The newly identified aquaporin coding sequences presented here pave the way for further insights into the plant–water relations in the commercial strawberry(Fragaria x ananassa).Aquaporins are water channel proteins that allow water to cross(intra)cellular membranes.In Fragaria x ananassa,few of them have been identified hitherto,hampering the exploration of the water transport regulation at cellular level.Here,we present new aquaporin coding sequences belonging to different subclasses:plasma membrane intrinsic proteins subtype 1 and subtype 2(PIP1 and PIP2)and tonoplast intrinsic proteins(TIP).The classification is based on phylogenetic analysis and is confirmed by the presence of conserved residues.Substrate-specific signature sequences(SSSSs)and specificity-determining positions(SDPs)predict the substrate specificity of each new aquaporin.Expression profiling in leaves,petioles and developing fruits reveals distinct patterns,even within the same(sub)class.Expression profiles range from leaf-specific expression over constitutive expression to fruit-specific expression.Both upregulation and downregulation during fruit ripening occur.Substrate specificity and expression profiles suggest that functional specialization exists among aquaporins belonging to a different but also to the same(sub)class.

AIM:To study the association between inflammatory bowel disease(IBD) and genetic variations in eosinophil protein X(EPX) and eosinophil cationic protein(ECP).METHODS:DNA was extracted from ethylene diamine tetraacetic acid blood of 587 patients with Crohn's disease(CD),592 with ulcerative colitis(UC) and 300 healthy subjects.The EPX405(G > C,rs2013109),ECP434(G > C,rs2073342) and ECP562(G > C,rs2233860) gene polymorphisms were analysed,by the 5'-nuclease allelic discrimination assay.For determination of intracellular content of EPX and ECP in granulocytes,39 blood samples was collected and extracted with a buffer containing cetyltrimethylammonium bromide.The intracellular content of EPX was analysed using an enzyme-linked immunosorbent assay.The intracellular content of ECP was analysed with the UniCAP system as described by the manufacturer.Statistical tests for calculations of results were χ 2 test,Fisher's exact test,ANOVA,Student-Newman-Keuls test,and Kaplan-Meier survival curve with Log-rank test for trend,the probability values of P < 0.05 were considered statistically significant.RESULTS:The genotype frequency for males with UC and with an age of disease onset of ≥ 45 years(n = 57) was for ECP434 and ECP562,GG = 37%,GC = 60%,CC = 4% and GG = 51%,GC = 49%,CC = 0% respectively.This was significantly different from the healthy subject's genotype frequencies of ECP434(GG = 57%,GC = 38%,CC = 5%;P = 0.010) and ECP562(GG = 68%,GC = 29%,CC = 3%;P = 0.009).The genotype frequencies for females,with an age of disease onset of ≥ 45 years with CD(n = 62),was for the ECP434 and ECP562 genotypes GG = 37%,GC =52%,CC = 11% and GG = 48%,GC = 47% and CC = 5% respectively.This was also statistically different from healthy controls for both ECP434(P = 0.010) and ECP562(P = 0.013).The intracellular protein concentration of EPX and ECP was calculated in μg/10 6 eosinophils and then correlated to the EPX 405 genotypes.The protein content of EPX was highest in the patients with the CC genotype of EPX405(GG = 4.65,GC = 5.93,and CC

Evaluation of Filter Material for Treatment of Different Types of Wastewater

This paper presents an evaluation of the suitability of a mixed absorbent based on peat and carbon-containing ash for treatment of wastewaters, such as wastewater from professional car washes, landfill leachate and stormwater. This mixture is very attractive, since it is a low-cost material which has a capability to simultaneously remove inorganic as well as organic pollutants. Since any filter material eventually needs to be replaced either due to saturation of pollutants or reduced infiltration capacity, it is important that the residual can be handled at low cost and that the environment will be not impaired. The tested mixture, used in filter beds, showed low leaching values and high simultaneous removal efficiency of metals as Cu, Cd and Pb, non-polar organic compounds such as PCBs. Polar organic compounds as phenols were also efficiently removed by microbial and/or chemical degradation in the studied treatment plants with the filter bed acted as a biofilter. Filter material used for three years in a full-scale plant for leachate treatment and four years in treatment plants for wastewater from car washes, had sufficiently high energy content indicating that energy recovery is a good alternative for handling after its usage. Results show that the presented filter material is excellent for both small scale applications (e.g. treatment systems for car wash wastewater with capacity between 250 - 3000 m3 per year) as well as large-scale applications (e.g. filter systems for landfill leachates with capacity above 30,000 m3 per year).

Daily life close to death involves physical, psychological, and social strain, exposing patients and their family members to major transitions affecting relational patterns and identity. For the individual family member, this often means sharing life with a changing person in a changing relationship, disrupting both individual identity and family identity. Our aim was to deepen the understanding of individual experiences that are important in constructing family identity close to death at home. We performed a secondary analysis of qualitative data collected through 40 interviews with persons with life-threatening illness and the family members who shared everyday life with them. The analysis resulted in interpretive descriptions which provided three patterns important for creating family identity, which we here call “we-ness” close to death. The patterns were: being an existential person, being an extension of the other, and being together in existential loneliness. Together, these three patterns seemed to play a part in the construction of family identity;we-ness, close to death. One important finding was the tension between the search for togetherness in “we-ness” while dealing with an existential loneliness, which seemed to capture an essential aspect of being a family of which one member is dying.

Aims and objectives: To explore any changes in nurses’ skills at communicating with residents with dementia disease when using the validation method, as observed in one-to-one videotaped conversations. Background: Communication difficulties due to cognitive impairment among residents with dementia disease may complicate care situations. Training can improve nurses’ communication skills and increase care quality. The validation method aims to facilitate communication with residents with dementia disease through empathic and confirmatory approaches. Evaluations of the validation method have primarily focused on the residents’ perspective, and reports on nurses’ experiences are sparse. Improved communication and relationships with residents after validation method training have been described previously. Videotaped data could provide additional information about these earlier results. Design: A descriptive qualitative design. Methods: Eight nurses participated in a year of validation method training, including videotaped conversations with eleven residents. Videotapes with at least five months between the first and last recording were analysed and compared qualitatively. Results: The analysis revealed an overall pattern: nurses’ movements within and between various paths when improving their communication skills. This was based on three sub-patterns: from controlling communication towards developing attentiveness in communication, from ambiguous communication towards developing coherence in communication, and from being open and attentive towards having a refined attuned communication. Conclusions: All nurses developed their communication skills during the programme, albeit to different degrees. The findings are in congruence with the experiences described by nurses, and so it is reasonable to believe that the programme helped to improve the nurses’ skills in communicating with residents with dementia disease. Objectives: To translate and evaluate the psychometric properties of the Swedish version of the Fear of Complications Questionnaire. Design: Cross-sectional study design and scale development. Settings: Totally, 469 adults (response rate 63.5%) with Type 1 diabetes completed the questionnaires. Participants were recruited from two university hospitals in Sweden. Participants: Eligible patients were those who met the following inclusion criteria: diagnosed with Type 1 diabetes, diabetes duration of at least 1 year and aged at least 18 years. Methods: The Fear of Complications Questionnaire was translated using the forward-backward translation method. Factor analyses of the questionnaire were performed in two steps using both exploratory and confirmatory factor analysis. Convergent validity was examined using the Hospital Anxiety and Depression Scale and the Fear of Hypoglycaemia Fear Survey. Internal consistency was estimated using Cronbach’s alpha. Results: Exploratory factor analysis supported a two-factor solution. One factor contained three items having to do with fear of kidney-related complications and one factor included the rest of items concerning fear of other diabetes-related complications, as well as fear of complications in general. Internal consistency was high Cronbach’s alpha 0.96. The findings also gave support for convergent validity, with significant positive correlations between measures (r = 0.51 to 0.54). Conclusion: The clinical relevance of the identified two-factor model with a structure of one dominant subdomain may be considered. We suggest, however a one-factor model covering all the items as a relevant basis to assess fear of complications among people with Type 1 diabetes.
