microRNA-455-5p alleviates neuroinflammation in cerebral ischemia/reperfusion injury

Neuroinflammation is a major pathophysiological factor that results in the development of brain injury after cerebral ischemia/reperfusion.Downregulation of microRNA(miR)-455-5p after ischemic stroke has been considered a potential biomarker and therapeutic target for neuronal injury after ischemia.However,the role of miR-455-5p in the post-ischemia/reperfusion inflammatory response and the underlying mechanism have not been evaluated.In this study,mouse models of cerebral ischemia/reperfusion injury were established by transient occlusion of the middle cerebral artery for 1 hour followed by reperfusion.Agomir-455-5p,antagomir-455-5p,and their negative controls were injected intracerebroventricularly 2 hours before or 0 and 1 hour after middle cerebral artery occlusion(MCAO).The results showed that cerebral ischemia/reperfusion decreased miR-455-5p expression in the brain tissue and the peripheral blood.Agomir-455-5p pretreatment increased miR-455-5p expression in the brain tissue,reduced the cerebral infarct volume,and improved neurological function.Furthermore,primary cultured microglia were exposed to oxygen-glucose deprivation for 3 hours followed by 21 hours of reoxygenation to mimic cerebral ischemia/reperfusion.miR-455-5p reduced C-C chemokine receptor type 5 mRNA and protein levels,inhibited microglia activation,and reduced the production of the inflammatory factors tumor necrosis factor-αand interleukin-1β.These results suggest that miR-455-5p is a potential biomarker and therapeutic target for the treatment of cerebral ischemia/reperfusion injury and that it alleviates cerebral ischemia/reperfusion injury by inhibiting C-C chemokine receptor type 5 expression and reducing the neuroinflammatory response.

Atherosclerosis is a fundamental pathological change in coronary heart disease,with vulnerable plaque formation leading to decreased plaque stability and plaque rupture,and is a major cause of acute cardiovascular events.The inflammatory response is an important mechanism in the development and progression of atherosclerosis,and plaque stability is closely related to the inflammatory response.In recent years,the role of NLRP3 inflammatory vesicles,which are involved in macrophage foaminess,in the inflammatory response to atherosclerosis has received increasing attention.Chinese medicine is able to modulate inflammatory vesicles to improve atherosclerotic plaque stability,showing good promise in the prevention and treatment of atherosclerotic vulnerable plaques.Based on the role of inflammatory vesicles in atherosclerosis and the results of previous studies that wind-extinguishing and phlegm-resolving medicine can effectively modulate the inflammatory response to intervene in the treatment of atherosclerosis,this paper aims to investigate the treatment of coronary atherosclerosis by wind-extinguishing and phlegm-resolving medicine from the direction of inflammatory vesicles.

Dear Editor,Hepatoblastoma(HB)is a predominant hepatic neoplasm that develops in children from 0 to 4 years of age at the rate of 2.16 per 1,000,000.It originates from abnormal differentiation of hepatocyte precursors(hepatoblasts)during embryogenesis(Sumazin et al.,2017).Approximately 20%of children with HB have metastasis in lung at diagnosis,which indicates poor prognosis(Angelico et al.,2019).While surgery in combination of chemotherapy and/or metastasectomy is the most popular therapy,relapse happens in a significant portion of HB patients(Zhang et al.,2021).Therefore,novel and less aggressive therapies targeting the pathogenesis of HB should be explored to prolong patients’s disease-free survival as well as to improve their quality of life.

中国电网新能源装机不断增加,对电力系统暂态电压稳定产生了显著影响。在新能源低电压穿越控制作用下,电力系统出现了新的电压失稳现象。文中从电力系统的新型P-V特性入手,研究系统的暂态行为,直观解释了实际运行中“持续低电压穿越”“反复低电压穿越”等新型失稳现象的物理机理,借助混杂系统理论建立了新型失稳现象的动力学模型。为克服混杂系统模型表达式复杂、难以解析分析的难题,文中使用现有的成熟计算工具,利用不同边界条件下的潮流计算结果,提出并验证了暂态电压失稳的判别方法。

Multicellular organisms such as plants contain various cell types with specialized functions.Analyzing the characteristics of each cell type reveals specific cell functions and enhances our understanding of organization and function at the organismal level.Guard cells(GCs)are specialized epidermal cells that regulate the movement of the stomata and gaseous exchange,and provide a model genetic system for analyzing cell fate,signaling,and function.Several proteomics analyses of GC are available,but these are limited in depth.Here we used enzymatic isolation and flow cytometry to enrich GC and mesophyll cell protoplasts and perform in-depth proteomics in these two major cell types in Arabidopsis leaves.We identified approximately 3,000 proteins not previously found in the GC proteome and more than600 proteins that may be specific to GC.The depth of our proteomics enabled us to uncover a guard cell-specific kinase cascade whereby Raf15and Snf1-related kinase2.6(SnRK2.6)/OST1(open stomata 1)mediate abscisic acid(ABA)-induced stomatal closure.RAF15 directly phosphorylated SnRK2.6/OST1 at the conserved Ser175 residue in its activation loop and was sufficient to reactivate the inactive form of SnRK2.6/OST1.ABAtriggered SnRK2.6/OST1 activation and stomatal closure was impaired in raf15 mutants.We also showed enrichment of enzymes and flavone metabolism in GC,and consistent,dramatic accumulation of flavone metabolites.Our study answers the long-standing question of how ABA activates SnRK2.6/OST1 in GCs and represents a resource potentially providing further insights into the molecular basis of GC and mesophyll cell development,metabolism,structure,and function.

Background:Highly emetogenic chemotherapy induces emesis in cancer patients without prophylaxis.The purpose of this study was to evaluate the efficacy and safety of a fosaprepitant-based triple antiemetic regimen for the prevention of chemotherapy-induced nausea and vomiting(CINV)in patients with solid malignant tumors,determine risk factors and externally validate different personalized risk models for CINV.Methods:This phase III trial was designed to test the non-inferiority of fosaprepitant toward aprepitant in cancer patients who were to receive the first cycle of single-day cisplatin chemotherapy.The primary endpoint was complete response(CR)during the overall phase(OP)with a non-inferiority margin of 10.0%.Logistic regression modelswere used to assess the risk factors ofCRand no nausea.To validate the personalized risk models,the accuracy of the risk scoring systems was determined by measuring the specificity,sensitivity and area under the receiver operating characteristic(ROC)curve(AUC),while the predictive accuracy of the nomogram was measured using concordance index(C-index).Results:A total of 720 patients were randomly assigned.CR during the OP in the fosaprepitant group was not inferior to that in the aprepitant group(78.1%vs.77.7%,P=0.765)with a between-group difference of 0.4%(95%CI,-5.7%to 6.6%).Female sex,higher cisplatin dose(≥70 mg/m2),no history of drinking and larger body surface area(BSA)were significantly associated with nausea.The AUC for the acute and delayed CINV risk indexes was 0.68(95%CI:0.66-0.71)and 0.66(95%CI:0.61-0.70),respectively,and the C-index for nomogram CINV prediction was 0.59(95%CI,0.54-0.64).Using appropriate cutoff points,the three models could stratify patients with high-or low-risk CINV.No nausea and CR rate were significantly higher in the low-risk group than in the high-risk group(P<0.001).Conclusions:Fosaprepitant-based triple prophylaxis demonstrated non-inferior control for preventing CINV in patients treated with cisplatin-base chemotherapy.Female cancer patients

The coronavirus disease 2019(COVID-19)has been declared as a pandemic by the World Health Organization.[1]Most COVID-19 patients exhibit mild to moderate symptoms,while approximately 15%progress rapidly to severe pneumonia,and about 5%eventually develop acute respiratory distress syndrome(ARDS),[2]which requires mechanical ventilation(MV)and even extracorporeal membrane oxygenation.The mortality of COVID-19 patients who received MV was reported to be as high as 66%.[3]Therefore,the treatments aiming to improve mortality should focus on two aspects:first,prevention of the aggravation of the disease in mild and moderate COVID-19 patients;second,the rescue therapy for patients in serious conditions.We have been applying high-dose intravenous vitamin C(HDIVC)in the treatment of critical illnesses for almost 10 years in our center.Our previous in vivo research showed that HDIVC protected hemorrhagic shock-related multiple organ failure(MOF)by inhibiting inflammatory cytokines and oxidative indicators through activating Sirtuin1 pathway.

高气压下三维旋转滑动弧放电光谱特性分析

为探究气体压力对滑动弧放电稳定性的影响规律,在高气压放电实验平台上,开展了滑动弧放电光谱特性研究。利用发射光谱法对滑动弧放电过程中的激发态物质进行诊断,并对不同气体压力条件下滑动弧放电的电子密度、振动温度、转动温度进行了计算。在滑动弧放电发射光谱中观测到氮气第二正带系N_(2)(C^(3)Π_(u)→B^(3)Π_(g))和氮气第一负带系N^(+)_(2)(B^(2)Σ^(+)_(u)→X^(2)Σ^(+)_(g))的发射谱线,随着气体压力升高,氮气第二正带系谱线发射强度增强,氮气第一负带系的发射强度变化幅度较小。当气体压力超过0.26 MPa之后,在滑动弧放电发射光谱中发现了氮气Gaydon s Green带系507.4 nm的发射谱线,其发射强度为3508.49 arb,随气体压力升高其谱线发射强度无明显变化。利用Stark展宽法对滑动弧放电过程中的电子密度进行了计算,得到电子密度在1023量级,并且随着气体压力的增加,滑动弧放电的电子密度呈线性增加。针对氮气分子第二正带系的5条谱线,采用玻尔兹曼图解法对滑动弧放电的振动温度进行计算,振动温度随气体压力的变化并非单调变化,在0.3 MPa之前振动温度变化幅度较小,气体压力超过0.3 MPa之后振动温度迅速增加。对氮气分子第一负带系390~391.6 nm处的发射谱线进行拟合计算滑动弧放电的转动温度,结果表明在0.24 MPa之前转动温度随气体压力升高变化幅度较大,气体压力超过0.24 MPa之后转动温度增长幅度变小。

Sporopollenin in the pollen cell wall protects male gametophytes from stresses.Phenylpropanoid derivatives,including guaiacyl(G)lignin units,are known to be structural components of sporopollenin,but the exact composition of sporopollenin remains to be fully resolved.We analyzed the phenylpropanoid derivatives in sporopollenin from maize and Arabidopsis by thioacidolysis coupled with nuclear magnetic resonance(NMR)and gas chromatography–mass spectrometry(GC–MS).The NMR and GC–MS results confirmed the presence of p-hydroxyphenyl(H),G,and syringyl(S)lignin units in sporopollenin from maize and Arabidopsis.Strikingly,H units account for the majority of lignin monomers in sporopollenin from these species.We next performed a genome-wide association study to explore the genetic basis of maize sporopollenin composition and identified a vesicle-associated membrane protein(ZmVAMP726)that is strongly associated with lignin monomer composition of maize sporopollenin.Genetic manipulation of VAMP726 affected not only lignin monomer composition in sporopollenin but also pollen resistance to heat and UV radiation in maize and Arabidopsis,indicating that VAMP726 is functionally conserved in monocot and dicot plants.Our work provides new insight into the lignin monomers that serve as structural components of sporopollenin and characterizes VAMP726,which affects sporopollenin composition and stress resistance in pollen.
