Clustered regulatory interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9 nuclease(Cas9),the third-generation genome editing tool,has been favored because of its high efficiency and clear system composition.In this technology,the introduced double-strand breaks(DSBs)are mainly repaired by non-homologous end joining(NHEJ)or homology-directed repair(HDR)pathways.The high-fidelity HDR pathway is used for genome modification,which can introduce artificially controllable insertions,deletions,or substitutions carried by the donor templates.Although high-level knock-out can be easily achieved by NHEJ,accurate HDR-mediated knock-in remains a technical challenge.In most circumstances,although both alleles are broken by endonucleases,only one can be repaired by HDR,and the other one is usually recombined by NHEJ.For gene function studies or disease model establishment,biallelic editing to generate homozygous cell lines and homozygotes is needed to ensure consistent phenotypes.Thus,there is an urgent need for an efficient biallelic editing system.Here,we developed three pairs of integrated selection systems,where each of the two selection cassettes contained one drug-screening gene and one fluorescent marker.Flanked by homologous arms containing the mutated sequences,the selection cassettes were integrated into the target site,mediated by CRISPR/Cas9-induced HDR.Positively targeted cell clones were massively enriched by fluorescent microscopy after screening for drug resistance.We tested this novel method on the amyloid precursor protein(APP)and presenilin 1(PSEN1)loci and demonstrated up to 82.0%biallelic editing efficiency after optimization.Our results indicate that this strategy can provide a new efficient approach for biallelic editing and lay a foundation for establishment of an easier and more efficient disease model.

As one of the most well-documented biogeographic patterns,the distance-decay relation-ship provides insights into the underlying mechanisms driving biodiversity distribution.Al-though wastewater treatment plants(WWTPs)are well-controlled engineered ecosystems,this pattern has been seen among microbial communities in activated sludge(AS).However,little is known about the relative importance of environmental heterogeneity and dispersal limitation in shaping AS microbial community across China;especially they are related to spatial scale and organism types.Here,we assessed the distance-decay relationship based on different spatial scales and microbial phylogenetic groups by analyzing 132 activated sludge(AS)samples across China comprising 3,379,20016S rRNA sequences.Our results in-dicated that the drivers of distance-decay pattern in China were scale-dependent.Microbial biogeographic patterns in WWTPs were mainly driven by dispersal limitation at both local and national scales.In contrast,conductivity,SRT,and pH played dominant roles in shaping AS microbial community compositions at the regional scale.Turnover rates and the drivers of beta-diversity also varied with microorganism populations.Moreover,a quantitative re-lationship between dispersal limitation ratio and AS microbial turnover rate was generated.Collectively,these results highlighted the importance of considering multiple spatial scales and micro-organism types for understanding microbial biogeography in WWTPs and pro-vided new insights into predicting variations in AS community structure in response to environmental disturbance.

Dynamic modeling and simulation of a pressurized system used in flight vehicle

For a typical pressurized system with a novel dual-stage gas pressure reducing regulator,a system model is established with modular models of various typical components. The simulation study on the whole working period shows that the general trends and magnitudes of simulation curves are in agreement with experimental measured curves. As the key component in the pressurized system, the regulator is studied by a series of numerical simulations to reveal the influences of various structure parameters on its stability. Furthermore, the variable ranges which can guarantee the stability of regulator and system are obtained to provide guidance for design. The modeling and analysis approach can be applied to other systems and components.

The smart distribution network(SDN)is integrat ing increasing distributed generation(DG)and energy storage(ES).Hosting capacity evaluation is important for SDN plan ning with DG.DG and ES are usually invested by users or a third party,and they may form friendly microgrids(MGs)and operate independently.Traditional centralized dispatching meth od no longer suits for hosting capacity evaluation of SDN.A quick hosting capacity evaluation method based on distributed optimal dispatching is proposed.Firstly,a multi-objective DG hosting capacity evaluation model is established,and the host ing capacity for DG is determined by the optimal DG planning schemes.The steady-state security region method is applied to speed up the solving process of the DG hosting capacity evalua tion model.Then,the optimal dispatching models are estab lished for MG and SDN respectively to realize the operating simulation.Under the distributed dispatching strategy,the dual-side optimal operation of SDN-MGs can be realized by several iterations of power exchange requirement.Finally,an SDN with four MGs is conducted considering multiple flexible resources.It shows that the DG hosting capacity of SDN oversteps the sum of the maximum active power demand and the rated branch capacity.Besides,the annual DG electricity oversteps the maximum active power demand value.

SMS4 is released by the Chinese government as part of the WAPI standard for the wireless networks.At ICICS 2007 and CRYPTO 2015, Lu and sun et al. constructed some 12-round impossible differentials and12-round zero correlation linear hulls, respectively. In this paper, it is proved that the distinguishers constructed by Lu and sun et al. are independent with the details of the non-linear layers though they concentrated on the specific S-boxes. This indicates that for the structure deduced by SMS4, there always exist 12-round impossible differentials and 12-round zero correlation linear hulls.

Homochiral metal-organic frameworks(MOFs)have attracted considerable attention in many fields of research,such as chiral catalysis and chiral chromatography.However,only few homochiral MOFs can be effectively used in capillary electrochromatography(CEC)and their performances are far from adequate.In this study,we successfully synthesized achiral nanocrystalline MIL-53.A facile post-synthetic modification strategy was then implemented to functionalize the product,yielding a homochiral MOF:L-His-NH-MIL-53.This MOF was then employed as a chiral coating in open-tubular CEC mode(OT-CEC),and,as such,it exhibited high enantioselectivities for several racemic drugs.The homochiral MOF and the fabricated capillary coating were systematically characterized using transmission electron microscopy,scanning electron microscopy(with energy-dispersive X-ray spectrometry),Fourier-transform infrared spectroscopy,X-ray diffractometry,thermogravimetric analysis,circular dichroism spectroscopy,Brunauer-Emmett-Teller surface area measurements,and X-ray photoelectron spectroscopy.This study is expected to provide a new strategy for the design and establishment of MOF-based chiral OT-CEC systems.

Cryptanalysis of Full-Round Magpie Block Cipher

Magpie is a lightweight block cipher proposed by Li et al. in Acta Electronica Sinica volumn 45,issue 10. It adopts an substitution-permutation network(SPN) structure with a block size of 64 bits and the key size of 96 bits, respectively. To achieve the consistency of the encryption and decryption, which is both hardware and software friendly, 16 bits of the key are used as control signals to select S-boxes and another 16 bits of the key are used to determine the order of the operations. As the designers claimed, the security might be improved as different keys generate different ciphers. This paper analyzes the security of Magpie, studies the difference propagation of Magpie, and finally finds that the cipher has a set of 280weak keys which makes the full-round encryption weak, and corrects the lower bound of the number of active S-boxes to 10 instead of 25 proposed by the designers. In the weak key model, the security of the cipher is reduced by the claimed 280to only 4×216.

Let be an n -Lie superalgebra. We study the double derivation algebra D ( ) D ( ) D() and describe the relation between D ( ) D ( ) D() and the usual derivation Lie superalgebra Der(). We show that the set D ( ) D ( ) D() of all double derivations is a subalgebra of the general linear Lie superalgebra gl() and the inner derivation algebra ad() is an ideal of D ( ) D ( ) D() . We also show that if is a perfect n -Lie superalgebra with certain constraints on the base field, then the centralizer of ad() in D ( ) D ( ) D() is trivial. Finally, we give that for every perfect n -Lie superalgebra , the triple derivations of the derivation algebra Der() are exactly the derivations of Der().

A Multi-mode Electronic Load Sensing Control Scheme with Power Limitation and Pressure Cut-off for Mobile Machinery

In mobile machinery,hydro-mechanical pumps are increasingly replaced by electronically controlled pumps to improve the automation level,but diversified control functions(e.g.,power limitation and pressure cut-off)are integrated into the electronic controller only from the pump level,leading to the potential instability of the overall system.To solve this problem,a multi-mode electrohydraulic load sensing(MELS)control scheme is proposed especially considering the switching stability from the system level,which includes four working modes of flow control,load sensing,power limitation,and pressure control.Depending on the actual working requirements,the switching rules for the different modes and the switching direction(i.e.,the modes can be switched bilaterally or unilaterally)are defined.The priority of different modes is also defined,from high to low:pressure control,power limitation,load sensing,and flow control.When multiple switching rules are satisfied at the same time,the system switches to the control mode with the highest priority.In addition,the switching stability between flow control and pressure control modes is analyzed,and the controller parameters that guarantee the switching stability are obtained.A comparative study is carried out based on a test rig with a 2-ton hydraulic excavator.The results show that the MELS controller can achieve the control functions of proper flow supplement,power limitation,and pressure cut-off,which has good stability performance when switching between different control modes.This research proposes the MELS control method that realizes the stability of multi-mode switching of the hydraulic system of mobile machinery under different working conditions.
