With recent climate warming trends, both the increase in thermal variance （i.e., diurnal temperature range; DTR） as well as increased mean temperature may impact many different organisms, especially poikilothermic invertebrates. Predictions of insect developmental rates using degree-days （thermal unit accumulations above the developmental base threshold of the insect） are based on daily mean temperatures, regardless of DTR. However, non-linearity and variance in the means and extremes are often ignored. The role of thermal variance （e.g., daily temperature extremes and DTR） was evaluated experimentally for two swallowtail butterfly sister species using a common day/night photoperiod of 18 ： 6 h photo ： scoto-phase and corresponding daytime thermophase and nighttime cryophase periods of 22 ： 22℃ （constant 22℃）, 24 ： 16℃, and 26 ： 10℃（all three treatments had the same daily mean and the same degree-day accumulations）. Although developmental rates ofpost-diapause pupae were largely unaffected for both species, our results show that sizes in P canadensis females （but not males） were smaller in the treatments with more variance （26℃ ： 10℃） compared to constant 22℃. Such potentially significant impacts of size reduction in P canadensis females were not observed in P glaucus males or females under the same series ofthermo-period treatments.

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.

Studies over the past decades have uncovered fusion genes, a class of oncogenes that provide immense diagnostic and therapeutic advantages because of their tumor-specific expression. Originally associated with hemotologic cancers, fusion genes have recently been discovered in a wide array of solid tumors, including sarcomas, carcinomas, and tumors of the central nervous system. Fusion genes are attractive as both therapeutic targets and diagnostic tools due to their inherent expression in tumor tissue alone. Therefore, the discovery and elucidation of fusion genes in various cancer types may provide more effective therapies in the future for cancer patients.

Development of ChiP-chip and ChlP-seq technologies has allowed genome-wide high-resolution profiling of chromatin-associated marks and binding sites for epigenetic regulators. However, signals for directing epigenetic modifiers to their target sites are not understood. In this paper, we tested the hypothesis that genome location can affect the involvement of epigenetic regulators using Chromatin Charting （CC） Lines, which have an identical transgene construct inserted at different locations in the Arabidopsis genome. Four CC lines that showed evidence for epigenetic silencing of the luciferase reporter gene were transformed with RNAi vectors individually targeting epigenetic regulators LHP1, MOM1, CMT3, DRD1, DRM2, SUVH2, CLF, and HD1. Involvement of a particular epigenetic regulator in silencing the transgene locus in a CC line was determined by significant alterations in luciferase expression after suppression of the regulator＇s expression. Our results suggest that the targeting of epigenetic regulators can be influenced by genome location as well as sequence context. In addition, the relative importance of an epigenetic regulator can be influenced by tissue identity. We also report a novel approach to predict interactions between epigenetic regulators through clustering analysis of the regulators using alterations in gene expression of putative downstream targets, including endogenous loci and transgenes, in epigenetic mutants or RNAi lines. Our data support the existence of a complex and dynamic network of epigenetic regulators that serves to coordinate and control global gene expression in higher plants.

Force Myography （FMG）, which monitors pressure or radial deformation of a limb, has recently been proposed as a po- tential alternative for naturally controlling bionic robotic prostheses. This paper presents an exploratory case study aimed at evaluating how FMG behaves when a person with amputation uses a hand prosthetic prototype. One volunteer （transradial amputation） participated in this study, which investigated two experimental cases： static and dynamic. The static case considered forearm muscle contractions in a fixed elbow and shoulder positions whereas the dynamic case included movements of the elbow and shoulder. When considering eleven different hand grips, static data showed an accuracy over 99%, and dynamic data over 86% （within-trial analysis）. The across-trial analysis, that takes into account multiple trials in the same data collection set, showed a meaningful accuracy respectively of 81% and 75% only for the reduced six grips setup. While further research is needed to increase these accuracies, the obtained results provided initial evidence that this technology could represent an in- teresting alternative that is worth exploring for controlling prosthesis.

Fragile X syndrome is the most common form of inherited mental retardation affecting up to 1 in 4000 individuals. The syn- drome is induced by a mutation in the FMR1 gene, causing a deficiency in its gene by-product FMRP. Impairment in the nor- mal functioning of FMRP leads to learning and memory deficits and heightened sensitivity to sensory stimuli, including sound （hyperacusis）. The molecular basis of fragile X syndrome is thoroughly understood; however, the neural mechanisms underly- ing hyperacusis have not yet been determined. As the inferior colliculus （IC） is the principal midbrain nucleus of the auditory pathway, the current study addresses the questions underlying the neural mechanism of hyperacusis within the IC of fragile X mice. Acute experiments were performed in which electrophysiological recordings of the IC in FMR1-KO and WT mice were measured. Results showed that Q-values for WT were significantly larger than that of FMR-1 KO mice, indicating that WT mice exhibit sharper tuning curves than FMR1-KO mice. We also found the ratio of the monotonic neurons in the KO mice was much higher than the WT mice. These results suggest that lack of FMRP in the auditory system affects the developmental maturation and function of structures within the auditory pathway, and in this case specifically the IC. The dysfunction ob- served within the auditory neural pathway and in particular the IC may be related to the increased susceptibility to sound as seen in individuals with fragile X syndrome. Our study may help on understanding the mechanisms of the fragile X syndrome and hyperacusis.
