Protective effects of Wuwei Xiaodu Drink against chronic osteomyelitis through Foxp3^(+)CD25^(+)CD4^(+) Treg cells via the IL-2/STAT5 signaling pathway

To explore the effectiveness and safety of a Chinese medicinal decoction Wuwei Xiaodu Drink(WWXDD)in inhibiting chronic osteomyelitis via regulatory T cells signaling.The effective constitutes of WWXDD and osteomyelitis related genes were screened.Target proteins were cross-validated using the Venny database.GO function and KEGG pathway analysis were performed for target proteins,while pharmacological network was constructed.The bone properties were analyzed by HE staining and the concentrations of immune factors were measured by ELISA.The expression of CTLA-4 and Foxp3 mRNA and STAT5,p-STAT5,CTLA-4 and Foxp3 protein were detected using Real-time PCR and Western blot,respectively.FACS was used to analyze the percentages of cells.A total of 117 genes overlapped between 785 target genes of the active compounds of WWXDD and 912 osteomyelitis related genes.Inflammation-related genes,including IL-6,TNFα,IL-1βand IL-2 showed high connection degree in the drug-compound-disease-target network.GO function and KEGG pathway analysis revealed that 117 intersection genes mainly enriched in virus infection related pathways,immune related pathways and chemokine signaling pathway.Furthermore,the development of chronic osteomyelitis was suppressed in model rats after treatment with WWXDD.Meanwhile,the concentrations of IL-2 and CD4^(+)CD25^(+)Foxp3 Treg perr centages together with the levels of p-STAT5,CTLA-4 and Foxp3 were also down-regulated.Furthermore,IL-2 and WWXDD drug-containing serum exhibited opposite effects on regulating IL-2,IL-10,TGF-β1,Foxp3,CTLA4 and STAT5.In addition,a STAT5 phosphorylation inhibitor suppressed the expression of Foxp3 and CTLA-4.WWXDD can treat chronic osteomyelitis through suppressing the main regulating factors of Tregs and interfere its immunodepression.Our results bring a new solution for chronic osteomyelitis.

Uncontrolled hemorrhage and subsequent trauma-induced coagulopathy (TIC) are still the principle causes for preventable death after trauma and early detection and aggressive management have been associated with reduced mortality. Despite increasing knowledge about trauma resuscitation, best practice to treat this newly defined entity is still under debate. A synopsis of best current knowledge with reference to the updated European trauma guideline on the management of severe trauma hemorrhage and TIC is presented. The implementation of evidence-based local protocols and algorithms including clinical quality and safety management systems together with parameters to assess key measures of bleeding control and outcome is advocated.

Rapid Detection of Cement Raw Meal Composition Based on Near Infrared Spectroscopy

The composition of cement raw materials was detected by near-infrared spectroscopy.It was found that the BiPLS-SiPLS method selected the NIR spectral band of cement raw materials,and the partial least squares regression algorithm was adopted to establish a quantitative correction model of cement raw materials with good prediction effect.The root-mean-square errors of SiO_(2),Al_(2)O_(3),Fe_(2)O_(3) and CaO calibration were 0.142,0.072,0.034 and 0.188 correspondingly.The results show that the NIR spectroscopy method can detect the composition of cement raw meal rapidly and accurately,which provides a new perspective for the composition detection of cement raw meal.

One-step synthesis of CoSn(OH)_6 nanocubes for high-performance all solid-state flexible supercapacitors

CoSn（OH）6 nanocubes were synthesized by a facile one-step co-precipitate method. Their supercapacitor performances were investigated via a traditional threeelectrode system and all solid-state flexible asymmetric supercapacitor. The electrode shows a maximum specific capacitance up to 695 F·g^-1. The flexible all solid-state asymmetric supercapacitor was fabricated based on CoSn（OH）6 nanocubes and active carbon as positive and negative electrode. The assembled asymmetric supercapacitor exhibits a specific capacitance（97.3 mF·cm^-2）, good rate/mechanical stability and a long cycling stability（5000 cycles）, demonstrating great promise for applications in all solid-state flexible supercapacitors.

Sulfur and Oxygen Isotopes of Sulfate Extracted from Early Cambrian Phosphorite Nodules： Implications for Marine Redox Evolution in the Yangtze Platform

Phosphorite nodule beds are discovered in the black shale of basal Niutitang Formation throughout the Yangtze Platform in South China, recording an important phosphorite-generation event. Platform-wide phosphorite precipitation requires special oceanographic and geochemical conditions, thus the origin of the Niutitang phosphorite nodules may provide valuable information about the ocean chemistry in the Early Cambrian. In this study, we measured sulfur and oxygen isotopic compositions of sulfate extracted from phosphorite nodules collected from the basal Niutitang Formation. Phosphorite associated sulfate（PAS） is a trace amount of sulfate that incorporates into crystal lattice during phosphorite precipitation, accordingly PAS records the geochemical signals during phosphorite nodule formation. Sulfur isotopic composition of PAS（δ^（34）S_（PAS）） ranges from-1.16‰ to ＋24.48‰（mean=＋8.19‰, n=11）, and oxygen isotopic value（δ^（18）O_（PAS）） varies between-5.3‰ and ＋26.3‰（mean=＋7.0‰, n=8）. Most phosphorite nodules have low δ^（34）SPAS and low δ^（18）O_（PAS） values, suggesting PAS mainly derived from anaerobic oxidation of H_2S within suboxic sediment porewater. We propose that phosphate was delivered to the Yangtze Platform by a series of upwelling events, and was scavenged from seawater with the precipitation of FeOOH. The absorbed phosphate was released into suboxic porewater by the reduction of FeOOH at the oxic-suboxic redox boundary in sediments, and phosphorite nodule precipitated by the reaction of phosphate with Ca^（2＋） diffused from the overlying seawater. The platform-wide deposition of phosphorite nodules in the basal Niutitang Formation implies the bottom water might be suboxic or even oxic, at least sporadically, in Early Cambrian. We speculate that the intensified ocean circulation as evident with frequent occurrences of upwelling events might be the primary reason for the episodic oxidation of the Yangtze Platform in Early Cambrian.

为了研究钢-混凝土组合梁在疲劳荷载作用下刚度的退化规律,设计并制作了6根栓钉试验梁进行了静力和疲劳试验。该研究对比了不同疲劳加载次数后试件的破坏形态,重点分析了组合梁在疲劳加载过程中荷载-挠度曲线、残余挠度以及相对滑移量的变化情况;将疲劳试验过程中每根试验梁的刚度退化与残余挠度、相对滑移增长之间作了相关性分析,探讨组合梁刚度退化的实质原因;基于刚度退化函数,通过试验数据拟合,建立了用于计算钢-混凝土组合梁刚度退化计算模型。结果表明:在疲劳荷载作用下,组合梁刚度会发生不可逆的退化,刚度退化实际上是材料疲劳损伤和抗剪连接程度退化的宏观表现;随疲劳循环次数增加,组合梁刚度退化规律呈现明显的“S”型单调递减趋势;该研究建立的组合梁刚度退化规律模型,计算值与试验值吻合良好,具有一定的适用性。该研究成果为组合梁疲劳荷载作用后刚度退化模型的建立提供了一种思路和方法。

针对现行规范中无法计算钢-混凝土组合梁在疲劳后的剩余承载力问题,提出考虑多组件疲劳损伤的组合梁剩余承载力的计算方法。基于材料剩余强度理论,分别引入组合梁各组件(混凝土板、钢梁和栓钉连接件)在疲劳荷载作用下的强度衰减模型;对疲劳荷载作用下的组合梁进行受力分析,得到在既定疲劳荷载幅值下各组件的疲劳应力幅;将各组件的疲劳损伤计入钢-混凝土组合梁剩余抗弯承载力计算中,并考虑疲劳加载过程中组合梁抗剪连接度的变化,建立完全抗剪连接和部分抗剪连接两种情形下的组合梁剩余承载力计算方法,并通过6个试验梁的剩余承载力试验进行验证。研究结果表明:在疲劳荷载作用下,组合梁的抗剪连接度逐渐降低,剩余承载力退化明显且不可忽略。该文建立的组合梁剩余承载力计算方法的计算值与试验值吻合较好,具有良好的计算精度与适用性,补充并完善了现有组合梁承载力的计算方法。

The discovery of superconducting states in the nickelate thin film with infinite-layer structure has paved a new way for studying unconventional superconductivity.So far,research in this field is still very limited due to difficulties in sample preparation.Here we report the successful preparation of the superconducting state of Nd_(0.8)Sr_(0.2)NiO_(2)thin film(T_(c)=8.0-11.1 K)and study the stability of such films in the ambient environment,water,and under electrochemical conditions.Our work demonstrates that the superconducting state of Nd_(0.8)Sr_(0.2)NiO_(2)is remarkably stable,which can last for at least 47-day continuous exposure to air at 20°C and 35%relative humidity.We also show that the superconductivity disappears after being immersed in de-ionized water at room temperature for 5 h.Surprisingly,it can also survive under ionic liquid gating conditions with an applied voltage of about 4 V,which is even more stable than conventional perovskite complex oxides.

Effects of Titanium and Silicon on the Swelling Behavior of 15–15Ti Steels by Heavy-Ion Beam Irradiation

The swelling behavior of 20% cold-worked 15-15Ti steels with （0.23-0.40） wt% titanium and （0.42-0.81） wt% silicon content after heavy-ion beam irradiation has been investigated by transmission electron microscopy. The results show that 15-15Ti steel with higher titanium content displays lower swelling. As silicon content increases, the cavity size and the swelling of the 15-15Ti steels decrease obviously. Titanium and silicon may play important roles in suppressing the cavity nucleation and growth in 15-15Ti steel.

Effect of Kang Ai injection + conventional chemotherapy on tumor markers, apoptotic molecules and immune response in patients with advanced NSCLC

Objective: To study the effects of Kang Ai injection + conventional chemotherapy on tumor markers, apoptotic molecules and immune response in patients with advanced non-small cell lung cancer (NSCLC). Methods: Patients diagnosed as advanced NSCLC in our hospital during March 2015 - October 2017 were randomly divided into the experimental group receiving Kang Ai injection + conventional chemotherapy and the control group receiving conventional chemotherapy. The contents of tumor markers, apoptotic molecules and immunologic cytokines in serum and the contents of immune cells in peripheral blood were determined before and after chemotherapy. Results: after chemotherapy, the content of CEA, CYFRA21-1, SCC-Ag, GDF15 and Livin in the serum of the two groups decreased significantly, and the content of PDCD5, P53 and Bax increased significantly, and the content of CEA, CYFRA21-1, SCC-Ag, GDF15, Livin in the serum of the experimental group were lower than those of the control group, and the content of PDCD5, P53 and Bax were higher than those of the control group;the content of CD3+ and CD4+ cells in peripheral blood and the content of IFN-γ, IL-2 in serum of control group significantly decreased, whereas the content of CD8+ cells in peripheral blood and the content of IL-4, IL-5 in serum significantly increased after chemotherapy;while the content of CD3+ and CD4+ cells in peripheral blood and the content of IFN-γ, IL-2 in serum of experimental group significantly increased, whereas the content of CD8+ cells in peripheral blood and the content of IL-4, IL-5 in serum significantly decreased after chemotherapy. Conclusion: Kang Ai injection + conventional chemotherapy in the treatment of advanced NSCLC can more significantly decrease the serum tumor markers, regulate the secretion of apoptotic molecules and anti-tumor immune response than conventional chemotherapy.
