r.talin Use Modifies Alcohol Effects in r.ts

Methylphenidate (MPD), known as r.talin, is a common dr.g pr.scr.bed for.those diagnosed with Attention Deficit Hyper.ctivity Disor.er.(ADHD).Ther. ar. r.por.s that many MPD user. consume alcohol, r.sulting in toxic effects and hospitalization. The goal of this study was to investigate the effects of ethanol in r.ts concomitant with acute and r.petitive MPD exposur.. r.ts wer. divided into four.gr.ups, contr.l (saline), 0.6 mg/kg MPD, 2.5 mg/kg MPD, and 10.0 mg/kg MPD gr.ups and lasted for.12 consecutive days. Ethanol was given after.r.peated MPD administr.tion as follows. On exper.mental day 1 (ED 1), all animals wer. tr.ated with saline to establish baseline, on ED 2 thr.ugh ED 7 either.saline or.MPD (0.6, 2.5, or.10.0 mg/kg) was given. On ED 11, after.thr.e days without tr.atment (ED 8 - 10), r.ts wer. tr.ated as they wer. on ED 2 - 7. At ED 12, 1 g/kg ethanol was administer.d, and one hour.of locomotor.activity was r.cor.ed after.alcohol administr.tion, using the open field assay. The data show a dose r.sponse char.cter.stic of incr.ased locomotor.activity with incr.asing doses of MPD. Ethanol administr.tion alone depr.sses locomotor.activity. The depr.ssive effect of alcohol was significantly attenuated in animals tr.ated with MPD, in a dose dependent manner. The higher.dose of MPD pr.viously administer.d r.sulted in a lar.er.attenuation of the ethanol’s suppr.ssive effect. These tr.nds demonstr.te that chr.nic MPD exposur. dir.ctly influences the effects of alcohol in r.ts. Under.these cir.umstances, it is r.asonable to assume that a subject will need to consume an incr.ased amount of ethanol in or.er.to attain the ethanol effect desir.d. This discr.pancy between effects and exposur. may be a liability for.ethanol toxicity.
