A highly sensitive bio-barcode immunoassay for multi-residue detection of organophosphate pesticides based on fluorescence antiquenching

Balancing the risks and benefits of organophosphate pesticides(OPs)on human and environmental health relies partly on their accurate measurement.A highly sensitive fluorescence anti-quenching multi-residue bio-barcode immunoassay was developed to detect OPs(triazophos,parathion,and chlorpyrifos)in apples,turnips,cabbages,and *** nanoparticles were functionalized with monoclonal antibodies against the tested *** oligonucleotides were complementarily hybridized with an RNA fluorescent label for signal *** detection signals were generated by DNA-RNA hybridization and ribonuclease H dissociation of the *** resulting fluorescence signal enables multiplexed quantification of triazophos,parathion,and chlorpyrifos residues over the concentration range of 0.01-25,0.01-50,and 0.1-50 ng/mL with limits of detection of 0.014,0.011,and 0.126 ng/mL,*** mean recovery ranged between 80.3% and 110.8% with relative standard deviations of 7.3%-17.6%,which correlate well with results obtained by liquid chromatography-tandem mass spectrometry(LC-mS/mS).The proposed bio-barcode immunoassay is stable,reproducible and reliable,and is able to detect low residual levels of multi-residue OPs in agricultural products.

Leptospira binds to host extracellular matrix(ECm)through surface exposed outer membrane proteins called adhesin in order to initiate *** various adhesins present on the surface of the spirochete, Leptospira-immunoglobulin like proteins(Lig proteins)and LipL32 are most abundant,widely distributed among pathogenic serovars and well *** fragments of Lig proteins(Ligcon4,Ligcon4-7.5,LigBcen2) and C-terminus fragment of LipL32 all of that bind to host ECm were fused,expressed and purified in soluble form as *** week hamsters were immunized subcutaneously with various fusion proteins emulsified in EmULSIGEN-D adjuvant and subsequently boosted at 3 *** protective efficacy of these novel fusion proteins was evaluated against subsequent challenge with highly virulent *** serovar Pomona(mLD50-100).Our results indicate that fusion protein based vaccine induced significant protection against acute infection with respect to PBS-adjuvant vaccinated controls as revealed by enhanced survival and reduced pulmonary ***,the protection mediated by these novel proteins was higher than that of conserved region of Lig protein(Ligcon,established protective antigen)and correlated to the level of ***32 failed to impart significant protection,however fusing its immunogenic C-terminus domain to Lig fragments slightly delayed the morbidity of the infected *** results demonstrate that this novel strategy could be promising in developing effective subunit vaccine to combat this zoonotic infection.

This paper describes the effect of Frat-1, Idax and Axam toward the Wnt signaling pathway. The Wnt pathway is an important intracellular signaling pathway that plays a crucial role in the cell cycle process at several levels of development. Understanding this pathway thoroughly will enable identification of new drug targets and thereby open avenues to more effective therapies for a range of Wnt pathway related diseases such as cancer, osteoporosis, and mesothelioma. Dishevelled （Dsh） is one of the multi-module proteins working m the Wnt pathway. The function of Dsh depends on the regulators which bind to it, components it interacts with and most importantly the Wnt signal. It is important to note that positive regulators are those factors that induce gene transcription. Conversely, the negative regulators induce β-Catenin degradation. The positive regulator such as Frat- 1 would promote the Wnt signaling pathway and downstream gene transcription. meanwhile the negative regulator such as Idax and Axam would inhibit the signaling pathway and promotes β-Catenin degradation which will thereby prevent aberrant gene transcription.

Night-time Rise in Rat Pineal N-Acetyltransferase due to Carbaryl Administration Is Reduced by Propranolol Treatment

The purpose of the present study was to examine the effects of administration of sublethal doses of carbaryl on nighttime rat pineal melatonin synthesis in the presence and absence of propranolol, a β-adrenergic receptor antagonist. Two groups of adult male albino rats were administered orally N-methyl-l-naphthylcarbamate (carbaryl) (8. 33mg/kg BW daily in corn oil) for six successive days; another two groups received corn oil *** the last day of carbaryl treatment, half of the animals received an intraperitoneal injection of propranolol (20 mg/kg body weight, one hour before lights off). The other two groups were given a saline injection. Four hours after darkness onset, pineal N-acetyltransferase (NAT) and hydroxyindole-O-methyltransferase (HIOmT) activities as well as pineal concentrations of 5-hydroxytryptophan (5HTP), serotonin (5HT),5-hydroxyindole acetic acid (5HIAA) and pineal and serum melatonin levels were measured. Nocturnal NAT activity was increased due to carbaryl administration but the pesticide was ineffective in stimulating NAT activity in rats treated with *** 5HT was decreased due to carbaryl administration but 5HTP and 5HIAA levels were unaffected. Pineal and serum melatonin levels were decreased due to propranolol treatment. The results indicate that carbaryl may influence pineal NAT activity by a mechanism that involves β-adrenergic neural transmission.

Bioreduction of nitrate in groundwater using a pilot-scale hydrogen-based membrane biofilm reactor

A long-term pilot-scale H_(2)-based membrane biofilm reactor(mBfR)was tested for removal of nitrate from actual groundwater.A key feature of this secondgeneration pilot mBfR is that it employed lower cost polyester hollow fibers and still achieved high loading *** steady-state maximum nitrate surface loading at which the effluent nitrate and nitrite concentrations were below the maximum Contaminant Level(mCL)was at least 5.9 g·N·(m^(2)·d)^(–1),which corresponds to a maximum volumetric loading of at least 7.7 kg·N·(m^(3)·d)^(–1).The steady-state maximum nitrate surface area loading was higher than the highest nitrate surface loading reported in the firstgeneration mBfRs using composite fibers(2.6 g·N·(m^(2)·d)^(–1)).This work also evaluated the H_(2)-utilization efficiency in *** measured H_(2)supply rate was only slightly higher than the stoichiometric H_(2)-utilization ***,H_(2)utilization was controlled by diffusion and was close to 100%efficiency,as long as biofilm accumulated on the polyester-fiber surface and the fibers had no leaks.

Fundamental movement skills training to promote physical activity in children with and without disability： A pilot study

Background： A positive association between fundamental movement skills （FmS） and physical activity （PA） has been shown in previous research of children with and without disability. This pilot study explored a causal mechanism for such relationship, and hypothesized that when FmS proficiency is improved, enhanced PA uptake will be found in children with and without disability. It was further hypothesized that improving FmS proficiency will have a greater impact on children with disability than those without disability. methods： Participants include typically developing （TD） children without disability and children with cerebral palsy （CP）, who were allocated to FmS training groups （CP-FmS n = 12, TD-FmS n = 13） and control groups （CP-C n = 12, TD-C n = 13）. Training groups practiced five FmS （run, jump, kick, throw, catch） in weekly 45-min sessions for 4 weeks. Control groups had their regular physiotherapy （CP） or physical education （TD） sessions. FmS were evaluated using process- and product-oriented measures, and PA was measured using accelerometers, before and after training. Results： It was verified that training groups gained improvements in FmS while control groups did not. No significant changes in weekday PA were found. Increased weekend moderate to vigorous physical activity （mVPA） was found in the CP-FmS group, while decreased weekend sedentary time was found in the CP-FmS and TD-FmS groups. The percentages of participants who exceeded the minimum detectable change （mDC9o） in mVPA and sedentary time were larger in children with CP than in children without disability. Conclusion： The findings suggest that improved FmS proficiency could potentially contribute to heightened PA and decreased sedentary time during weekends for children. Such effect of improved FmS proficiency on PA appears to be greater in those with physical disability than in those without disability. It is recommended that the findings of this pilot study should be further examined in future

modelling Dust Emissions from a Source Using Dust monitoring and meteorological Data

This paper describes a study into the development of more robust dust emission factors by means of dust and meteorological monitoring. Emission factors for nuisance dusts in the literature are scarce, with estimates of dust output given for many processes in mass per unit area per year. Temporal variations and the extent and conditions in which maximum concentrations occur can therefore be impossible to predict with any accuracy. This investigation aims to improve predic-tions by “back calculating” emission levels based on dust monitoring around known dust sources. Nuisance dust and meteorological monitoring has been undertaken at a sand and gravel quarry in the UK for a consecutive period of two years. Sticky pad directional dust monitors were used to collect dust at eight locations at and around the site with meteorological data collected at an elec-tronic weather station within the site. Air quality modelling software (ADmS) was used to test emission factors from the European Environment Agency (EEA) and the US Environmental Protec-tion Agency (EPA) for emissions from mineral workings. Predictions were compared with the dust monitoring data to assess accuracy, with results showing limited poor correspondence (r2 mission predictions were poorest in winter;this is likely because most emis-sion calculations are not weather dependent and seasonal fluctuations will occur. Dust emission rate calculations were altered with respect to the dust monitoring data for one monitoring location on the mineral site boundary and the model was run again. Results were then tested at two different locations up to 200 m from the site boundary, with very positive correlations (r2 0.89) and similar maximum concentrations (mission rates can be produced in combination with site boundary sticky pad dust monitoring in order to accurately derive estimations elsewhe
