SARS-coV-2 productively infects primary human immune system cells in vitro and in coviD-19 patients

The severe acute respiratory syndrome coronavirus 2(SARS-covV-2)infection is associated with a hyperinflammatory state and lymphocytopenia,a hallmark that appears as both signature and prognosis of disease severity *** cytokine storm and a sustained inflammatory state are commonly associated with immune cell depletion,it is still unclear whether direct SARS-cov-2 infection of immune cells could also play a role in this scenario by harboring viral *** found that monocytes,as well as both B and T lymphocytes,were susceptible to SARS-cov-2 infection in vitro,accumulating double-stranded RNA consistent with viral RNA replication and ultimately leading to expressive T cell *** addition,flow cytometry and immunofluorescence analysis revealed that SARS-cov-2 was frequently detected in monocytes and B lymphocytes from coronavirus disease 2019(coVID-19)*** rates of SARS-cov-2-infected monocytes in peripheral blood mononuclear cells from coVID-19 patients increased over time from symptom onset,with SARS-coV-2-positive monocytes,B cells,and cD4+T lymphocytes also detected in postmortem lung *** results indicated that SARS-coV-2 infection of blood-circulating leukocytes in covID-19 patients might have important implications for disease pathogenesis and progression,immune dysfunction,and virus spread within the host.

In this work,we report long-term trends in the abundance and breeding performance of Adélie penguins(Pygoscelis adeliae)nesting in three Antarctic colonies(i.e.,at Martin Point,South Orkneys Islands;Stranger Point/cabo Funes,South Shetland Islands;and Esperanza/Hope Bay in the Antarctic Peninsula)from 1995/96 to 2022/*** yearly count data of breeding groups selected,we observed a decline in the number of breeding pairs and chicks in crèche at all colonies ***,the magnitude of change was higher at Stranger Point than that in the remaining ***,the index of breeding success,which was calculated as the ratio of chicks in crèche to breeding pairs,exhibited no apparent trend throughout the study ***,it displayed greater variability at Martin Point compared to the other two colonies under *** the number of chicks in crèche of Adélie penguins showed a declining pattern,the average breeding performance was similar to that reported in gentoo penguin colonies,specifically,those undergoing a population increase(even in sympatric colonies facing similar local conditions).consequently,it is plausible to assume a reduction of the over-winter survival as a likely cause of the declining trend observed,at least in the Stranger Point and Esperanza ***,we cannot rule out local effects during the breeding season affecting the Adélie population of Martin Point.

BAcKGROUND Autoimmunity has emerged as a probable disease modifier in patients with clinically diagnosed type 2 diabetes mellitus(T2DM),that is,patients who have insulin resistance,obesity,and other cardiovascular risk factors,suggesting that the presence of glutamic acid decarboxylase(anti-GAD65),islet antigen 2(anti-IA2),and zinc transporter 8(anti-Zn8T)antibodies could have deleterious effects on beta cell function,causing failure and earlier requirement for insulin *** To evaluate anti-GAD65,anti-IA2 and anti-Zn8T as predictors of early insulin requirement in adolescents with a clinical diagnosis of *** This was a case–control study in patients with clinically diagnosed with T2DM(68 cases and 64 controls with and without early insulin dependence respectively),male and female,aged 12–18 ***,blood pressure,glucose,insulin,c-peptide,glycated hemoglobin A1c,and lipid profiles were *** was used to measure anti-GAD65,anti-IA2,and anti-Zn8T *** statistics,Pearson'sχ2 test,Student's t test,and logistic regression was performed.Pconsidered statistically *** There were 132 patients(53.8%female),with a mean age was 15.9±1.3 years,and there was a disease evolution time of 4.49±0.88 *** presence of anti-GAD65,anti-IA2,and anti-Zn8T positivity was found in 29.5%,18.2%,and 15.9%,*** the groups by early or no insulin dependence showed that the group with insulin had a higher frequency of antibody positivity:anti-GAD65 odds ratio(OR):2.42(1.112–5.303,P=0.026);anti-IA2:OR:1.55(0.859–2.818,P=0.105);and anti-Zn8T:OR:7.32(2.039–26.279,P=0.002).cONcLUSION Anti-GAD65 positivity was high in our ***-GAD65 and anti-Zn8T positivity showed a significantly depleted beta cell reserve phenotype,leading to an increased risk of early insulin dependence.

Loggerhead Sea Turtles Hand-Reared in captivity: Isotopic Insights into Their Inherent Dietary Variation

Stable isotope analysis is a widely used method for gathering ecological insights into the diet and feeding habitats of various species. While captive studies often limit lethal sampling and differ from wild conditions, they offer valuable insights into inherent isotopic variations among individuals, which are often assumed to reflect differences between natural populations. In the Sea Turtle conservation Program, loggerhead turtle hatchlings from different nests were fed. Necropsies were conducted on turtles that died during this period, obtaining bone fragments for analysis. We evaluated the isotopic variation of carbon (δ13c) and nitrogen (δ15N) in bone tissue across six turtle nests (n = 66 samples) and assessed differences in Straight carapace Length (ScL, n = 71 samples). Using SIBER and nicheROVER in R, we calculated niche width and overlap, while the simmr package determined primary prey assimilation. Despite feeding the hatchlings the same prey, we observed variations in nitrogen isotope assimilation between nests. Nests 4 and 6 had a niche width >1.8‰, indicating consistent consumption frequencies across all prey and >70% niche overlap with other nests. In contrast, nests 1 and 2 showed a narrower niche width (Mugil sp. constituted the primary diet component (>40%) across all groups. This study demonstrates how factors like competition or prey preference can influence the assimilation of diet, even when the source remains constant (inherent variation).

The following paper reports on a comparative study of the effects of two types of carbon nanotubes,namely multiwall(MWcNT)and single-wall(SWcNT)carbon nanotube,on the properties of 3D-printed parts produced with acrylonitrile-butadiene-styrene(ABS)nanocomposites with various cNT loadings of5-10 wt.%.Quasi-static tensile properties and Vicat softening temperature of 3D-printed parts were enhanced with the increasing cNT *** highest enhancement in tensile properties was observed for the ABS/cNT nanocomposites at 10 wt.%filler loading.3D-printed ABS/SWcNT composites showed higher tensile modulus,better creep stability and higher Vicat ***,the strength of ABS/SWcNT 3D samples is relatively lower than that of ABS/*** addition,3D-printed parts exhibited anisotropic electrical conductive behaviour,which has a conductivity of through-layer of about2-3 orders of magnitude higher than *** highest conductivity of 3D-printed samples reached25.2 S/m,and 9.3 S/m for ABS/MWcNT and ABS/SWcNT composites at 10 wt.%,*** results obtained,*** successful fuse filament fabrication and the consequent electromechanical properties,confirm that these 3D printable nanocomposite could be properly utilized for the production,and application up to about 90°c,of thermoelectric devices and/or resistors for flexible circuits.

Body temperature,activity patterns and hunting in free-living cheetah:biologging reveals new insights

As one of the few felids that is predominantly diurnal,cheetahs(Acinonyx jubatus)can be exposed to high heat loads in their natural *** is known about long-term patterns of body temperature and activity(including hunting)in cheetahs because long-term concurrent measurements of body temperature and activity have never been reported for cheetahs,or,indeed,for any free-living *** report here body temperature and locomo-tor activity measured with implanted data loggers over 7 months in 5 free-living cheetahs in *** tem-perature ranged from a maximum of 39°c in summer to−2°c in *** had higher(~0.4°c)max-imum 24-h body temperatures,later acrophase(~1 h),with larger fluctuations in the range of the 24-h body temperature rhythm(approximately 0.4°c)during a hot-dry period than during a cool-dry period,but main-tained homeothermy irrespective of the climatic *** ambient temperatures increased,the cheetahs shifted from a diurnal to a crepuscular activity pattern,with reduced activity between 900 and 1500 hours and increased nocturnal *** timing of hunts followed the general pattern of activity;the cheetahs hunted when they were on the *** hunted if an opportunity presented itself;on occasion they hunted in the midday heat or in total darkness(new moon).Biologging revealed insights into cheetah biology that are not ac-cessible by traditional observer-based techniques.

Isolation by phage display and characterization of a single-chain antibody specific for O^6-methyldeoxyguanosine

New approaches of making single chain Fv antibodies against O6-methyl-2’ -deoxyguanosine (O6MdG) have been demonstrated by using the phage antibody display system. Using O6MdG as an antigen, 21 positive clones were identified by ELISA from this library, one of which, designated H3, specifically binds to O6MdG with high affinity. The H3 scFv antibody has an affinity constant (Kaff) of 5.94×1011(mol/L)-1. H3 scFv has been successfully used to detect O6MdG in DNA hydrolyses from yeast or E. coli cells treated with a DNA methylating agent. To our knowledge, this is the first report of the selection of a specific scFv against DNA adducts. The results demonstrate the potential applications of the phage display technology for the detection of DNA lesions caused by mutagens and carcinogens.

A cosmic Dimension： c. S. Lewis＇s Engagement with the Politics of Power and the challenge of Jesus

This paper focuses on some literary expressions of the role of faith in our engagement with things political from the pen of c. S. Lewis, whose writings are distinguished by a combination of intellectual force and imaginative flair. In an increasingly secular climate, he sought to bring God back into the equation, and it is not surprising to find some current issues addressed within his imaginative representations of a vibrant spiritual reality. His ideas and ideology are inextricably linked to his christian faith. Several critical essays reflect his interest in both theology and political issues including： the ethical use of technology, civil liberties, ecology, pacifism, and animal rights. This material forms the layers of his science-fiction trilogy which includes themes of spiritual warfare, factions, faith, and a fairy tale ending. His theology was not confined to the disciplines of a particular school of thought and his approach to things political follows no party line. His critical essays and stories are concerned with the practicalities of life, but are never reduced to a merely social gospel; they always have a cosmic dimension.

Midwifery Students’ confidence in New-Born care at completion of Training in Selected Midwifery Schools in Zambia. A Multicentric Study: A case of Zambia

Background: Quality education is one of the important factors in empowering midwives to provide quality health care services to mothers and newborns irrespective of their practice settings. Due to lack of standardization of the duration, content, and structure of midwifery education and different pathways to midwifery across the world, midwives may lack the confidence to deliver quality health care services in the main domain of the IcM competences. Aim: To assess the confidence of final year midwifery students in the domain of new-born care based on the IcM midwifery competencies. Methods: The first phase consisted of a quick situation analysis of midwifery education and regulation in Zambia, followed by a cross-sectional survey through a self-administered questionnaire to assess the confidence of final year midwifery students. Results: Most (51.9%) of the respondents were aged between 18 and 28 years, and majority of them were placed in Levels 1, 2 and 3 Hospitals and also clinics for their clinical practice, with between one hundred to ten thousand annual births, followed by 16.9% who were placed in Level 3 hospitals only. Most students (38.3%) reported receiving classroom instruction by both Lecture and Demonstration. Few students (22.7%) responded that they were not satisfied with their clinical supervision, while 77.3% were satisfied. There was significant association between confidence to perform skills independently and having enough time to practice on models in the labs (p value = 0.024), confidence when practiced for the first time on mothers (p value = 0.007) and satisfaction with supervision during clinical practice (p value = 0.000). Binary logistic regression of characteristics associated with confidence to perform new-born skills independently revealed significant association between satisfaction with clinical supervision and confidence to perform skills independently, 95% cI [1.83,
