The sa.ety of na.opa.ticle-ba.ed drug delivery systems(DDSs)for ca.cer trea.ment is still a.challenge,restricted by the intrinsic cytotoxicity of drug ca.riers a.d lea.a.e of loa.ed ***,we propose a.novel na.oca.rier’s cytotoxicity a.oida.ce stra.egy by synthesizing a. enca.sula.ion core–shell structure of zeolitic imida.ola.e fra.ework-8(ZIF-8)-ba.ed colloid pa.ticles(CPs)with a. a.orphous ZIF-8 *** enca.sula.ion structure a.hieves a. ultra.high loa.1}ng ra.e(LR)of 90%(i.e.,9 mg doxorubicin(DOX)per 1 mg ZIF-8)for DOX a.d the protection of DOX from ***,to deliver unit-dose drug,this ultra.high LR of 90%significa.tly reduces the usa.e of ZIF-8 to 1.2%(2 orders of ma.nitude)compa.ed to that of DOX@ZIF-8 with a.10%LR,in which cytotoxicity of ZIF-8 could well below the sa.ety limit a.d then be rela.ively ***,drug delivery effica.y,sca.e-up a.ility,a.d universa.ity of this enca.sula.ion structure have been further *** findings suggest the grea. potentia. of this ZIF-8-ba.ed enca.sula.ion core–shell structure in the field of drug delivery.
