State-of-the-Art management of knee osteoarthritis

Osteoarthritis(OA) is the most common type of arthritis found in the United States' population and is also the most common disease of joints in adults throughout the world with the knee being the most frequently affected of all joints. As the United States' population ages along with the increasing trends in obesity prevalence in other parts of the world, it is expected that the burden of OA on the population, healthcare system, and overall economy will continue to increase in the future without making major improvements in managing knee OA. Numerous therapies aim to reduce symptoms of knee OA and continued research has helped to further understand the complex pathophysiology of its disease mechanism attempting to uncover new potential targets for the treatment of OA. This review article seeks to evaluate the current practices for managing knee OA and discusses emerging therapies on the horizon. These practices include non-pharmacological treatments such as providing patient education and self-management strategies, advising weight loss, strengthening programs, and addressing biomechanical issues with bracing or foot orthoses. Oral analgesics and anti-inflammatories are pharmacologicals that are commonly used and the literature overall supports that some of these medications can be helpful for managing knee OA in the short-term but are less effective for long-term management. Additionally, more prolonged use significantly increases the risk of serious associated side effects that are not too uncommon. Diseasemodifying osteoarthritis drugs are being researched as a treatment modality to potentially halt or slow disease progression but data at this time is limited and continued studies are being conducted to further investigate their effectiveness. The realization of a mixed-phase microstructure in strained BiFeO_(3)(BFO)thin films has led to numerous novel effects derived from the coexistence of the tetragonal-like monoclinic phase(T phase)and rhombohedral-like monoclinic phase(R phase).Strong strain and polarization diiferences between the phases should result in a high level of transformation plasticity,which enables the continuous alteration of the relative proportion of R and T states in response to external *** the potential for utilizing such plasticity to control mixed-phase populations under external stimuli is evident,direct experi・mental evidence backed by equilibrium predictions has not yet been fully *** we demonstrate deterministic control of mixed-phase populations in an epitaxially strained BFO thin film through the application of localized stresses and electric fields in a reversible manne匚The results illustrate and rationalize deterministic control of mixed phases in strained BFO films,which could be crucial in tuning their functional *** findings also highlight a new multiparametric technique in the scanning probe lithography toolbox based on tip-assisted electric and strain field manipulation of functional properties that might find application beyond the ferroelectric domain and structural phase lithography.

Obesity-induced sperm DNA methylation changes at satellite repeats are reprogrammed in rat offspring

There is now strong evidence that the paternal contribution to offspring phenotype at fertilisation is more than just DNA. However, the identity and mechanisms of this nongenetic inheritance are poorly understood. One of the more important questions in this research area is： do changes in sperm DNA methylation have phenotypic consequences for offspring？ We have previously reported that offspring of obese male rats have altered glucose metabolism compared with controls and that this effect was inherited through nongenetic means. Here, we describe investigations into sperm DNA methylation in a new cohort using the same protocol. Male rats on a high-fat diet were 30% heavier than control-fed males at the time of mating （16-19 weeks old, n = 14/14）. A small （0.25%） increase in total 5-methyl-2＇-deoxycytidine was detected in obese rat spermatozoa by liquid chromatography tandem mass spectrometry. Examination of the repetitive fraction of the genome with methyI-CpG binding domain protein-enriched genome sequencing （MBD-Seq） and pyrosequencing revealed that retrotransposon DNA methylation states in spermatozoa were not affected by obesity, but methylation at satellite repeats throughout the genome was increased. However, examination of muscle, liver, and spermatozoa from male 27-week-old offspring from obese and control fathers （both groups from n = 8 fathers） revealed that normal DNA methylation levels were restored during offspring development. Furthermore, no changes were found in three genomic imprints in obese rat spermatozoa. Our findings have implications for transgenerational epigenetic reprogramming. They suggest that postfertilization mechanisms exist for normalising some environmentally-induced DNA methylation changes in sperm cells.

Disappearance and appearance of an indigestible marker in feces from growing pigs as affected by previous-and currentdiet composition

Background： Indigestible markers are commonly utilized in digestion studies, but the complete disappearance or maximum appearance of a marker in feces can be affected by diet composition, feed intake, or an animal＇s *** objectives of this study were to determine the impact of previous（Phase 1, P1） and current-（Phase 2, P2）diet composition on marker disappearance（Cr） and appearance（Ti） in pigs fed 3 diets differing in NDF ***： When pigs were maintained on the 25.1, 72.5, and 125.0 g/kg NDF diets, it took 5.1, 4.1, and 2.5 d, respectively,for Cr levels to decrease below the limit of quantitation; or 4.6, 3.7, or 2.8 d, respectively, for Ti to be maximized. These effects were not, however, independent of the previous diet as indicated by the interaction between P1 and P2 diets on fecal marker concentrations（P 〈 0.01）. When dietary NDF increased from P1 to P2, it took less time for fecal Cr to decrease or fecal Ti to be maximized（an average of 2.5 d）, than if NDF decreased from P1 to P2 where it took longer for fecal Cr to decrease or fecal Ti to be maximized（an average of 3.4 d）.Conclusions： Because of the wide range in excretion times reported in the literature and improved laboratory methods for elemental detection, the data suggests that caution must be taken in considering dietary fiber concentrations of the past and currently fed diets so that no previous dietary marker addition remains in the digestive tract or feces such that a smal amount of maker is present to confound subsequent experimental results, and that marker concentration have stabilized when these samples are col ected.

Surface energy-mediated fibronectin adsorption and osteoblast responses on nanostructured diamond

Nanostructured diamond have potential applications in many biomedical related fields and demonstrated extraordinary capacity to influence cellular responses. Studying the surface property of nanodiamond and its influence to protein adsorption and subsequent cellular responses along with the mechanism behind such capacity becomes more important. Here the role of surface energy associated with nanostructured diamond in modulating fibronectin and osteoblast(OB, bone forming cells) responses was investigated. Nanocrystalline diamond(NCD) and submicron crystalline diamond(SMCD) films with controllable surface energy were prepared by microwave-enhanced plasma chemical vapor deposition(MPCVD) techniques. Fibronectin adsorption on the diamond films with varied surface energy values was measured via the enzyme-linked immunosorbent assay(ELISA) and the relationship between the surface energy and fibronectin adsorption was studied. The result indicated that fibronectin adsorption on nanostructured surfaces was closely related to both surface energy and material microstructures. The spreading and migration of OB aggregates(each containing 30–50 cells) on the NCD with varied surface energy values were also studied. The result indicates a correlation between the cell spreading and migration on nanodiamond and the surface energy of nanostructured surface.

Ultrafast Preparation of Nonequilibrium FeNi Spinels by Magnetic Induction Heating for Unprecedented Oxygen Evolution Electrocatalysis

Carbon-supported nanocomposites are attracting particular attention as high-performance,low-cost electrocatalysts for electrochemical water *** are mostly prepared by pyrolysis and hydrothermal procedures that are time-consuming(from hours to days)and typically difficult to produce a nonequilibrium ***,for the first time ever,we exploit magnetic induction heating-quenching for ultrafast production of carbon-FeNi spinel oxide nanocomposites(within seconds),which exhibit an unprecedentedly high performance towards oxygen evolution reaction(OER),with an ultralow overpotential of only+260 mV to reach the high current density of 100 mA cm^(-2).Experimental and theoretical studies show that the rapid heating and quenching process(ca.10^(3)K s^(-1))impedes the Ni and Fe phase segregation and produces a Cl-rich surface,both contributing to the remarkable catalytic *** from this study highlight the unique advantage of ultrafast heating/quenching in the structural engineering of functional nanocomposites to achieve high electrocatalytic performance towards important electrochemical reactions.

Extrapolating plot-scale CO_(2) and ozone enrichment experimental results to novel conditions and scales using mechanistic modeling
