A novel alternative for pyrogen detection based on a transgenic cell line

Pyrogen,often as a contaminant,is a key indicator affecting the safety of almost all parenteral drugs(including biologicals,chemicals,traditional Chinese medicines and medical devices).It has become a goal to completely replace the in vivo rabbit pyrogen test by using the in vitro pyrogen test based on the promoted‘reduction,replacement and refinement’principle,which has been highly considered by regulatory agencies from different *** used NF-κB,a central signalling molecule mediating inflammatory responses,as a pyrogenic marker and the monocyte line THP-1 transfected with a luciferase reporter gene regulated by NF-κB as an in vitro model to detect pyrogens by measuring the intensity of a fluorescence ***,we show that this test can quantitatively and sensitively detect endotoxin(lipopolysaccharide from different strains)and nonendotoxin(lipoteichoic acid,zymosan,peptidoglycan,lectin and glucan),has good stability in terms of NF-κB activity and cell phenotypes at 39 cell passages and can be applied to detect pyrogens in biologicals(group A&C meningococcal polysaccharide vaccine;basiliximab;rabies vaccine(Vero cells)for human use,freeze-dried;Japanese encephalitis vaccine(Vero cells),inactivated;insulin aspart injection;human albumin;recombinant human erythropoietin injection(CHO Cell)).The within-laboratory reproducibility of the test in three independent laboratories was 85%,80%and 80%and the interlaboratory reproducibility among laboratories was 83.3%,95.6%and 86.7%.The sensitivity(true positive rate)and specificity(true negative rate)of the test were 89.9%and 90.9%,*** summary,the test provides a novel alternative for pyrogen detection.

Autotandem catalysis(ATC)is fascinating since a single catalyst can promote tandem reactions involving two or more mechanisms in a single reactor and is a highly desirable approach to realize multifarious transformations in organic synthetic ***,a palladium-catalyzed asymmetric autotandem hydrogenation ofα,β-unsaturated lactones is disclosed,providing chiral saturated *** this reaction,the C=C bond inα,β-unsaturated lactones was first hydrogenated to give the dihydrocoumarins with high enantioselectivity,followed by hydrogenation of the ester group with the same catalytic ***,after decreasing the reaction temperature,hydrogenation of the single C=C bond proceeded smoothly,delivering the dihydrocoumarin products with high yield and *** experiments and density functional theory calculations were carried out to elucidate the cause of this unusual asymmetric autotandem ***,the homogeneous palladium catalyst was successfully applied in the hydrogenation of esters under neutral conditions.

Background Saline nasal irrigation is an effective therapy for relieving common cold *** study aimed to investigate and explore the efficacy of physiological seawater nasal irrigation(PSNI)on children with mild and asymptomatic infection with *** This randomized controlled trial was conducted in Shanghai,China,and 403 children with mild and asymptomatic infection with Omicron were *** children were allocated into the PSNI group and the control *** primary outcome was the duration of viral shedding(DVS),and the secondary outcome was the change in clinical *** The median age of all participants was 5.59(6.26)years *** DVS was significantly shorter in the PSNI group[2.40(1.13)]than in the control group[3.09(2.14)](P=0.014).The multivariable Cox regression model also showed that patients in the PSNI group had an increased probability of shorter DVS compared with patients in the control group[hazard ratio(HR),1.27;95%confidence interval(CI),1.04–1.55;P=0.017].Subgroup analysis suggested that the DVS of patients without full vaccination was significantly reduced in the PSNI *** proportions of runny nose and stuffy nose were apparently reduced in the first three days in the PSNI group or the control group,but there was no evidence showing that PSNI contributes to the benefit compared with the control *** PSNI can reduce the DVS of patients with mild and asymptomatic infection with SARS-CoV-2 Omicron BA.2 variant.

Tal6b/AvrXa27A,a hidden TALE targeting the susceptibility gene OsSWEET11a and the resistance gene Xa27 in rice

Xanthomonas oryzae ***(Xoo)secretes transcription activator-like effectors(TALEs)to activate rice susceptibility(S)genes,causing bacterial blight(BB),as well as resistance(R)genes,leading to de-fense against *** activation follows a gene-for-gene paradigm that results in an arms race between the TALE of the pathogen and effector-binding elements(EBEs)in the promoters of host *** this study,we characterized a novel TALE,designated Tal6b/AvrXa27A,that activates the rice S gene OsSWEET11a and the rice R gene ***6b/AvrXa27A is a member of the AvrXa27/TalAO class and contains 16 repeat variable diresidues(RVDs);one RVD is altered and one is deleted in Tal6b/AvrXa27A compared with AvrXa27,a known avirulence(avr)effector of ***6b/AvrXa27A can transcriptionally activate the expression of Xa27 and OsSWEET11a via EBEs in their corresponding promoters,leading to effector-triggered immunity and susceptibility,*** 16 RVDs in Tal6b/AvrXa27A have no obvious similarity to the 24 RVDs in the effector PthXo1,but EBETal6b and EBEPthXo1 are overlapped in the OsSWEET11a ***6b/AvrXa27A is prevalent among Asian Xoo isolates,but PthXo1 has only been reported in the Philippine strain *** editing of EBETal6b in the OsSWEET11a pro-moter further confirmed the requirement for OsSWEET11a expression in Tal6b/AvrXa27A-dependent susceptibility to ***,Tal6b/AvrXa27A resulted in higher transcription of Xa27 than of OsSWEET11a,which led to a strong,rapid resistance response that blocked disease *** suggest that Tal6b/AvrXa27A has a dual function:triggering resistance by activating Xa27 gene expression as an avirulence factor and inducing transcription of the S gene OsSWEET11a,resulting in ***,Tal6b/AvrXa27A,but not AvrXa27,can bind to the promoter of *** underlying recognition mechanism for this binding remains unclear but appears to deviate from the currently accepted TALE code.

Cationic engineered nanodiamonds for efficient antibacterial surface with strong wear resistance

The spread of diseases caused by bacterial adhesion and immobilization in public places constitutes a serious threat to public *** of bacteria spread by the construction of an antibacterial surface takes precedence over post-infection ***,we demonstrate an effective antibacterial surface with strong wear resistance by constructing cationic engineered nanodiamonds(C-NDs).The C-NDs with positive surface potentials interact effectively with bacteria through electrostatic interactions,where the C-NDs act on the phospholipid bilayer and lead to bacterial membrane collapse and rupture through hydrogen bonding and residual surface oxygen-containing reactive *** this case,bactericidal rate of 99.99%and bacterial biofilm inhibition rate of more than 80%can be achieved with the C-NDs concentration of 1 mg/*** addition,the C-NDs show outstanding antibacterial stability,retaining over 87%of the antibacterial effect after stimulation by adverse environments of heat,acid,and external ***,an antibacterial surface with high wear resistance obtained by integrating C-NDs with commercial plastics has been *** antibacterial surface with a mass fraction of 1 wt.%C-NDs improved abrasion resistance by 3981 times,with 99%killing of adherent *** work provides an effective strategy for highly efficient antibacterial wear-resistant surface,showing great practical applications in public health environments.
