of TB patients (from 19.2 to 21.6, p Conclusion: The evaluation showed that a low-cost one-time family–based educational intervention with a psychological component can be effective in improving treatment outcomes of TB patients.

Background & Aim: Employees’ engagement is a significant organizational competence. Turnover intention is an institutional concern that needs critical analysis of identifying its predictors. Studies have supported the relationship between nurses’ engagement and their desire to leave the organization. This study aims to explore the relationship between work engagement and turnover intention among nurses working in the Kingdom of Bahrain. Methods: A cross-sectional design was used for this study. A group of 922 nurses from a major tertiary hospital in the Kingdom of Bahrain was invited to complete the online self-administered survey. The work engagement was assessed using the Utrecht Work Engagement Scale (UWES). The turnover intention was assessed using the 3-item TI Scale based on Mobley et al. survey. The study was conducted in December 2019. Results: 610 nurses completed the survey on a response rate of (66.2%). The result suggests that nurses expressed high level of work engagement Mean ± Standard Deviation [M = 4.85, SD ± 0.70], and low level of turnover intention [M = 2.29, SD ± 0.94]. Pearson correlation revealed a significant negative relationship between work engagement and turnover intention at p Conclusion/practical implication: It’s paramount for the nurses’ leaders and human resource development practitioners to focus on work engagement dimensions, and developing effective interventions to improve it, which eventually decreases nursing staff’s turnover intention. Retention is more preferable than recruiting, employing, and training new staff. This study confirms the significant relationship between work engagement and turnover intention among nurses.

AIM To investigate the regulation of Myopalladin(Mypn) and identify its gene network involved in restrictive cardiomyopathy(RCM).METHODS Gene expression values were measured in the heart of a large family of BXD recombinant inbred(RI) mice derived from C57BL/6J and DBA/2J. The proteomics data were collected from Mypn knock-in and knock-out mice. Expression quantitative trait locus(eQ TL) mapping methods and gene enrichment analysis were used to identify Mypn regulation,gene pathway and co-expression networks.RESULTS A wide range of variation was found in expression of Mypn among BXD strains. We identified upstream genetic loci at chromosome 1 and 5 that modulate the expression of Mypn. Candidate genes within these loci include Ncoa2,Vcpip1,Sgk3,and Lgi2. We also identified 15 sarcomeric genes interacting with Mypn and constructed the gene network. Two novel members of this network(Syne1 and Myom1) have been confirmed at the protein level. Several members in this network are already known to relate to cardiomyopathy with some novel genes candidates that could be involved in RCM. CONCLUSION Using systematic genetics approach,we constructed Mypn co-expression networks that define the biological process categories within which similarly regulated genes function. Through this strategy we have found several novel genes that interact with Mypn that may play an important role in the development of RCM.
