Hydrophobic protein in colorectal cancer in relation to tumor stages and grades

AIM: To identify differentially expressed hydrophobic proteins in colorectal cancer. METHODS: Eighteen pairs of colorectal cancerous tissues in addition to tissues from normal mucosa were analysed. Hydrophobic proteins were extracted from the tissues, separated using 2-D gel electrophoresis and analysed using Liquid Chromatography Tandem Mass Spectrometry (LC/MS/MS). Statistical analysis of the proteins was carried out in order to determine the significance of each protein to colorectal cancer (CRC) and also their relation to CRC stages, grades and patients’ gender. RESULTS: Thirteen differentially expressed proteins which were expressed abundantly in either cancerous or normal tissues were identified. A number of these proteins were found to relate strongly with a particular stage or grade of CRC. In addition, the association of these proteins with patient gender also appeared to be ***: Stomatin-like protein 2 was found to be a promising biomarker for CRC, especially in female patients. The differentially expressed proteins identified were associated with CRC and may act as drug target candidates.

A core assumption of sexual selection theory is that sexually selected weapons,specialized morphological structures used directly in male con-tests,can improve an individual's reproductive success but only if the bearer can overcome associated costs,the negative effects on the bear-er's fitness ***,recent studies have shown that producing and wielding exaggerated weapons may not necessarily be ***,some traits can be selected for supporting,or compensating for,the expense of producing and wielding such exaggerated weapons.n the ant-mimicking jumping spider Myrmarachne gisti,exaggerated chelicerae are borne only by adult males and not females,showing sexual dimorphism and steep positive allometry with body ***,we determine the potential benefits of bearing exaggerated chelicerae during male contests and explore the potential for costs in terms of prey-capture efficiency and compensation between chelicera size and neighboring trait *** males with longer chelicerae won most of their male-male contests,we found no significant differences in prey-capture effi-ciency between males and females regardless of whether prey was winged or ***'elongated chelicerae thus do not impede their efficiency at capturing ***,we found that the sizes of all neighboring traits are positively correlated with chelicera size,suggesting that these traits may be under correlational *** together,our findings suggest that *** males armed with the exaggerated chelicerae that function as weapons win more fights at limited cost for performance in prey capture and compensate for neighboring structures.

The SARS-CoV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOC)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOCD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUC¼0.91,95%CI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Nano-hydroxyapatite(nHAP)has been widely used in bone repair as an osteo-inductive and naturally-occurring ***,the optimal applied form of nHAP and the underlying mechanisms involved remain ***,to investigate into these,a range of corresponding models were designed,including three applied forms of nHAP(Free,Coating and 3D)that belong to two states(Free or fixed).The results indicate that when fixed nHAP was applied in the 3D form,optimal osteogenesis was induced in human bone marrow stem cells(hBMSCs)with increased bone volume via integrinα7(ITGA7)-mediated upregulation of the PI3K-AKT signaling pathway,while contrary results were observed with free *** osteogenesis experiments in mice subcutaneous transplantation model further confirmed the different tendencies of ITGA7 expression and osteogenesis of hBMSCs in free and fixed states of *** results revealed that the two states of nHAP play a different regulatory role in cell morphology and osteogenesis through the valve role of ITGA7,providing cues for better application of nanoparticles and a potential new molecular target in bone tissue engineering.

Cytochrome P4502J2(CYP2J2)metabolizes arachidonic acid(AA)to cardioprotective epoxyeicosatrienoic acids(EETs).Dronedarone,an antiarrhythmic drug prescribed for treatment of atrial fibrillation(AF)induces cardiac adverse effects(AEs)with poorly understood *** previously demonstrated that dronedarone inactivates CYP2J2 potently and irreversibly,disrupts AA-EET pathway leading to cardiac mitochondrial toxicity rescuable via EET *** this study,we investigated if mitigation of CYP2J2 inhibition prevents dronedarone-induced cardiac *** first synthesized a deuterated analogue of dronedarone(termed poyendarone)and demonstrated that it neither inactivates CYP2J2,disrupts AA-EETs metabolism nor causes cardiac mitochondrial toxicity in *** patch-clamp experiments demonstrated that pharmacoelectrophysiology of dronedarone is unaffected by ***,we show that dronedarone treatment or CYP2J2 knockdown in spontaneously beating cardiomyocytes indicative of depleted CYP2J2 activity exacerbates beat-to-beat(BTB)variability reflective of proarrhythmic *** contrast,poyendarone treatment yields significantly lower BTB variability compared to dronedarone in cardiomyocytes indicative of preserved CYP2J2 ***,poyendarone and dronedarone display similar antiarrhythmic properties in the canine model of persistent AF,while poyendarone substantially reduces beat-to-beat variability of repolarization duration suggestive of diminished proarrhythmic *** findings prove that deuteration of dronedarone prevents CYP2J2 inactivation and mitigates dronedarone-induced cardiac AEs.

Since December 2019,the novel coronavirus(severe acute respiratory syndrome coronavirus 2(SARS-CoV-2))has spread to many countries around the world,developing into a global pandemic with increasing numbers of deaths reported *** data,although some vaccines have been developed,there are no ideal drugs to treat novel coronavirus pneumonia(coronavirus disease 2019(COVID-19)).By examining the structure of the coronavirus and briefly describing its possible pathogenesis based on recent autopsy reports conducted by various teams worldwide,this review analyzes the possible structural and functional changes of the human body upon infection with *** observed that the most prominent pathological changes in COVID-19 patients are diffuse alveolar damage(DAD)of the lungs and microthrombus formation,resulting in an imbalance of the ventilation/perfusion ratio and respiratory *** direct evidence of viral infection can also be found in other organs and tissues,the viral load is relatively *** conclusion that the injuries of the extrapulmonary organs are directly caused by the virus needs further investigation.

We report herein a class of superatomic AU13 clusters stabilized by different N-heterocyclic carbenes(NHCs).The clusters show diverse metal surface structures,properties and functions as exemplified by:(1)the first anionic AU13 cluster[Au1g(NHC-1)sBr6],which has bulky NHC-1 ligands that lead to a rather open metal surface contributing to its high catalytic activity;(2)the tricationic cluster[Au13(NHC-2)sBr2]+which has bidentate,benzyl-rich NHC-2 ligands that make it ultra-stable and highly-luminescent,suitable for bio-imaging;and(3)by bearing two pyridyl groups on NHC-3,the dicationic cluster[Au:a(NHC-3)gClg]f+exhibits reversible and stable visible absorption and solubility responses to protonation/deprotonation cycles,making it a potential pH sensor(NHC-1=1,3-disopropylbenzimidazolin-ylidene;NHC-2=1,3-bis(1-benzyl-1H-benzimidazol-1-ium-yl)propane;NHC-3=1,3-bis(icolyI)benzimidazolin-ylidene).The study nicely demonstrates the importance of ligands in designing metal nanoclusters with desired functionalities.

Copper-hydrides have been intensively studied for a long time due to their utilization in a variety of technologically important chemical ***,poor stability of the species severely hinders its isolation,storage and operation,which is worse for nano-sized *** report here an unprecedented strategy to access to ultrastable copper-hydride nanoclusters(NCs),namely,using bidentate N-heterocyclic carbenes as stabilizing ligands in addition to *** this work,a simple synthetic protocol was developed to synthesize the first large copper-hydride nanoclusters(NCs)stabilized by N-heterocyclic carbenes(NHCs).The NC,with the formula of Cu3i(RS)25(NHC)3H6(NHC=1,4-bis(1-benzyl-1 H-benzimidazol-1-ium-3-yl)butane,RS=4-fluorothiophenol),was fully characterized by high resolution Fourier transform ion cyclotron resonance mass spectrum,nuclear magnetic resonance,ultra-violet visible spectroscopy,density functional theory(DFT)calculations and single-crystal X-ray ***,the title cluster exhibits unprecedented Cu4 tetrahedron-based vertex-sharing(TBVS)superstructure(fusion of six Cu4 tetrahedra).Moreover,the ultrahigh thermal stability renders the cluster a model system to highlight the power of NHCs(even other carbenes)in controlling geometrical,electronic and surface structure of polyhydrido copper clusters.
