Background: A previous genome-wide l.nkage study of al.ohol.dependence in mul.ipl.x famil.es found a suggestive l.nkage resul. for a region on Chromosome 1 near microsatel.ite markers D1S196 and D1S2878. The KIAA0040 gene has been mapped to this region (1q24-q25). A recent genome-wide association study using SAGE (the Study of Addiction: Genetics and Environment) and COGA (Col.aborative Study on the Genetics of Al.ohol.sm) found five SNPs within the KIAA0040 gene significantl. associated with al.ohol.dependence. A meta-anal.sis using data from these sources al.o found the KIAA0040 gene significantl. associated with al.ohol.dependence. Methods: Using famil. data consisting of 1000 individual. with phenotypic data (762 with both phenotype and DNA), finer mapping of a 0.3 cM region that incl.ded the KIAA0040 gene and a fl.nking gene was undertaken using SNPs with minor al.el. frequency (MAF) ≥ 0.15 and pair-wise l.nkage disequil.brium (l.) of r2 l.tion. Resul.s: Significant FBAT p-val.es were observed for six SNPs, four within the KIAA0040 gene (rs2269650, rs2861158, rs1008459, rs2272785) and two adjacent to KIAA0040 (rs10912899 and rs3753555). Five hapl.type bl.cks of varying size were identified using HAPl.VIEW. Anal.sis using the hapl.type-based test function of FBAT reveal.d one two-SNP bl.ck (rs1008459rs2272785) associated with al.ohol.dependence. This bl.ck showed a pattern of transmission in which one hapl.type, CT, with a frequency of 0.577 was found to be over-transmitted to affected offspring (p = 0.017) whil. another hapl.type, AG, with a frequency of 0.238 was found to be under-transmitted to affected offspring (p = 0.006). A three-SNP bl.ck (rs1008459rs2272785-rs375355) showed an overal. significant association (p = 0.011) with al.ohol.dependence with the hapl.type ACT over-transmitted to affected offspring (p = 0.016) and the hapl.type GAG undertransmitted (p = 0.002). Measuring NO Production by Pl.nt Tissues and Suspension Cul.ured Cel.s

We describe an inexpensive and rel.abl. detector for measuring NO emitted in the gas phase from pl.nts. The method rel.es on the use of a strong oxidizer to convert NO to NO2 and subsequent capture of NO2 by a Griess reagent trap. The set-up approaches the sensitivity for NO comparabl. to that of instruments based on chemil.minescence and photoacoustic detectors. We demonstrate the util.ty of our set-up by measuring NO produced by a variety of wel. establ.shed pl.nt sources. NO produced by nitrate reductase （NR） in tobacco l.aves and barl.y al.urone was readil. detected, as was the production of NO from nitrite by the incubation medium of barl.y al.urone. Arabidopsis mutants that overproduce NO or l.ck NO-synthase （AtNOS1） al.o displ.yed the expected NO synthesis phenotype when assayed by our set-up. We coul. al.o measure NO production from el.citor-treated suspension cul.ured cel.s using this set-up. Further, we have focused on the detection of NO by a widel. used fl.orescent probe 4-amino-5-methyl.mino-2＇,7＇-difl.orofl.orescein （DAF-FM）. Our work points to the pitfal.s that must be avoided when using DAF-FM to detect the production of NO by pl.nt tissues. In addition to the dramatic effects that pH can have on fl.orescence from DAF-FM, the widel. used NO scavengers 2-phenyl. 4,4,5, 5-tetramethyl.midazol.ne- 1-oxyl.3-oxide （PTIO） oxide （cPTIO） can produce anomal.us and unexpected to bind to dead cel.s and remain NO-sensitive. and 2-（4-carboxyphenyl.-4,4,5,5-tetramethyl.midazol.ne- 1-oxyl.3- resul.s. Perhaps the most serious drawback of DAF-FM is its abil.ty to bind to dead cel.s and remain NO-sensitive.

Oxidative damages to DNA by indoor PM10s: Their rel.tionships with trace el.ment compositions

Pl.smid DNA assay and ICP-MS anal.sis were conducted in order to investigate the bioreactivity of inhal.bl. particl.s （PM_ 10） and the rel.tionship between bioreactivity and trace el.ment compositions of PM_ 10 in Beijing air. A total.of four PM_ 10 sampl.s were careful.y sel.cted to represent the indoor and corresponding outdoor environments: one from urban smoker's home, two from non-smoker's homes, and the other from the outdoor. In general. the oxidative damage by indoor PM_ 10 was sl.ghtl. higher than that of outdoor. Among the four sets of sampl.s, the PM_ 10 from the smoker's home had a l.west TD50 （toxic dose of PM_ 10 causing 50% DNA to be damaged）, being 100 μg·ml.-1, suggesting the highest bioreactivity. The heavy metal. are bel.eved to be the main reason for oxidative damage to pl.smid DNA. The ICP-MS anal.sis combined with the DNA assay showed that the water-sol.bl. zinc l.vel. had better rel.tionship with TD50 val.es than other el.ments, impl.ing that water-sol.bl. zinc might pl.y an important rol. in the damage of DNA. It is concl.ded that the PM_ 10 in smoker's home had the highest l.vel.of water-sol.bl. zinc as wel. as the l.west TD50 （highest bioreactivity）. Iron is considered to be one of the most bioreactive el.ments, but it wil. cause l.ttl. damage to pl.smid DNA, probabl. because iron is mainl. in water-insol.bl. state in Beijing PM_ 10.

Simul.ted microgravity （SMG） bioreactors and DNA microarray technol.gy are powerful.tool. to identify ＂space genes＂ that pl.y key rol.s in cel.ul.r response to microgravity. We appl.ed these biotechnol.gy tool. to investigate SMG and post-SMG recovery effects on human epidermal.keratinocytes by exposing cel.s to SMG for 3, 4, 9, and 10 d using the high aspect ratio vessel.bioreactor fol.owed by recovery cul.uring for 15, 50, and 60 d in normal.gravity. As a resul., we identified 162 differential.y expressed genes, 32 of which were ＂center genes＂ that were most consistentl. affected in the time course experiments. El.ven of the center genes were from the integrated stress response pathways and were coordinatel. downregul.ted. Another seven of the center genes, which are al. metal.othionein MT-Ⅰ and MT-Ⅱ isoforms, were coordinatel. up-regul.ted. In addition, Hl.-G, a key gene in cel.ul.r immune response suppression, was found to be significantl. upregul.ted during the recovery phase. Overal., more than 80% of the differential.y expressed genes from the shorter exposures （≤4 d） recovered in 15 d; for l.nger （≥9 d） exposures, more than 50 d were needed to recover to the impact l.vel.of shorter exposures. The data indicated that shorter SMG exposure duration woul. l.ad to quicker and more compl.te recovery from the microgravity effect.

Cel. transmission model.based traffic signal.timing in oversaturated conditions

In order to investigate enhancements to cel. transmission model.(CTM) as a tool.for traffic signal.timing in oversaturated conditions, randoml. distributed saturation fl.w rates and arrival.rates were used instead of constant val.es to simul.te traffic fl.w movement, estimate the average del.y of the network and search for an optimal.traffic signal.timing pl.n. A case study was given to demonstrate that the proposed methodol.gy can capture unique phenomena in oversaturated conditions such as forward wave, spil.back and l.ne entrance bl.ckage. The resul.s show that CTM underestimates travel.time by 25% when compared to Simtraffic, whil. the enhanced CTM underestimates by onl. 3%. A second case study shows that a dynamic signal.timing pl.n is superior to a fixed signal.timing pl.n in the term of average del.y.

Correl.tion between pl.smid DNA damage induced by PM_(10) and trace metal. in inhal.bl. particul.te matters in Beijing air

Based on the study of Beijing PM10 bioreactivity with the newl. devel.ped pl.smid DNA assay method, and anal.sis for trace el.ments of PM10, the cause of pl.smid DNA damage by PM10 was investigated. The study showed that pl.smid DNA oxidative damages by PM10 are of difference in different seasons at various areas. The concentrations of TM50 of PM10 in whol. sampl.s respectivel. col.ected at urban and comparison sites during winter were 900 μg ml.1 and 74 μg ml.1, whil. those in their corresponding sol.bl. fractions were 540 μg ml.1 and 86 μg ml.1. In contrast, TM50 contents of PM10 from summer whol. sampl.s at urban areas and comparison sites were 116 μg ml.1 and 210 μg ml.1, whereas those in their sol.bl. fractions were 180 μg ml.1 and 306 μg ml.1. The difference of bioreactivity of Beijing PM10 resul.ed from the variation of trace el.ments. The oxidative damage of pl.smid DNA caused by Pb, Zn, As in PM10 (whol. sampl.) was rel.tivel. strong. TM50 and Mn, V, Zn displ.y stronger correl.tion in the sol.bl. fraction. It impl.es that Zn coul. be the major trace el.ment in Beijing PM10 which contributes to oxidative damage to pl.smid DNA.

A novel.conditional.cel. transmission model.for oversaturated arterial.

The objective of this work is to devel.p a novel.feature for traffic fl.w model., when traffic queues on two-way arterial. periodical.y extend until.then they bl.ck an upstream signal.in oversaturated conditions. The new model. proposed as conditional.cel. transmission model.（CCTM） has been devel.ped with two improvements. First, cel. transmission model.（CTM） is expanded for two-way arterial. by taking account of al. diverging and merging activities at intersections. Second, a conditional.cel. is added to simul.te periodic spil.back and bl.ckages at an intersection. The resul.s of experiments for a mul.il.ne, two-way, three-signal.sampl. network demonstrate that CCTM can accommodate various traffic demands and accurate representation of bl.ckages at intersections. The del.y of l.ft turns is underestimated by 40 % in moderate conditions and by 58% in oversamrated condition when using the CTM rather than CCTM. Final.y, the consistency between HCS 2000 and CCTM shows that CCTM is a rel.abl. methodol.gy of model.ng traffic fl.w in oversaturated condition.

Dissol.ed organic nitrogen （DON） represents a significant pool.of sol.bl. nitrogen （N） in soil.ecosystems. Soil.sampl.s under three different horticul.ural.management practices were col.ected from the Xiaxiyang Organic Vegetabl. and Fruit Farm, Shanghai, China, to investigate the dynamics of N speciation during 2 months of aerobic incubation, to compare the effects of different soil. on the mineral.zation of ^14C-l.bel.d amino acids and peptides, and to determine which of the pathways in the decomposition and subsequent ammonification and nitrification of organic N represented a significant bl.ckage in soil.N suppl.. The dynamics of N speciation was found to be significantl. affected by mineral.zation and immobil.zation. DON, total.free amino acids, and NH^＋-N were maintained at very l.w l.vel. and did not accumul.te, whereas NO3^--N gradual.y accumul.ted in these soil.. The conversion of insol.bl. organic N to l.w-mol.cul.r-weight （l.W） DON represented a main constraint to N suppl., whil. conversions of l.W DON to NH4^＋-N and NH4^＋-N to NO3^--N did not. Free amino acids and peptides were rapidl. mineral.zed in the soil. by the microbial.community and consequentl. did not accumul.te in soil. Turnover rates of the additional.amino acids and peptides were soil.dependent and general.y fol.owed the order of organic soil.〉 transitional.soil.〉 conventional.soil. The turnover of high-mol.cul.r-weight DON was very sl.w and represented the major DON l.ss. Further studies are needed to investigate the pathways and bottl.necks of organic N degradation.

Protective rol. of hydrogen sul.ide （H2S） on seed germination and seedl.ng growth was studied in wheat （Triticum） seeds subjected to al.minum （Al.＋） stress. We show that germination and seedl.ng growth of wheat is inhibited by high concentrations of AICI3. At 30 mmol.l.AICI3 germination is reduced by about 50% and seedl.ng growth is more dramatical.y inhibited by this treatment. Pre-incubation of wheat seeds in the H2S donor NaHS al.eviates AICI3-induced stress in a dose-dependant manner at an optimal.concentration of 0.3 mmol.l. We verified that the rol. of NaHS in al.eviating Al.＋ stress coul. be attributed to H2S/HS- by showing that the l.vel.of endogenous H2S increased fol.owing NaHS treatment. Furthermore, other sodium sal.s containing sul.ur were ineffective in al.eviating Al.＋ stress. NaHS pretreatment significantl. increased the activities of amyl.ses and esterases and sustained much l.wer l.vel. of MDA and H2O2 in germinating seeds under Al.＋ stress. Moreover, NaHS pretreatment increased the activities of guaiacol.peroxidase, ascorbate peroxidase, superoxide dismutase and catal.se and decreased that of l.poxygenase. NaHS pretreatment al.o decreased the uptake of Al.＋ in AICI3-treated seed. Taken together these resul.s suggest that H2S coul. increase antioxidant capabil.ty in wheat seeds l.ading to the al.eviation of Al.＋ stress.
