Consumer Perceptions of Noni j.ice Health Benefits during a 90-Day In-Home Use Test

Noni j.ice, made from the fruit of Morinda citrifolia, is a health-promoting food from the Pacific Islands with an extensive history of traditional use. Human clinical trials have revealed that noni j.ice has antioxidant, immunomodulatory and anti-inflammatory properties. Previously, regular noni j.ice consumers in Europe and traditional healers in Fij. were surveyed to identify potential noni j.ice health benefits. But to better understand the perception of its health benefits among those with no previous experience with noni j.ice, an in-home study was conducted in the United States. Three hundred and nineteen adults with untreated chronic health conditions were provided with a 90-day supply of a blinded (unlabeled) commercial noni j.ice product (Tahitian Noni® j.ice), instructed to consume 4 fluid ounces (1/2 cup) each day, and responded to a questionnaire given on days 30, 60 and 90 to evaluate product appeal and perceived effectiveness. A total of 299 people (94%) felt that their health improved in some way after drinking TNj.for 90 days. More than 80% of the subj.cts reported feeling better or healthier in general. Most participants also concurred with more than two dozen additional positive health perception statements, especially on days 60 and 90. More than 70% of consumers also indicated that noni j.ice helped them feel better than other wellness supplements they had tried in the past, including other so-called “super fruit” products. These findings seem to corroborate the results of clinical trials and previous consumer surveys regarding the potential for noni j.ice to provide discernable health benefits when used consistently over time. Such findings may help guide additional placebo-controlled clinical trials in which specific effects may be evaluated more rigorously.

The Influence of Morinda citrifolia(Noni) Fruit j.ice on Collagen Deposition in the Skin: A Minireview

The fruit of Morinda citrifolia, commonly known as noni, has an extensive history of use as a food and medicine throughout the tropics. Among its many uses, noni was believed to promote skin health. In vivo wound healing studies reveal that noni fruit j.ice ingestion increases collagen production and deposition via increased expression of extracellular matrix protein genes. Noni j.ice also appears to promote fibroblast proliferation. In vitro studies indicate that noni j.ice also protects fibroblasts via activation of the transcription factor Nrf2 and protects extracellular matrix collagen by inhibiting matrix metalloproteinases. In vitro and in vivo study findings are corroborated by a clinical trial where hospitalization times were reduced, and wound granulation quality was improved in pediatric burn patients. The findings of other human studies also indicate that noni j.ice has the potential to protect skin collagen. These include reduced plasma reactive oxygen species in cigarette smokers, lowered skin glycation levels in overweight and obese adults, as well as reduced skin glycation levels among noni j.ice consumers within the general population. These in vitro, in vivo and human studies reveal that there are multiple mechanisms of action through which noni j.ice promotes collagen synthesis and inhibits collagen degradation. The procollagen properties of noni j.ice aid wound healing, increase skin elasticity, and improve the overall appearance of the skin.

Morinda citrifolia(Noni) Fruit j.ice Inhibits SARS-CoV-2 Spike Protein Binding of Angiotensin-Converting Enzyme 2 (ACE2)

COVID-19 is a global pandemic that has claimed millions of lives. This disease is caused by a coronavirus, SARS-CoV-2, which requires the binding of its spike protein to angiotensin-converting enzyme 2 (ACE2) for infection of the host cell. Morinda citrifolia (noni) fruit j.ice has antiviral activity that involves enhancement of immune system function. SARS-CoV-2 spike-ACE2 interaction experiments were carried out to further investigate the antiviral properties of noni j.ice and its maj.r iridoids. Noni j.ice inhibited binding by approximately 69%. Scandoside was the most active of the three iridoids evaluated, reducing average spike protein-ACE2 interaction by 79.25%. The iridoids worked synergistically towards inhibiting spike protein binding when assayed together, improving activity by more than 22% above the expected level. But the modest activity of the most abundant iridoid, deacetylasperulosidic acid, indicates that other phytochemicals (i.e. scopoletin, quercetin, rutin and kaempferol) are also involved. Our results suggest that the presence of several biological active phytochemicals in noni j.ice enhances resistance to SARS-CoV-2 by interfering with its ability to bind ACE2. This is a new and significant anti-viral mechanism of noni j.ice that does not directly involve its immunomodulatory properties.

A Face Serum Containing Palmitoyl Tripeptide-38, Hydrolyzed Hyaluronic Acid, Bakuchiol and a Polyherbal and Vitamin Blend Improves Skin Quality

A face serum composed of a combination of biologically active compounds was evaluated for safety and efficacy in vitro, in a repeat insult patch test and in a human clinical efficacy trial. The serum inhibited tyrosinase activity modestly, decreased collagenase activity and exhibited notable free radical scavenging activity in vitro. It is gentle to the skin, as the serum did not irritate the skin or produce symptoms of allergic contact dermatitis in the 55 healthy adults that participated in the repeat insult patch test. In the efficacy trial, daily application of the face serum for 30 days significantly increased skin hydration, with all 35 volunteers experiencing improvement. Substantial improvements in skin elasticity, roughness (fine lines and wrinkles), and brightness also occurred during the trial. Dermatological examination also revealed a trend for reduced comedone count with use of the serum. Self-assessment responses revealed that all volunteers experienced improvements in multiple skin quality parameters and that participant perceptions are consistent with the results of the instrumental analyses. These findings indicated that the measured improvements in skin quality are not only statistically significant but are also clinically relevant as they were great enough for users of the face serum to feel and recognize.

Vitamin C, Grape Seed Extract and Citrus Bioflavonoids Protect the Skin against Photoaging: A Review

The skin is a maj.r protective organ of the body. It is constantly exposed to the environment and is very resilient. But exposure to ultraviolet (UV) rays from the sun results in the production of reactive oxygen species (ROS) and subsequent inflammatory responses that can overwhelm the innate protective mechanisms of the skin. This results in damage and premature aging. Strategies to mitigate this premature photoaging might include avoidance of sunlight. However, some sunlight exposure is beneficial to health. One notable example of this is the production of vitamin D. A more practical approach to preventing adverse effects of UV light in the skin is antioxidant supplementation. Dietary antioxidants may help control ROS propagation following UV light exposure. To further evaluate the utility of antioxidants in protecting the skin, in vitro, in vivo and human studies of three well known dietary antioxidants are reviewed and discussed. The data clearly demonstrate that vitamin C, grape seed extract and citrus bioflavonoids have the potential to reduce the damaging effects of excess sun exposure via antioxidant, anti-inflammatory and immunomodulating mechanisms. As such, regular ingestion of dietary antioxidants appears to be a useful strategy for protecting the skin against photoaging.

Lignans from Morinda citrifolia(Noni) Fruit Inhibit Fatty Acid Amide Hydrolase and Monoacylglycerol Lipase in Vitro

Morinda citrifolia (noni) fruit j.ice has exhibited a variety of biological activities in human clinical trials, indicating that it influences multiple systems of the body. Since the 1990s, the endocannabinoid system (ECS) has been found to modulate the activity of other organ systems. To investigate noni’s potential impact on the ECS, extracts from freeze-dried noni fruit were evaluated in fatty acid amide hydrolase (FAAH) and monoacylglycerol lipase (MAGL) inhibition assays. The ethyl acetate extract demonstrated the greatest activity against both enzymes. Lignans in this extract also inhibited enzyme activities, with americanin A being the most active in both assays. Americanoic acid and 3,3’-bisdemethylpinoresinol were the next most active compounds. These results suggest that lignans in noni fruit may influence endocannabinoid levels within the body via FAAH and MAGL inhibition. This reveals another set of probable mechanisms of action by which noni j.ice affects human health.

Morinda citrifolia (noni) fruit j.ice has been shown to have a wide variety of potential health benefits in human clinical trials. It may also influence the endocannabinoid system of the body. Since the main ingredient of the product studied in these clinical trials was j.ice made from noni fruit puree from French Polynesia, it was evaluated for its ability to inhibit the two maj.r endocannabinoid degradation enzymes in vitro. Noni fruit j.ice inhibited both fatty acid amide hydrolase (FAAH) and monoacylglycerol lipase (MAGL) in a concentration-dependent manner, suggesting that it may help maintain anandamide and 2-arachidonoylglycerol levels. Samples of the puree were also analyzed for the presence of characteristic phytochemical markers of authentic noni fruit such as scopoletin, rutin, quercetin, deacetylasperulosidic acid and asperulosidic acid, all of which were present. Also present was scandoside, which is reported for the first time as being identified in noni fruit or its j.ice. Some of these compounds may contribute to the FAAH and MAGL inhibiting activity of noni j.ice. These results reveal another set of mechanisms by which noni j.ice possibly supports mental health, maintains j.int health, relieves discomfort and modulates the immune system.

Noni Seed Oil Topical Safety, Efficacy, and Potential Mechanisms of Action

Background: Noni fruit and leaf j.ices were used to treat acne in Polynesian folk medicine. However, the potential benefit of noni seed oil (NSO) for human skin health is not as well known. Obj.ctive: The topical safety of NSO was evaluated in a repeat insult patch test with 49 adult volunteers. Comedogenicity was evaluated in 23 adolescent volunteers for four weeks. Anti-inflammatory activity was determined in-vitro with COX-2 and 5-LOX enzyme inhibition assays. Results: No reactions to NSO were observed in any of the volunteers. NSO also reduced the number of open and closed comedones in the comedogenicity test. NSO inhibited COX-2 and 5-LOX enzymes in a concentration dependent manner, but with COX-2 inhibition being more pronounced than 5-LOX. Conclusion: Noni seed oil is safe for topical use and is non-comedogenic. It also has potential anti-inflammatory activity, being a dual inhibitor of COX-2 and 5-LOX enzymes. These results indicate the potential utility of NSO for skin care applications, as well as warrant further investigation of efficacy in 5-LOX and COX-2 related pathophysiological conditions.

Retrospective Assessment of Changes in Perceived Health Status during Continuous Use of a Dietary Supplement

Dietary factors play an important role in human health. Specifically, nutritional interventions may have an impact on the hypothalamic-pituitary axis resulting in positive health outcomes. A retrospective health status survey of 386 respondents was conducted to investigate the potential of a dietary supplement, NutrifiiTM Renew, for improving various health outcomes in comparison to a multivitamin/mineral control group. Among those who ingested Renew, 95% reported experiencing at least some improvement in one or more health categories. Compared to the multivitamin/mineral group, the Renew group reported earlier and/or greater improvements in several health status categories including sleep, reproductive health, memory, energy/stamina, skin and hair quality, flexibility, exercise recovery and feeling rested in the morning. A clear maj.rity (84.9%) of Renew consumers reported experiencing noticeable improvement in some aspect of their health within one month. Among those who felt an improvement in sleep, most reported experiencing a change within one week. These improvements were likely due to the synergistic effects of L-arginine, L-citrulline, L-lysine, and botanical extracts on nitric oxide production, growth hormone (GH) release, and sleep quality. The observed improvements in skin elasticity and hair growth may be related to increased GH levels and IGF-1 production. Overall, these findings suggest that Renew may have potential as a dietary intervention for improving various health outcomes beyond those provided by multivitamin/mineral supplementation alone. Further studies are needed to confirm these findings and elucidate the underlying mechanisms of action.
