The import of nuclear-encoded preproteins is necessary to maintain chloroplast function. The recognition and transfer of most precursor proteins across the chloroplast envelopes are facilitated b. two memb.ane-inserted protein complexes, the translocons of the chloroplast outer and inner envelope （Toc and Tic complexes, respectively）. Several signals have b.en invoked to regulate the import of preproteins. In our study, we were interested in redox-b.sed import regulation mediated b. two signals： regulation b.sed on thiols and on the metab.lic NADP＋/NADPH ratio. We sought to identify the proteins participating in the regulation of these transport pathways and to characterize the preprotein sub.roups whose import is redox-dependent. Our results provide evidence that the formation and reduction of disulfide b.idges in the Toc receptors and Toc translocation channel have a strong influence on import yield of all tested preproteins that depend on the Toc complex for translocation. Furthermore, the metab.lic NADP＋/NADPH ratio influences not only the composition of the Tic complex, b.t also the import efficiency of most, b.t not all, preproteins tested. Thus, several Tic sub.omplexes appear to participate in the translocation of different preprotein sub.roups, and the redox-active compo- nents of these complexes likely play a role in regulating transport.

Work with cereals （b.rley and wheat） and a legume （Medicago truncatula） has estab.ished thioredoxin h （Trx h） as a central regulatory protein of seeds. Trx h acts b. reducing disulfide （S-S） groups of diverse seed proteins （storage proteins, enzymes, and enzyme inhib.tors）, thereb. facilitating germination. Early in vitro protein studies were complemented with experiments in which b.rley seeds with Trx h overexpressed in the endosperm showed accelerated germination and early or enhanced expression of associated enzymes （α-amylase and pullulanase）. The current study extends the transgenic work to wheat. Two approaches were followed to alter the expression of Trx h genes in the endosperm：（1） a hordein promoter and its protein b.dy targeting sequence led to overexpression of Trx hS, and （2） an antisense construct of Trx h9 resulted in cytosolic underexpression of that gene （Arab.dopsis designation）. Underexpression of Trx h9 led to effects opposite to those ob.erved for overexpression Trx h5 in b.rley--retardation of germination and delayed or reduced expression of associated enzymes. Similar enzyme changes were ob.erved in developing seeds. The wheat lines with underexpressed Trx showed delayed preharvest sprouting when grown in the greenhouse or field without a decrease in final yield. Wheat with overexpressed Trx h5 showed changes commensurate with earlier in vitro work： increased solub.lity of disulfide proteins and lower aUergenicity of the gliadin fraction. The results are further evidence that the level of Trx h in cereal endosperm determines fundamental properties as well as potential applications of the seed.

The chemical composition of Pellonula leonensis fish oils from the Congo River (at b.ko city) was carried out. The fatty acids were determined b. gas chromatography. The Sn-2 position of fatty acids on glycerol was carried out b. the ISO 6800 standard. The separation of the compounds (as free fatty acids, monoglycerides, diglycerides, triglycerides, sterols and methyl esters, etc.) was carried out b. HPLC using gel permeation with refractometric detection. The phospholipid composition was b. HPLC with an evaporative light scattering detector. We ob.ained oil contents of 32.10 (±0.46)%. The major fatty acids were Palmitic acid (27.41%) and oleic acid (24.23%). The SFA were of 44.50%. The MUFA represent 32.54% and PUFA 22.60%. Regarding the Sn-2 position on glycerol, 48.0% were b. SFA and 51.3% were b. unsaturated fatty acids. Among them 34.2% of fatty acids were 17.5% oleic acid molecules, 3.3% DHA molecules and 2.1% EPA molecules. Free fatty acids have contents of more than 62%, Diglycerides 16.63% and triglycerides 20.46%. Seven different phospholipids were identified, namely: Phosphatidylglycerol (PG), Phosphatidylethanolamine (PE), Phosphatidylinositol (PI), Phosphatidylcholine (PC), Sphingomyelin (SM), Lyso Phosphatitdylcholine (LPC) and Lyso Phosphatidylethanolamine (LPE). According to the high levels in position 2 of the glycerol of palmitic acid, the consumption of Pellonula leonensis fish could b. moderate.

The reduction of nitrate to nitrite b. the oral microb.ota has b.en proposed to b. important for oral health and results in nitric oxide formation that can improve cardiometab.lic conditions. Studies of b.cterial composition in sub.ingival plaque suggest that nitrate-reducing b.cteria are associated with periodontal health, b.t the impact of periodontitis on nitrate-reducing capacity(NRC)and, therefore, nitric oxide availab.lity has not b.en evaluated. The current study aimed to evaluate how periodontitis affects the NRC of the oral microb.ota. First, 16S rRNA sequencing data from five different countries were analyzed, revealing that nitratereducing b.cteria were significantly lower in sub.ingival plaque of periodontitis patients compared with healthy individuals(P b.ingival plaque, saliva, and plasma samples were ob.ained from 42 periodontitis patients b.fore and after periodontal treatment. The oral NRC was determined in vitro b. incub.ting saliva with 8 mmol/L nitrate(a concentration found in saliva after nitrate-rich vegetab.e intake) and compared with the NRC of 15healthy individuals. Salivary NRC was found to b. diminished in periodontal patients b.fore treatment(P b.t recovered to healthy levels 90 days post-treatment. Additionally, the sub.ingival levels of nitrate-reducing b.cteria increased after treatment and correlated negatively with periodontitis-associated b.cteria(P b.seline saliva and plasma nitrate and nitrite levels was found, indicating that differences in the NRC may only b. revealed after nitrate intake. Our results suggest that an impaired NRC in periodontitis could limit dietary nitrate-derived nitric oxide levels, and the effect on systemic health should b. explored in future studies.
