Threonine 32 （Thr32） of FoxO3 is critical for TGF-β-induced apoptosis via Bim in hepatocarcinoma cells

Transforming growth factor-β （TGF-β） exerts apoptotic effects on various types of malignant ceils, including liver cancer cells. However, the precise mechanisms by which TGF-β induces apoptosis remain poorly known. In the present study, we have showed that threonine 32 （Thr32） residue of FoxO3 is critical for TGF-β to induce apoptosis via Bim in hepatocarcinoma Hep3B cells. Our data demonstrated that TGF-βinduced FoxO3 activation through specific de-phosphorylation at Thr32. TGF-β-activated FoxO3 cooperated with Smad2/ 3 to mediate Bim up-regulation and apoptosis. FoxO3 （de）phosphorylation at Thr32 was regulated by casein kinase I-ε （CKI-E）. CKI inhibition by small molecule D4476 could abrogate TGF-β-induced FoxO/Smad acti- vation, reverse Bim up-regulation, and block the sequential apoptosis. More importantly, the deregulated levels of CKI-ε and p32FoxO3 were found in human malignant liver tissues. Taken together, our findings suggest that there might be a CKI-FoxO/Smad-Bim engine in which Thr32 of FoxO3 is pivotal for TGF-β- induced apoptosis, making it a potential therapeutic target for liver cancer treatment.

MRI and diffusion tensor imaging in assessing correlation of activation of cortical motor function and manifestations of corticospinal tract with muscle strength in patients with ischemic stroke

BACKGROUND： Ischemic stroke is often followed by the abnormalities of neurons and corticospinal tract, which can lead to corresponding clinical symptoms and signs. Recently, with the continuous perfection of high-field MRI instrument, it becomes possible to assess and investigate the cortical function and structural reconstruction following stroke by using MRI and diffusion tensor imaging （DTI）. OBJECTIVE： To observe the cortical motor function and changes of corticospinal tracts by using MRI and DTI in the patients with ischemic stroke at acute period, compare with the normal subjects, and assess the damage of corticospinal tract and muscle strength.DESIGN ： A case-control observation.SETTING： Department of Medical Imaging, Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA. PARTICIPANTS ： Nine inpatients （5 males and 4 females） with injury of motor function induced by acute ischemic stroke were selected from Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA between August and December in 2005, they aged 16-87 years with an average of 51 years old, and those with obvious conscious disturbances and severe cognitive disorders were excluded. At the same time, nine healthy right-handed physical examinees matched by age and gender with the patients were also selected, and they all had no nervous disease, epilepsy, mental diseases, cerebrovascular abnormalities and injury history, etc. All the subjects were informed with the detected items and agreed to participate in. METHODS： All the 9 patients with ischemic stroke at acute period and 9 healthy subjects were examined with MRI and DTI.① A block-based design was used in the MRI, the passive finger-to-finger exercise was used as the stimulative task, and the static condition was taken as the baseline task. Prediction of Wordle Scores Based on ARIMA and LSTM Models

This paper examines the effectiveness of the Differential autoregressive integrated moving average (ARIMA) model in comparison to the Long Short Term Memory (LSTM) neural network model for predicting Wordle user-reported scores. The ARIMA and LSTM models were trained using Wordle data from Twitter between 7th January 2022 and 31st December 2022. User-reported scores were predicted using evaluation metrics such as MSE, RMSE, R2, and MAE. Various regression models, including XG-Boost and Random Forest, were used to conduct comparison experiments. The MSE, RMSE, R2, and MAE values for the ARIMA(0,1,1) and LSTM models are 0.000, 0.010, 0.998, and 0.006, and 0.000, 0.024, 0.987, and 0.013, respectively. The results indicate that the ARIMA model is more suitable for predicting Wordle user scores than the LSTM model.

微网内分布式资源分属于不同主体,市场主体的趋利性市场行为影响微网的源荷功率特性,进而影响电力设备和配电网的可靠性。文中提出了一种考虑微网日前市场交易影响的配电网可靠性分析模型。首先,构建各类市场参与者的日前市场投标策略和市场出清模型。其次,考虑不同时段源荷水平对可靠性的影响,提出载荷相关的元件老化失效模型,并针对源、荷、投标及外部电网电价的随机性及时序性,生成'源—荷—市'联合时序场景。最后,在改进的RBTS BUS 6算例系统上进行仿真分析。结果表明,需求侧分布式资源参与市场能降低峰值时段的元件失效概率且增加系统容量备用,起到一定的削峰填谷作用,根据不同电源的出力水平合理地配置备用容量能有效提高供电可靠性。

An XPS Study of Nitrogen Structures in Soil Humic Substances

X-ray photoelectron spectroscopy (XPS) was applied to examine the N structures of soil humic substances and some of their analogues. It was found that for soil humic substances XPS method gave similar results as those obtained by 15N CPMAS NMR (cross-polarization magic-angle spinning nuclear magnetic resonance) method. 70%～86% of total N in soil humic substances was in the form of amide, and 6%～13% was presented as amines, with the remaining part as heterocyclic N. There was no difference in the distribution of the forms of N between the humic substances from soils formed over hundreds or thousands of years and the newly formed ones. For fulvic acid from weathered coal and benzoqu inone- (N H-4 )-2 S O-4 polymer the XPS results deviated significantly from the 15N CPMAS NMR data.

Systemic chemotherapy for metastatic breast cancer

Breast cancer is the leading cause of cancer among women worldwide and the most common cancer in China. Many factors influence the treatment strategy for metastatic breast cancer （MBC）. Chemotherapy should be administered to patients with hormone receptor-negative tumors, symptomatic visceral metasta- sis, and a short disease-free interval. Sequential single-agent chemotherapy has similar efficacy as combi- nation agents in terms of overall survival and quality of life. Anthracyclines are the cornerstone of first-line treatment for MBC, and taxanes represent the second treatment option after resistance. When progression or intolerable toxicity occurs after optimal treatment, the alternative treatments include capecitabine, vinorel- bine, and gemcitabine. Ixabepilone and eribulin are relatively new effective single agents. A combination of cytotoxic agents for patients with rapid clinical progression can further improve the overall response rate and time to progression compared to single-agent treatment. For patients with MBC who were pretreated with anthracyclines in the neoadjuvant/adjuvant setting, a taxane-containing regimen such as docetaxel plus capecitabine or gemcitabine plus paclitaxel should be administered. Platinum-based therapies such as cisplatin or carboplatin have a role in the treatment of triple-negative breast cancer. Meanwhile, the efficacy of the addition of targeted drugs such as iniparib, bevacizumab, and catuximab to chemotherapy remains unproven. Maintenance chemotherapy is routinely recommended in clinical practice at present. Patients who were previously treated with paclitaxel and gemcitabine have better progression-free and overall sur- vival with maintenance chemotherapy according to a Korean phase III clinical trial. Sequential maintenance treatment with capecitabine monotherapy after capecitabine-based combination chemotherapy （X-based X） appears favorable based on a series of domestic studies.

Psychosis has increasingly become a social problem,emphasizing the need to understand the relationship between mental disorders and *** study aimed to investigate the relationship between mental disorders and personality among psychiatric outpatients based on real-world *** Checklist 90(SCL-90)and Eysenck Personality Questionnaire(EPQ)were used to evaluate the personality and psychopathological symptoms of patients(n=8409)in the Psychiatric Outpatient Department at Nanjing Drum Tower Hospital.t-test was used to compare scores between patients and national ***’s correlation coefﬁcient and path analysis were used to explore the relationship between mental health status and *** correlation coefﬁcient between the neuroticism(N)score and each factor score of the SCL-90 test,as well as the correlation between psychoticism(P)and hostility and paranoia,exceeded *** analysis revealed that the standardized path coefﬁcients of N score and SCL-90 were all higher than *** addition,the standardized path coefﬁcient of hostility and paranoia on P score were 0.313 and 0.280,*** sensitivity,depression and obsessive-compulsive symptoms were affected by extraversion(E)score,with standardized path coefﬁcients of-0.149,-0.138,and-0.105,*** path analysis also showed the direct and indirect effects of age,gender,education,and marital status on *** characterized as melancholic had higher scores in all factors of *** conclusion,mental health was related to personality traits of neuroticism,psychoticism and introversion.

Inflammatory bowel diseases(IBD)are the incurable chronic recurrent gastrointestinal disorders and currently lack in safe and effective *** this study,patchouli alcohol,a main active compound of traditional Chinese herb patchouli,was developed into biomimetic liposomes for macrophagetargeting delivery for IBD *** developed lactoferrin-modified liposomes(LF-lipo)can specifically bind to LRP-1 expressed on the activated colonic macrophages and achieve cell-targeting anti-inflammatory ***-lipo reduced the levels of inflammatory cytokines and ROS and suppressed the MAPK/NF-κB ***-lipo also suppressed the formation of NLRP3 inflammasome and the consequent IL-1β***-lipo showed improved therapeutic efficacy in a DSS-induced colitis murine model,evidenced by the reduced disease activity index,the improved colon functions,and the downregulated inflammatory cytokines in the ***-lipo provided an effective and safe macrophagetargeting delivery and therapeutic strategy for addressing the unmet medical need in IBD management.
