Cucumber mosaic virus（CMV） can infect a wide range of host species. For the lacking of CMV resistant varieties of tomato, RNA interference（RNAi） can be used as a fast and effective method for the generation of transgenic resistant varieties. In this current study, five intron-spliced hairpin RNA（ihp RNA） plant expression vectors aimed at five genes of CMV have been constructed. Transgenic tomatoes were obtained by Agrobacterium tumefaciens-mediated transformation with expression vectors. Highly resistant generations of transgenic plants were employed as rootstocks and grafted onto non-transgenic tomatoes that resulted in the successful transfer of resistance to the scions. Using a novel method of plant cuttings for rootstock propagation, we obtained large quantities of disease-resistant material. Further, this method produces scions that can remain undetectable for transgenic resistance marker genes that may provide novel approaches to evade collective concerns about genetically-modified organism（GMO） biosafety.

Highway frost heave and thawing settlement caused by water migration towards the freezing front and ice lens development is widespread in the alpine meadow area of the southeast QinghaiTibet Plateau(QTP). A laboratory experiment on a highway reconstruction and expansion project in the QTP was carried out in this work to analyze the effects of fine particle content, initial water content, and the number of freeze-thaw cycles(FTCs) on frost depth, temperature gradient(Grad T), total water intake, and water intake flux. Based on the results of the laboratory experiment, a modified model of migration potential related to fine particle content, freeze-thaw history, and freezing time was established. The results show that, with the increase of fine particle content, the frost depth of soil decreases, the curve of total water intake over time is transformed from an Sshape to an arch, and the curve of water intake flux over time is transformed from a peak shape to descending shape. The variation trend of migration potential with freezing time and the freeze-thaw history is the same as that of water intake flux with freezing time and freeze-thaw history. The variation trend of soil intake flux can be used as a reference to determine the variation trend of soil migration potential. This study provides a reference for the design and construction of highway subgrade in the alpine meadow area of the QTP.

Numerical modeling of tidal currents,sediment transport and morphological evolution in Hangzhou Bay,China

A 2D depth-averaged numerical model is set up to simulate the macro-scale hydrodynamic characteristics,sediment transport patterns and morphological evolution in Hangzhou Bay,a large macro-tidal estuary on the eastern coast of *** incorporating the shallow water equations,the suspended sediment transport equation and the mass-balance equation for sediment;short-term hydrodynamics,sediment transport and long-term morphological evolution for Hangzhou Bay are simulated and the underly.ng physical mechanisms are *** model reproduces the spatial distribution patterns of suspended sediment concentration(SSC) in Hangzhou Bay,characterized by three high SSC zones and two low SSC *** also correctly simulates the residual flow,the residual sediment transport and the sediment accumulation patterns in Hangzhou *** model results are in agreement with previous studies based on field *** residual flow and the residual sediment transport are landwards directed in the northern part of the bay and seawards directed in the southern *** accumulation takes place in most areas of the *** analysis revealed that the tide is flood-dominant in the northern part of the bay and ebb-dominant in the southern part of the *** strength of the flood-dominance increases landwards along the northern Hangzhoii *** turn sediment transport in Hangzhou Bay is controlled by this tidal asymmetry *** addition,the direction of tidal propagation in the East China Sea,the presence of the archipelago in the southeast and the funnel-shaped geometry of the bay,play important roles for the patterns of sediment transport and sediment accumulation respectively.

The improvement of the accuracy of simulated cloud-related variables,such as the cloud fraction,in global climate models(GCMs)is still a challenging problem in climate *** this study,the influence of cloud microphysics schemes(one-moment versus two-moment schemes)and cloud overlap methods(observation-based versus a fixed vertical decorrelation length)on the simulated cloud fraction was assessed in the BCC_AGCM2.0_CUACE/*** with the fixed decorrelation length method,the observation-based approach produced a significantly improved cloud fraction both globally and for four representative *** utilization of a two-moment cloud microphysics scheme,on the other hand,notably improved the simulated cloud fraction compared with the one-moment scheme;specifically,the relative bias in the global mean total cloud fraction decreased by 42.9%–84.8%.Furthermore,the total cloud fraction bias decreased by 6.6%in the boreal winter(DJF)and 1.64%in the boreal summer(JJA).Cloud radiative forcing globally and in the four regions improved by 0.3%−1.2% and 0.2%−2.0%,***,our results showed that the interaction between clouds and climate through microphysical and radiation processes is a key contributor to simulation uncertainty.

Jincheng orange(Citrus sinensis Osbeck)is widely grown in Chongqing,China,and is commonly consumed because of its characteristic aroma contributed by the presence of diverse volatile *** changes in aroma during the development and maturation of fruit are indicators for ripening and harvest ***,the influence of growth stages on the volatile compounds in Jincheng orange remains *** addition,volatiles originate from fatty acids,most of which are the precursors of volatile *** this basis,gas chromatography-mass spectrometry(GC-MS)was performed to elaborate the changes in volatile constituents and fatty acids as *** study tested proximately 60 volatiles and 8 fatty acids at 9 growth and development stages(AF1-AF9).Of those compounds,more than 92.00%of total volatiles and 87.50%of fatty acids were terpenoid and saturated fatty acids,*** shown in the PCA plot,the AF5,AF6,and AF9 stages were confirmed as completely segregated and appeared *** addition,most of the volatiles and fatty acids first increased at the beginning of the development stage,then decreased from the AF6 development stage,and finally increased at the AF9 maturity ***,the highest contents of terpenoid,alcohols,aldehydes,ketones,and saturated fatty acids in Jincheng orange peel oil were d-limonene,linalool,octanal,cyclohexanone,and stearic acid during development stages,*** results found that the growth stages significantly affected the volatile constituents and precursors in Jincheng orange peel oil.

Heat stress(HS)can be detrimental to the gut health of *** negative outcomes induced by HS are increasingly recognized as including modulation of intestinal *** turn,the intestinal microbiota is a unique ecosystem play.ng a critical role in mediating the host stress ***,we aimed to characterize gut microbiota of pigs’exposure to short-term HS,to explore a possible link between the intestinal microbiota and HS-related changes,including serum cytokines,oxidation status,and intestinal epithelial barrier *** findings showed that HS led to intestinal morphological and integrity changes(villus height,serum diamine oxidase[DAO],serum D-lactate and the relative expressions of tight junction proteins),reduction of serum cytokines(interleukin[IL]-8,IL-12,interferongamma[IFN-g]),and antioxidant activity(higher glutathione[GSH]and malondialdehyde[MDA]content,and lower superoxide dismutase[SOD]).Also,16S rRNA sequencing analysis revealed that although there was no difference in microbial a-diversity,some HS-associated composition differences were revealed in the ileum and cecum,which partly led to an imbalance in the production of short-chain fatty acids including propionate acid and valerate *** networks revealed that HS-derived changes in bacterial genera and microbial metabolites,such as Chlamydia,Lactobacillus,Succinivibrio,Bifidobacterium,Lachnoclostridium,and propionic acid,were correlated with oxidative stress,intestinal barrier dysfunction,and inflammation in ***,our observations suggest that intestinal damage induced by HS is probably partly related to the gut microbiota dysbiosis,though the underly.ng mechanism remains to be fully elucidated.

The Community Microwave Emission Model (CMEM) developed by the European Centre for Me-dium-Range Weather Forecasts (ECMWF) can provide a link between surface states and satellite observations and simulate the passive microwave brightness temperature of the surface at low frequencies (from 1 GHz to 20 GHz).This study evaluated the performance of the CMEM cou-pled with the Community Land Model (CLM) (CMEM-CLM) using C-band (6.9 GHz) microwave brightness temperatures from the Advanced Microwave Scanning Radiometer on Earth Observing System (AMSR-E) over East *** results support the argument that the simulated brightness temperatures of CMEM-CLM from July 2005 to June 2010 are comparable to AMSR-E observational ***-CLM performed better for vertical polarization,for which the root mean square error was approximately 15 K,compared to over 30 K for horizontal *** evaluation performed over seven sub-regions in China indicated that CMEM-CLM was able to capture the temporal evolution of C-band brightness temperatures well,and the best correlation with AMSR-E appeared over western Northwest China (over 0.9 for vertical polarization).However,larger biases were found over southern North China and the middle and lower reaches of the y.ngtze River.
