Prokineticin 2/Bv8 is expressed in Kupffer cells in liver and is down regulated in human hepatocellular carcinoma

AIM： TO study the implication of prokineticin 1 （PKI/EGVEGF） and prokineticin 2 （PK2/13v8） in hepatocellular carcinoma angiogenesis.METHODS： The gene induction of PK1/EG-VEGF and PK2/Bv8 was investigated in 10 normal, 28 fibrotic and 28 tumoral livers by using real time PCR. Their expression was compared to the expression of VEGF （an angiogenesis marker）, vWF （an endothelial cell marker） and to CD68 （a monocyte/macrophage marker）. Furthermore, the rnRNA levels of PK1/EG-VEGF, PK2/Bv8, prokineticin receptor 1 and 2 were evaluated by real time PCR in isolated liver cell populations. Finally, PK2/Bv8 protein was detected in normal liver paraffin sections and in isolated liver cells by immunohistochernistry and immunocytochemistry.RESULTS： PK2/Bv8 mRNA but not PK1/EG-VEGF was expressed in all types of normal liver samples examined. In the context of liver tumor development, we reported that PK2/13v8 correlates only with CD68 and showed a significant decrease in expression as the pathology evolves towards cancer. Whereas, VEGF and vWF mRNA were significantly upregulated in both fibrosis and HCC,as expected. In addition, out of all isolated liver cells examined, only Kupffer cells （liver resident macrophages） express significant levels of PK2/Bv8 and its receptors, prokineticin receptor 1 and 2.CONclUSION： In normal liver PK2/Bv8 and its receptors were specifically expressed by Kupffer cells. PK2/Bv8 expression decreased as the liver evolves towards cancer and did not correlate with HCC angiogenesis.

Neurodegeneration and choroidal vascular features on OCT in the progression to advanced age-related macular degeneration

AIM:To quantify and compare longitudinal thickness changes of the ganglion cell complex(GCC)and the choroid in patients with different patterns of age-related macular degeneration(AMD)***:Retrospective cohort analysis of anonymized data from participants aged 50y or more and diagnosed with early/intermediate AMD in at least one eye(with no evidence of advanced AMD).A total of 64 participants were included from the Instituto de Retina de Lisboa(IRL)study(IPL/2022/MetAllAMD_ESTeSL)and divided into 4 groups according to the Rotterdam classification for *** domain optical coherence tomography(SD-OCT)was used to assess and quantify GCC and choroid thickness at two time points(first visit vs last visit)with a minimum interval of ***:In the GCC inner ring,a thinner thickness(P=0.001)was observed in the atrophic AMD group(51.3±21.4μm)compared to the early AMD(84.3±11.5μm),intermediate AMD(77.6±16.1μm)and neovascular AMD(88.9±16.3μm)*** thickness quantification showed a generalized reduction in the central circle(P=0.002)and inner ring(P=0.001).Slight reductions in retinal thickness were more accentuated in the inner ring in the atrophic AMD(-13%;PclUSION:The variation of the analyzed structures could be an indicator of risk of progression with neurodegenerative(GCC)or vascular(choroid)pattern in the intermediate and atrophic *** quantification of both structures can provide important information about the risk of disease progression in the early and intermediate stages but also for the evolution pattern into late stages(atrophic or neovascular).

Tailoring sub-5 nm Fe-doped CeO2 nanocrystals within confined spaces to boost photocatalytic hydrogen evolution under visible light

This work aimed to study the efficiency of the reverse micelle（RM） preparation route in the syntheses of sub-5 nm Fe-doped CeO2nanocrystals for boosting the visible-light-driven photocatalytic hydrogen production from methanol aqueous *** effectiveness of confining precipitation reactions within micellar cages was evaluated through extensive physicochemical cha *** particula r,the nominal composition（0-5 mol% Fe） was preserved as ascertained by ICP-MS analysis,and the absence of separate iron-containing crystalline phases was supported by X-ray *** effective aliovalent doping and modulation of the optical properties were investigated using UV-Vis,Raman,and photoluminescence spectroscopies.2.5 mol% iron was found to be an optimal content to achieve a significant decrease in the band gap,enhance the concentration of oxygen vacancy defects,and increase the charge carrier *** photocatalytic activity of Fe-doped CeO2prepared at different Fe contents with RM preparation was studied and compared with undoped *** optimal iron load was identified to be2.5 mol%,achieving the highest hydrogen production(7566 μmol L-1after 240 min under visible light).Moreover,for comparison,the conventional precipitation（P） method was adopted to prepare iron containing CeO2at the optimal content（2.5 mol% Fe）.The Fe-doped CeO2catalyst prepared by RM showed a significantly higher hydrogen production than that obtained with the sample prepared by the P *** optimal Fe-doped CeO2,prepared by the RM method,was stable for six reuse ***,the role of water in the mechanism of photocatalytic hydrogen evolution under visible light was studied through the test in the presence of *** obtained results evidenced that hydrogen was produced from the reduction of H+by the electrons promoted in the conduction band,while methanol was preferentially oxidized by the photogenerated positive holes.

矮塔斜拉桥通常采用对称悬臂法施工,客观条件可能导致该方法无法采用,需要研究非对称悬臂施工的可行性。文章针对双塔双索面预应力混凝土矮塔斜拉桥开展实桥监测,建立有限元模型,分析合理成桥状态和合理施工状态。考虑背景工程无法采用对称悬臂法的实际情况,基于合理施工状态提出边跨现浇段非对称悬臂施工的2种方案,对比采用2种方案完成边跨现浇段施工后对合理施工状态下主梁的挠度、应力及斜拉索索力的影响。结果表明:采用应力平衡法计算成桥恒载弯矩可行域,并以此为约束条件利用基于影响矩阵法原理的未知荷载系数法可计算合理成桥索力;利用考虑施工阶段的未知荷载系数法计算合理初张拉索力及二次张拉索力,可得到合理施工状态;与方案1相比,采用方案2的主梁最大挠度更小且分布更合理,边跨无索区主梁上缘拉应力更小,斜拉索索力分布更均匀;方案2对矮塔斜拉桥的合理施工状态影响较小,最终可达到合理成桥状态。

Combined Effect of Honey from Central West Brazil on Bacterial Membrane Permeability

Honey, an apicultural product with a complex chemical composition, contains numerous bioactive compounds with potential antimicrobial effects. This study investigated the effect of Apis mellifera honey from Brazil’s Central-West Region, combined with antibiotics, on bacterial membrane permeability, exploring the contributions of bioactive compounds and the botanical origin of honey. Six fresh Apis mellifera honey samples and their fractions (hexane and ethyl acetate) were analyzed, for a total of 18 samples. The bacteria Staphylococcus epidermidis, Helicobacter pylori and Enterococcus faecalis were used for antibacterial activity tests, which included minimum inhibitory concentration (MIC) determination and synergistic effect (checkerboard) assays. The total polyphenol and flavonoid contents were quantified, and the botanical origin was determined based on pollen analysis. The tested honey samples significantly affected bacterial membrane permeability when combined with rifampicin and clarithromycin. Although many honey-derived bioactive compounds, when isolated, did not exhibit significant activity against these bacteria, the additive or synergistic effect of multiple compounds acting on different targets appears to potentiate the antibacterial action. Descriptive statistical analysis, including means and 95% confidence intervals, confirmed the relevance of the findings. This study has provided an important discovery: Honey has an effect on bacterial membrane permeability, although the specific mechanisms involved in this process require further investigation.

Dynamics of Water Flow in the Atmosphere-Aerial Roots Continuum

The study of the parameters influencing the absorption of atmospheric humidity not condensed by plants with aerial roots is a challenge in the current context of climatic disturbance where methods of adapting plants to water stress have become the concern of many scientific researchers. Based on Poiseuille and Fick laws, leaf water potential depending on solar radiation and atmospheric water potential around aerial roots;the influence of temperature, relative humidity, inclination angle of roots and solar radiation wavelength on the radial and axial water flow through a root segment is simulated. The various simulations show that a low temperature of the air surrounding the root favors a significant entry of water into the root as well as a relative humidity of the high air approaching 100%. The angle of inclination has an influence on the quantity of water entering the root and that rising in the xylem. These different streams of water are optimized for root tilting of 60°. The wavelength of the red radiation received by the leaves favors more entry and rise of water in the root. Future studies for transgenes should be taken into account the influence of all these parameters.

This article is part of the ongoing study of the moderate geomagnetic activity of corotation. The aim here is to examine ionospheric anomalies through variations in Total Electronic Content (TEC) recorded at the Koudougou GPS station over solar cycle 24 during periods of corotation activity. Through the diurnal variations by solar phase, we observed a TEC peak at 14:00 TL. The TEC data analysed revealed winter, annual and semi-annual anomalies. However, the equinoctial anomaly was insignificant during corotation activity at the Koudougou station during solar cycle 24.

Very-high-dose olanzapine for treatment-resistant schizophrenia

Treatment-resistant schizophrenia has an extremely negative impact on mental health and social life. If clozapine, the gold standard treatment, fails, there are very few options left. The literature suggests that high-dose olanzapine (20 - 60 mg/day) is a possible alternative. We report two cases in which very high doses of olanzapine were administered, with significant clinical improvements above 60 mg/day. clinical, metabolic and cardiac tolerance was good. This report highlights the usefulness of very-high-dose olanzapine in treatment-resistant schizophrenia. The main hypotheses concerning the psychopharmacological mechanisms of very-high-dose olanzapine are discussed.
