Silvestrol alleviates glioblastoma progression through ERK pathway modulation and MANBA and NRG-1 expression

Background:Glioblastoma,a notably malignant tumor within the central nervous system,is distinguished by its aggressive behavior.Silvestrol,a robust inhibitor of the RNA helicase eukaryotic initiation factor 4A(eIF4A),has shown significant potential as an anticancer compound.Yet,the impact of silvestrol on glioblastoma,especially its molecular mechanisms,has not been fully elucidated.Methods:This investigation employed a variety of in vitro assays,such as cell counting kit-8(CCK-8),clonogenic,5-ethynyl-2′-deoxyuridine(EDU),wound healing,and flow cytometry,to evaluate cell cycle progression,apoptosis,cell viability,and migration.Western blot analysis was also performed to study the apoptosis and extracellular regulated kinase(ERK)pathways.After the ERK pathway was inhibited,differentially expressed genes(DEGs)in U87 cells were identified,followed by an analysis of target genes using the gene expression profiling interactive analysis(GEPIA)database.Results:Silvestrol significantly suppressed the proliferation,migration,and colony formation of glioma cells.It caused cell cycle arrest and enhanced apoptosis in these cells.Additionally,silvestrol stimulated the ERK pathway,with these effects being reversible by an ERK phosphorylation inhibitor.Transcriptome combined with GEPIA,GSCA,UALCAN,TIMER database screened 4 potential drug targets of silvestrol:chromosome 1 open reading frame 226(C1ORF226),mannosidase beta A(MANBA),IQ motif and Sec7 domain 2(IQSEC2),neuregulin 1(NRG-1).Among them,C1ORF226 was lower risk gene while MANBA,IQSEC2,and NRG-1 were high-risk genes.Furthermore,silvestrol notably reduced MANBA mRNA levels,which could be reversed by inhibiting ERK phosphorylation.Furthermore,silvestrol markedly decreased NRG-1 protein levels,with an additional reduction observed when the ERK pathway was blocked.Conclusion:Silvestrol’s anti-glioma effects are primarily due to the suppression of MANBA expression via the ERK pathway and possibly by hindering the translation of NRG-1 protein,thus reducing its expression.The do

Influence of friction-lubrication characteristics varying with rolling speed on instability of chatter and slip in cold tandem rolling process

In the high-speed cold tandem rolling process of thin plate,chatter or slip instability gives rise to the deterioration of equipment and product quality.Macroscopic instability behavior is closely related to interfacial friction-lubrication con-dition which is generally characterized by the friction coefficient.However,with higher and higher speed requirements,the commonly used model of friction coefficient is no longer applicable and accurate.A novel approach is suggested to calculate the speed-dependent friction coefficient,in which the viscosity-pressure-temperature effects of the lubricant,surface roughness states of work rolls and rolled piece are comprehensively involved and the mixed flm lubrication theory is applied.Subsequently,the influences of friction coefficient on the instability of slip and chatter are investigated for a five-stand cold tandem rolling mill.On one side,the critical friction coefficient for each stand is determined by calculating the corresponding slip factor;on the other hand,the friction coefficient varying with rolling speed is combined with the model of theoretical critical rolling speed presented under constant friction coefficient,so that the speed threshold is determined.Furthermore,the stable and unstable regions corresponding to the rear three stands are individually discussed,and the technical strategies are proposed to suppress both slip and chatter frequently occurring in the actual rolling process.

A Review of Intelligent Configuration and Its Security for Complex Networks

Complex networks are becoming more complex because of the use of many components with diverse technologies. In fact, manual configuration that makes each component interoperable has breed latent danger to system security. There is still no comprehensive review of these studies and prospects for further research. According to the complexity of component configuration and difficulty of security assurance in typical complex networks, this paper systematically reviews the abstract models and formal analysis methods required for intelligent configuration of complex networks, specifically analyzes, and compares the current key technologies such as configuration semantic awareness, automatic generation of security configuration, dynamic deployment, and verification evaluation. These technologies can effectively improve the security of complex networks intelligent configuration and reduce the complexity of operation and maintenance. This paper also summarizes the mainstream construction methods of complex networks configuration and its security test environment and detection index system, which lays a theoretical foundation for the formation of the comprehensive effectiveness verification capability of configuration security. The whole lifecycle management system of configuration security process proposed in this paper provides an important technical reference for reducing the complexity of network operation and maintenance and improving network security.

Influence of different magnetic forces on the effect of colonic anastomosis in rats

BACKGROUND Despite much work having been conducted on magnetic compression anastomo-sis(MCA)in the digestive tract,there are no reports on the influence of magnetic force on the anastomosis.AIM To investigate the effect of different magnetic force magnets on the MCA of the digestive tract.METHODS Two groups of magnets of the same sizes but different magnetic forces were designed and produced.A total of 24 Sprague-Dawley rats were randomly assigned into two groups(powerful magnet group and common magnet group),with 12 rats in each group.Two types of magnets were used to complete the colonic side-to-side anastomosis of the rats.The operation time and magnet discharge time were recorded.The anastomotic specimens were obtained 4 wk after the operation and then the burst pressure and diameter of the anastomosis were measured,and the anastomosis was observed via the naked eye and subjected to histological examination.RESULTS The magnetic forces of the powerful and common magnet groups at zero distance were 8.26 N and 4.10 N,respectively.The colonic side-to-side anastomosis was completed in all 24 rats,and the operation success rate and postoperative survival rate were 100%.No significant difference was noted in the operation time between the two groups.The magnet discharge time of the powerful magnet group was slightly longer than that of the common magnet group,but the difference was not statistically significant(P=0.513).Furthermore,there was no statistical difference in the burst pressure(P=0.266)or diameter of magnetic anastomosis(P=0.095)between the two groups.The gross specimens of the two groups showed good anastomotic healing,and histological observation indicated good mucosal continuity without differences on healing.CONCLUSION In the rat colonic side-to-side MCA model,both the powerful magnet with 8.26 N and the common magnet with 4.10 N showed no significant impact on the anastomosis establishment process or its effect.

Effects of Atmospheric Pressure on Developmental Characteristics and the Stability of Air Entraining Agent for Concrete

In order to ascertain the effects of atmospheric pressure on developmental characteristics and the stability of AEA(air-entraining agent)solution bubbles,AEA solution experiments and AEA solution bubble experiments were,respectively,conducted in Peking(50 m,101.2 kPa)and Lhasa(3,650 m,63.1 kPa).Surface tensions and inflection-point concentrations were tested based on AEA solutions,whilst developmental characteristics,thicknesses and elastic coefficients of liquid films were tested based on air bubbles of AEA solutions.The study involved three types of AEAs,which were TM-O,226A,and 226S.The experimental results show that initial sizes of TM-O,226A,and 226S are,respectively,increased by 43.5%,17.5%,and 3.8%.With the decrease of ambient pressure,the drainage rate and the drainage index of AEA solution bubbles increase.Interference experiments show that the liquid film thicknesses of all tested AEA solution bubbles are in micron scales.When the atmospheric pressure decreases from 101.2 to 63.1 kPa,the liquid film thicknesses of three types of AEA solutions decrease in various degrees;and film elasticities at critical thicknesses increase.Liquid film of 226S solution bubbles is the most stable,presenting as a minimum thickness variation.It should be noted that elastic coefficient of liquid film only represents the level at critical thickness,thus it can not be applied as the only evaluating indicator of bubble stability.For a type of AEA,factors affecting the stability of its bubbles under low atmospheric pressure include initial bubbles size,liquid film thickness,liquid film elasticity,ambient temperature,etc.

A new interacting capillary bundle model on the multiphase flow in micropores of tight rocks

Surfactants are widely used in the fracturing fluid to enhance the imbibition and thus the oil recovery rate. However, current numerical models cannot capture the physics behind capillary imbibition during the wettability alteration by surfactants. Although the interacting capillary bundle(ICB) model shows potential in characterizing imbibition rates in different pores during wettability alteration, the existing ICB models neglect the influence of wettability and viscosity ratio on the imbibition behavior, making it difficult to accurately describe the oil-water imbibition behavior within the porous media. In this work,a new ICB mathematical model is established by introducing pressure balance without assuming the position of the leading front to comprehensively describe the imbibition behavior in a porous medium under different conditions, including gas-liquid spontaneous imbibition and oil-water imbibition.When the pore size distribution of a tight rock is known, this new model can predict the changes of water saturation during the displacement process in the tight rock, and also determine the imbibition rate in pores of different sizes. The water saturation profiles obtained from the new model are validated against the waterflooding simulation results from the CMG, while the imbibition rates calculated by the model are validated against the experimental observations of gas-liquid spontaneous imbibition. The good match above indicates the newly proposed model can show the water saturation profile at a macroscopic scale while capture the underlying physics of the multiphase flow in a porous medium at a microscopic scale. Simulation results obtained from this model indicate that both wettability and viscosity ratio can affect the sequence of fluid imbibition into pores of different sizes during the multiphase flow, where less-viscous wetting fluid is preferentially imbibed into larger pores while more-viscous wetting fluid tends to be imbibed into smaller pores. Nervonic acid(NA)is a very-long-chain monounsaturated fatty acid with pharmaceutical and nutraceutical functions that plays an important role in treating several neurological disorders.One major source of NA is plant seed oil.Here we report fatty acid profiles of seeds and germplasm diversity of six plant species,including three woody plants with high amounts of NA-enriched seed oil,Malania oleifera,Macaranga adenantha,and M.indica.M.oleifera had the largest seed(average 7.40 g single seed),highest oil content(58.71%),and highest NA level(42.22%).The germplasm diversity of M.oleifera is associated with its habitat but not elevation.Seeds of M.adenantha contained higher NA levels(28.41%)than M.indica(21.77%),but M.indica contained a significantly higher oil content(29.22%)and seed yield.M.adenantha germplasm varied among populations,with one population having seeds with high oil content(22.63%)and NA level(37.78%).Although M.indica grow naturally at a range of elevations,no significant differences were detected between M.indica populations.These results suggest that M.indica and M.oleifera have greater potential as a source of NA,which will contribute to constructing a germplasm resource nursery and establishing a selection and breeding program to improve the development of NA-enriched plants.

Precast system and assembly connection of cement concrete slabs for road pavement: A review

Prefabricated pavement is increasingly applied worldwide due to the rapid construction on-site.This paper presents a state-of-the-art review of the precast systems and assembly connection of concrete slabs,as well as the precast pavement features.The previous research indicates that:(1)both super-slab and Michigan systems are recommended with the satisfied road performance close to the cast-in-place;(2)flexible base material is suggested for the fabricated pavement for the satisfied leveling and stress distribution;(3)to prevent the voiding phenomenon of the fabricated pavement,the sand cushion course,dry mixed mortar or self-leveling mortar,and other flowable materials could be used for the secondary leveling of the base after the pavement splicing;(4)two-direction dowel bars are recommended for slab connections of the fabricated pavement,which helps to improve the load transfer capacity of the joints and enhance the durability of the fabricated pavement structure;(5)the sealing treatment of precast slab joints needs strengthening to reduce the impact of surface runoff on the base course;(6)the further research focuses are designing with functional,composite,mechanized,intelligent,lightweight,and flexible pavement slabs.Besides,pavement mechanical properties induced by temperature overlapping traffic loads need to be revealed.

在生物燃料制备工艺中,甘油是重要的副产物之一,具有产量大、价格低、同时热值尚可的特点,精炼过后的产品可作为工业中的补充燃料。本文设计了一个可控的旋流喷雾燃烧平台,通过分析不同雾化空燃比(ALR)下精甘油的实际火焰、OH*荧光信号以及燃烧室流场信息,以探究ALR变化对于精甘油喷雾燃烧的影响。此外,对精甘油喷雾燃烧后烟气中的NO、CO和CO_(2)含量随ALR变化的趋势进行分析。结果表明,随着ALR的升高,旋流喷雾火焰的回流区域扩大,甘油火焰中心的OH*信号强度和范围增加,甘油的雾化质量提高。排放方面,NO浓度大体随ALR增加而提升,随当量比增加先升高后下降;CO则是随ALR升高而降低,随当量比增加先降低,后有一定程度升高。提升ALR有利于降低喷雾液滴粒径,促进液滴蒸发及混合,在一定工况范围内能实现低NO_(x)和CO的甘油高效稳定燃烧。

Bioinspired polarized skylight navigation,which can be used in unfamiliar territories,is an important alternative autonomous navigation technique in the absence of Global Navigation Satellite System(GNSS).However,the polarization pattern in night environment with noise effects and model uncertainties is a less explored area.Although several decades have passed since the first publication about the polarization of the moonlit night sky,the usefulness of nocturnal polarization navigation is only sporadic in previous researches.This study demonstrates that the nocturnal polarized light is capable of providing accurate and stable navigation information in dim light outdoor environment.Based on the statistical characteristics of Angle of Polarization(Ao P)error,a probability density estimation method is proposed for heading determination.To illustrate the application potentials,the simulation and outdoor experiments are performed.Resultingly,the proposed method robustly models the distribution of Ao P error and gives accurate heading estimation evaluated by Standard Deviation(STD)which is 0.32°in a clear night sky and 0.47°in a cloudy night sky.
