Racotumomab monoclonal antibody is a murine anti-idiotypic antibody. This monoclonal antibody mimics N-glycolyl-GM3 gangliosides has been tested in several clinical trials Phase I/II for breast, melanoma and non-small cell lung cancer patients as an anti-idiotypic cancer vaccine. The early production process was performed in vivo from mice ascites fluid. This process was transferred to bioreactor-based method at pilot scale followed to the scale-up of the fermentation. In this work we present a comprehensive molecular characterization of racotumomab MAb produced by the two different production scales in order to determine the impact of the manufacturing process in vaccine performance. We observed differences in glycosylation pattern and charge heterogeneity between racotumomab produced in both scales. Interestingly, these modifications had no significant impact on biological activity elicited in chickens. So, changes in primary structure like glycosylation, charge heterogeneity and oxidation did not affect biological activity of the vaccine.

Characterizing the diversity and structure of host–parasite communities is crucial to understandingtheir eco-evolutionary dynamics.Malaria and related haemosporidian parasites are responsible forfitness loss and mortality in bird species worldwide.However,despite exhibiting the greatest ornithologicalbiodiversity,avian haemosporidians from Neotropical regions are quite unexplored.Here,we analyze the genetic diversity of bird haemosporidian parasites(Plasmodium andHaemoproteus)in 1,336 individuals belonging to 206 bird species to explore for differences in diversityof parasite lineages and bird species across 5 well-differentiated Peruvian ecoregions.Wedetected 70 different haemosporidian lineages infecting 74 bird species.We showed that 25 out ofthe 70 haplotypes had not been previously recorded.Moreover,we also identified 81 new host–parasite interactions representing new host records for these haemosporidian parasites.Our outcomesrevealed that the effective diversity(as well as the richness,abundance,and Shannon–Weaver index)for both birds and parasite lineages was higher in Amazon basin ecoregions.Furthermore,we also showed that ecoregions with greater diversity of bird species also had highparasite richness,hence suggesting that host community is crucial in explaining parasite richness.Generalist parasites were found in ecoregions with lower bird diversity,implying that the abundanceand richness of hosts may shape the exploitation strategy followed by haemosporidian parasites.These outcomes reveal that Neotropical region is a major reservoir of unidentified haemosporidianlineages.Further studies analyzing host distribution and specificity of these parasites inthe tropics will provide important knowledge about phylogenetic relationships,phylogeography,and patterns of evolution and distribution of haemosporidian parasites.

The aim of this study was to evaluate injectable,in situ cross-linkable elastin-like recombinamers(ELRs)for osteochondral repair.Both the ELR-based hydrogel alone and the ELR-based hydrogel embedded with rabbit mesenchymal stromal cells(rMSCs)were tested for the regeneration of critical subchondral defects in 10 New Zealand rabbits.Thus,cylindrical osteochondral defects were filled with an aqueous solution of ELRs and the animals sacrificed at 4months for histological and gross evaluation of features of biomaterial performance,including integration,cellular infiltration,surrounding matrix quality and the new matrix in the defects.Although both approaches helped cartilage regeneration,the results suggest that the specific composition of the rMSC-containing hydrogel permitted adequate bone regeneration,whereas the ELR-based hydrogel alone led to an excellent regeneration of hyaline cartilage.In conclusion,the ELR cross-linker solution can be easily delivered and forms a stable well-integrated hydrogel that supports infiltration and de novo matrix synthesis.

Purpose: To examine the association of physical activity(PA) intensity levels and sedentary time with health-related quality of life(HRQoL) in women with fibromyalgia and whether patients meeting the current PA guidelines present better HRQoL.Methods: This cross-sectional study included 407 women with fibromyalgia aged 51.4 ± 7.6 years. The time spent(min/day) in different PA intensity levels(light, moderate, and moderate-to-vigorous physical activity(MVPA) and sedentary time were measured with triaxial accelerometry. The proportion of women meeting the American PA recommendations(>150 min/week of MVPA in bouts >10 min) was also calculated.HRQoL domains(physical function, physical role, bodily pain, general health, vitality, social functioning, emotional role, and mental health), as well as physical and mental components, were assessed using the 36-item Short-Form Health Survey.Results: All PA intensity levels were positively correlated with different HRQoL dimensions(r_(partial) between 0.10 and 0.23, all p < 0.05). MVPA was independently associated with social functioning(p < 0.05). Sedentary time was independently associated with physical function, physical role, bodily pain, vitality, social functioning, and both the physical and mental component summary score(all p < 0.05). Patients meeting the PA recommendations presented better scores for bodily pain(mean = 24.2(95%CI: 21.3-27.2) vs. mean = 20.4(95%CI: 18.9-21.9), p = 0.023)and better scores for social functioning(mean = 48.7(95%CI: 43.9-44.8) vs. mean = 42.3(95%CI: 39.8-44.8), p = 0.024).Conclusion: MVPA(positively) and sedentary time(negatively) are independently associated with HRQoL in women with fibromyalgia. Meeting the current PA recommendations is significantly associated with better scores for bodily pain and social functioning. These results highlight the importance of being physically active and avoiding sedentary behaviors in this population.

Real-time interventional MRI（I-MRI）would be very helpful to track the position of the surgical device during MR-guided neurosurgery.This would improve the patient outcome.Specially,in deep brain stimulation（DBS）,the visualization of the surgical procedure in real-time using I-MRI could improve the accuracy of the electrode placement.However,the reconstruction of interventional images using high undersampling rate and fast reconstruction speed for real-time imaging pose a great challenge.We proposed a feature-based convolutional neural network（FbCNN）for reconstructing for reconstructing interventional images from golden-angle radially sampled data,based on recent advances in deep learning（DL）.The reconstruction is composed of two steps: 1)reconstruction of the interventional feature;2)feature refinement and postprocessing.We showed that the interventional feature can be reconstructed with a cascade CNN,using an undersampling of only 5 radially sampled spokes.We obtained a final interventional image from a refined feature and a fully sampled reference image.FbCNN was compared with traditional reconstruction techniques and recent DL-based method,and it was shown that only FbCNN could reconstruct the interventional feature and the final interventional image.Furthermore,FbCNN demonstrated that had the potential in real-time I-MRI applications with a reconstruction time of ～500ms per frame and an acceleration factor of ～80.

BACKGROUND： Balance problems are common in people of all ages and can lead to falls, thus causing fractures with consequent disability. Qigong practice has long been part of daily life in Chinese culture, and has good effects on physical health maintenance. OBJECTIVE： The present work describes the change in balance in young, healthy women after practising Qigong for eight weeks. DESIGN, SETTING, PARTICIPANTS AND INTERVENTIONS： The study took the form of a controlled, randomised longitudinal trial, and involved 30 women aged 18-25 years. The subjects had no prior experience of Qigong or Tai Chi and were unaware of the aims of the study. Subjects were randomly assigned to a Qigong intervention group or a control group. Those in the Qigong intervention group performed ＂exercises in 20 figures for health and long-life＂（Wang Ziping） for 1 h twice per week, for 4 weeks. The control group undertook no exercise at all. MAIN OUTCOME MEASURES： The main outcome measure was the stabilometry values. These were obtained in a unipodal support test, using a plantar pressure platform with optical sensors. RESULTS： The Qigong subjects showed a significant improvement in their stabilometry results （40.1% pre-intervention and 56.4% post-intervention）（P〈0.045）, while no improvement was seen in the control group （51.2% pre-intervention and 53.5% post-intervention）. At the beginning of the intervention, the stabilometry values recorded for the Qigong intervention group were worse than those recorded for the control group （40.15% and 51.21% respectively; P=0.121）. However, a comparison of the post-intervention values between these groups showed that these differences have disappeared （P=0.653）. CONCLUSION： Qigong can improve balance in healthy, young women.

Background:Lung hypoplasia,pulmonary persistent hypertension of the newborn and its morphological changes are the main features in congenital diaphragmatic hernia(CDH).This study was undertaken to investigate if antenatal use of sildenafil and/or bosentan attenuates vascular remodeling,promotes branching,and improves alveolarization in experimental nitrofeninduced CDH.Methods:Nitrofen(100 mg)was gavage-fed to pregnant rats at post conception day(PCD)9 to induce CDH.The rats were randomized to 5 groups:1)control;2)nitrofen;3)nitrofen+sildenafil 100 mg/kg per day at PCD 16-20;4)nitrofen+bosentan 30 mg/kg per day,at PCD 16-20,and 5)nitrofen+bosentan+sildenafil,same doses and administration days.After cesarean delivery,the offsprings were sacrifi ced.The diaphragmatic defect and pulmonary hypoplasia were identified,and the lungs were dissected.Arterial wall thickness,bronchiolar density and alveolarization were assessed.Results:The offsprings with CDH were characterized by severe pulmonary hypoplasia(lung weight-to-body weight ratio:0.0263[95%confidence interval(CI)0.0242-0.0278)]in the nitrofen group versus 0.0385(95%CI 0.0355-0.0424)in the control group(P=0.0001).Pulmonary arterial wall thickness was decreased to 3.0(95%CI 2.8-3.7)μm in the nitrofen+sildenafil group versus 5.0(95%CI 4.1-4.9)μm in the nitrofen group(P=0.02).Terminal bronchioles increased to 13.7(95%CI 10.7-15.2)μm in the nitrofen+bosentan group in contrast to 8.7(95%CI 7.2-9.4)μm in the nitrofen group(P=0.002).More significant differences(P=0.0001)were seen in terminal bronchioles in the nitrofen+sildenafil+bosentan group than in the nitrofen group[14.0(95%CI 12.5-15.4)μm versus 8.5(95%CI 7.1-9.3)μm].Pulmonary arterial wall thickness was also decreased in the former group.Conclusions:In this rat model,antenatal treatment with sildenafil attenuates vascular remodeling.Bosentan promotes the development of terminal bronchioles in nitrofen-induced CDH.

Upper eyelid blepharoplasty is one of the most frequently performed surgeries by plastic surgeons worldwide. A prospective observational study was conducted to describe the anthropometric measurements of the upper eyelid and eyebrow in a Mexican population. The researcher measured the anthropometry of the eyelids and eyebrows to establish the diagnosis and surgical plan according to the checklist created and applied by the plastic and reconstructive surgery service. In anatomical position with a caliper instrument, the marginal reflex distance (MRD1), the tarsal thickness, the length of the vertical and horizontal slit, and finally the distance at which the external canthus is located with respect to the internal canthus were measured in millimeters. For the anthropometry of the eyebrows, the distance from the eyebrows to the hairline, the position in millimeters of the orbital rim with respect to the eyebrow, and the angle of its apex were measured. A sample of 210 patients was obtained, of which 65 were male and 145 females, divided by age groups. It was observed that in the three age groups, females presented a vertical palpebral fissure with a higher mean than males. Similar is the case of tarsal thickness, in which the difference was significantly greater in the female of the three age groups. On the other hand, it was observed that the horizontal slit in males was higher than that of females, while the MDR1 was similar in both sexes. Finally, it was identified that in the group of 41 to 50 years old significant decreases are observed both in the vertical cleft, tarsal thickness and the position of the external canthus compared to the group of 20 to 30 years old, this is due to the aging process. When evaluating the anthropometrics of the eyebrows, a greater distance from the trichion to the eyebrow was observed in males than in females, in contrast to the position of the eyebrow with respect to the supraorbital rim and its apex, in which it was shown that in females there is a greater distance. Background:Recently,sildenafil was introduced to treat pulmonary arterial hypertension (PAH);however,there are currently few studies on the pharmacokinetics of sildenalfil in children.Therefore,we aimed to carry out a pharmacokinetic study of sildenafil in children with PAH using a single dose.Methods:Twelve children diagnosed with PAH,consisting of with ten males and two females,were recruited for the study after obtaining written consent from their parents or guardians.Blood samples were obtained predose and at 0.25,0.5,1,2,4,8 and 12 hours after the oral administration of 1 mg/kg of sildenafil using an extemporal pediatric formulation developed in our laboratory.The samples were analyzed using a previously validated high performance liquid chromatography method.Results:A pharmacokinetic analysis using the WinNonlin 3.1 program that considered the Akaike information criterion (AIC) for sdecting a more adjustable model was performed.The following pharmacokinetic parameters were obtained:maximal concentration (Cmax):366+179 ng/mL,time to maximal concentration:0.92±0.30 hours,elimination half-life (t1/2):2.41±1.18 hours,total clearance (CLto/F):5.85±2.81 L/hour,volume of distribution (Vd/F):20.13±14.5 L,absorption rate constants (Ka):0.343 hour-1,elimination rate (Ke):0.35 hour-1,area under curve from zero to infinity:2061±618 ng/mL/hour.The data of all patients adjusted to the model of one compartment were corroborated using AIC.Conclusions:The parameters Ka,Ke and t1/2 were found to be similar to those reported in adults;however,the values of Cmax and Vd/F were significantly higher.Based on these findings,we propose that treatment regimen of sildenafil be adjusted in children with PAH.
