Mechanisms of phototherapy of Alzheimer’s disease during sleep and wakefulness:the role of the meningeal lymphatics

With the increase in the aging population,the global number of people with Alzheimer’s disease(AD)progressively increased *** situation is aggravated by the fact that there is no the efective pharmacological therapy of ***(PBM)is non-pharmacological approach that has shown very promising results in the therapy of AD in pilot clinical and animal ***,the mechanisms of therapeutic efects of PBM for AD are poorly *** this study on mice,we demonstrate that photodynamic efects of 5-aminolevulenic acid and laser 635 nm cause reduction of network of the meningeal lymphatic vessels(MLVs)leading to suppression of lymphatic removal of beta-amyloid(Aβ)from the right lateral ventricle and the *** the original protocol of PBM under electroencephalographic monitoring of wakefulness and sleep stages in non-anesthetized mice,we discover that the 7-day course of PBM during deep sleep *** provides better restoration of clearance of Aβfrom the ventricular system of the brain and the *** results shed light on the mechanism of PBM and show the stimulating efects of PBM on the brain lymphatic drainage that promotes transport of Aβvia the lymphatic *** efects of PBM on the brain lymphatics in sleeping brain open a new niche in the study of restorative functions of sleep as well as it is an important informative platform for the development of innovative smart sleep technologies for the therapy of AD.

Fibroblast growth factor21(FGF21)is a hormone that can balance nutrient fluctuations,control metabolic processes,and maintain energy *** FGF21 is produced by a variety of cell types,including hepatocytes,adipocytes,bone cells,myocardial cells,and pancreas cells,and act on various effector tissues such as brain,adipose tissue,liver,heart,and skeletal muscle.1 The target that FGF21 interacts with is the cell-surface receptor composed of FGF receptors in complex with the single-pass transmembrane proteinβ-*** receptors are abundantly expressed,both in peripheral tissues and some regions of the CNS,such as the hypothalamus and amygdala and the locus coeruleus(LC).2 Physiologically,FGF21 can be induced by various metabolic stresses,including hunger,protein deficiency,monosaccharides,and alcohol,by acting onβ-Klotho receptor in corresponding tissues,ultimately exerting regulatory effects(Fig.1)

Nanometallocarbosilanes： Synthesis, Physicochemical Properties and Structure

Highly efficient synthesis methods have been developed and characteristics of nanometallocarbosilanes molecular structure were studied by the research team of GNIIChTEOS （State Research Institute for Chemistry and Technology of Organoelement Compounds）. Nanometallocarbosilanes were synthesized by thermal co-condensation of oligocarbosilanes and alkyl amides of refractory metals. Initial, intermediate and final products of side reactions were characterized by 1H, 13C, 29Si NMR （nuclear magnetic resonance）, IR （infra-red） spectroscopy, GPC （gel-penetrating chromatography）, TGA （thermal gravimetric analysis）, TEM （transmission electron microscopy）, SEM （scanning electron microscopy）, RES （X-ray phase analysis） and elemental analysis. The proposed synthesis method of nanometallocarbosilanes was lbund to produce fusable soluble organosilicon oligomers with homogeneous distribution of nanoscale （10-20 nm） metal particles in the oligomer matrix. A computational model of the group and elemental composition of nanometallocarbosilanes was developed; it was shown that they are molecular globules of near-spherical shape and rigid polycyclic structure. Thermochemical treatment of nanometallocarbosilanes leads to SiC-nanoceramics （a high yield of up to 75-80 mass%） modified by metal nanoparticles （20-30 nm） contributing to its stabilization. The application of preceramic oxygen-free nanometallocarbosilanes will make it possible to advance in solving the problem of ceramic composite materials with long-term resistance at temperatures above 1,500 ℃ in oxidizing environments.
