Use of a lyophilized bovine pericardium graft to repair tunical defect in patients with Peyronie＇s disease： experience in a clinical setting

Many different materials are available for grafting during surgery for Peyronie＇s disease （PD）. To study the outcomes associated with the use of a lyophilized bovine pericardium graft （Peri-Guard） to repair tunical defects in patients with PD, descriptive study of 43 patients with disabling penile curvature was performed. Curvature was measured before and after the intervention using a standardized procedure. Surgical technique was plaque excision and grafting of the tunical defect carried out by a single surgeon. A lyophilized bovine pericardium patch was used for grafting. We measured the change in penile curvature and length, the ability to perform successful sexual intercourse without further pharmacological or surgical treatment, and the satisfaction with treatment. Follow-up data were available for a total of 41 patients. Median age was 50 years （IQR 48-52）; mean follow-up time was 14 ± 6 months. Complete penile straightening was achieved in 33 patients （80.5%） and five patients （12.2%） had curvature under 20. Three months after surgery and stretching therapy, mean （s.d.） flaccid penile length increased from 11.2 ± 2.8 cm to 12.1 ± 2.9 cm （P = 0.062）. Seventy-five percentage of patients reported being able to achieve successful intercourse without further treatment. Eighty-five percentage of patients reported to be satisfied with the treatment. There was no evidence of tissue rejection or infection, and only four patients suffered an adverse event. Plaque excision and grafting of the tunical defect with lyophilized bovine pericardium seems to be a safe and effective treatment for patients with PD.

DET1-mediated COP1 regulation avoids HY5 activity over second-site gene targets to tune plant photomorphogenesis

DE-ETIOLATED 1(DET1)and CONSTITUTIVE PHOTOMORPHOGENESIS 1(COP1)are two essential repressors of Arabidopsis photomorphogenesis.These proteins can associate with CULLIN4 to form independent CRL4-based E3 ubiquitin ligases that mediate the degradation of several photomorphogenic transcription factors,including ELONGATED HYPOCOTYL 5(HY5),thereby controlling multiple gene-regulatory networks.Despite extensive biochemical and genetic analyses of their multi-subunit complexes,the functional links between DET1 and COP1 have long remained elusive.Here,we report that DET1 associates with COP1 in vivo,enhances COP1-HY5 interaction,and promotes COP1 destabilization in a process that dampens HY5 protein abundance.By regulating its accumulation,DET1 avoids HY5 association with hundreds of second-site genomic loci,which are also frequently targeted by the skotomorphogenic transcription factor PHYTOCHROME-INTERACTING FACTOR 3.Accordingly,ectopic HY5 chromatin enrichment favors local gene repression and can trigger fusca-like phenotypes.This study therefore shows that DET1-mediated regulation of COP1 stability tunes down the HY5 cistrome,avoiding hyper-photomorphogenic responses that might compromise plant viability.

Comparison of the patient and partner satisfaction with 700CX and Titan penile prostheses

Most widespread three-component penile prosthesis models are 700CXTM and Titan~. Our purpose is to assess patient and partner satisfaction after the first implant. This is a multicenter, retrospective, nonrandomized study in which all patients who met the inclusion criteria between 2009 and 2013 were included. In total, 248 patients agreed to participate. To evaluate patient satisfaction, a validated but modified 11-question questionnaire was completed （EDITS）; and a nonvalidated two-item questionnaire was given to the partner. Statistical analysis used an ordinal logistic regression model. Two hundred and forty-eight patients （194 with 700CXTM vs 54 with Titan） and 207 couples completed the questionnaire （165 with 700CXTM vs 42 with Titan）. Overall satisfaction was high. Both showed great reliability for sexual intercourse and high compliance with prior expectations. Most patients were able to manage the penile prosthesis correctly within 6 months. Postoperative penile shortening led to some dissatisfaction in 42% and 46% of cases （700CX/Titan）. Significant differences were found in three questions of patients＇ questionnaire. There were more patients satisfied with the 700CXTM （P= 0.0001）. No patient with Titan implant took longer than 6 months to optimal management. Only 4% of patients with 700CXTM implant were dissatisfied with the deflation, in contrast to 24% with the Titan （P = 0.0031）. Of the two partners＇ questions, one showed a statistically significant difference （P = 0.0026）. It seems that group 700CX＂ would recommend to re-implant the prosthesis with a greater tendency. The overall satisfaction was very high for both prostheses. The final aspect of the erected and flaccid penis was satisfactory, but both groups showed significant discontent with its final size. Partners＇ overall satisfaction was high.

Physical and chemical characterization of sediments from an Andean river exposed to mining and agricultural activities: The Moquegua River, Peru

This study presents an analysis of sediments in an Andean river impacted by both natural conditions and anthropogenic activities. Fifty samples were collected from selected sites throughout the Moquegua River drainage basin, and Tambo River headwaters at Pasto Grande, in Peru, and analysed with X-ray diffraction, energy dispersive X-ray spectroscopy, scanning electron microscopy, Fourier-transform infrared spectroscopy and thermogravimetric analysis. Si, Ca, Al, Fe, and O, common constituents of soils, were dominant, along with K, N, and P. The latter originating as primary macronutrients or chemical residuals from fertilizers used in agriculture. Higher concentrations of macronutrients and organic components were found in agricultural and urban areas, respectively. Arsenic minerals were also detected, which occur naturally, but possibly at levels augmented by anthropogenic activity. The application of cluster analysis revealed clustering between arsenic, arsenolite, and potassium but no significant geospatial correlation between sample sites in the drainage basin.

Blockchain applied to the construction supply chain:A case study with threat model

The construction industry has long faced the challenge of introducing collaborative systems among multiple stakeholders.This challenge creates a high level of rigidity in terms of processing shared information related to different processes,robust holistic regulations,payment actualizations,and resource utilization across different nodes.The need for a digital platform to crossconnect all stakeholders is necessary.A blockchain-based platform is a prime candidate to improve the industry in general and the construction supply chain(CSC)in particular.In this paper,a literature review is presented to establish the main challenges that CSC faces in terms of its effects on productivity and efficiency.In addition,the effect of applying blockchain platforms on a case study is presented and analyzed from performance and security level.The analysis aims to emphasize that blockchain,as presented in this paper,is a viable solution to the challenges in the CSC regardless of the risks associated with the security and robustness of the flow of information and data protection.Moreover,a threat analysis of applying a blockchain model on the CSC industry is introduced.This model indicates potential attacks and possible countermeasures to prevent the attacks.Future work is needed to expand,quantify,and optimize the threat model and conduct simulations considering proposed countermeasures for the different blockchain attacks outlined in this study.

Last Planner System and Scrum: Comparative analysis and suggestions for adjustments

This study provides a critical review of the concepts of Agile,Lean,Scrum,and Last Planner®System(LPS).A comparative analysis is conducted between LPS and Scrum to expand LPS by considering Scrum’s best practices.Eight dimensions,namely,1)origins,2)main purpose,3)overall system/framework process,4)tools or artifacts maintained by the team,5)team composition and main roles,6)regular events or team meetings,7)metrics/dashboards,and 8)approach to learning,are evaluated.After analyzing side by side the eight dimensions,it was found that many aspects from Scrum already exist in LPS in the same or similar form.However,the authors identify four main elements from Scrum that can be leveraged to improve the LPS benchmark,such as considering the Scrum“Increment”concept into LPS,having a clear definition of roles and responsibilities,or adding an equivalent to a Scrum Master to have a designated"rule keeper”in LPS.These opportunities to be considered in new LPS benchmarks need to be tested and validated with real applications.To the best of the authors5 knowledge,this work is the first to comprehensively compare Scrum(Agile)and LPS(Lean)and could be seen as a contribution toward the evolution of the Last Planner System for the academic and industrial environments.
