An efficient and eco-friendly carbon nitride nanosheet(NM-g-C3N4)-catalyzed decarboxylative coupling reaction of imidazo-fused heterocycles(i.e.,imidazo[1,2-a]pyridines,benzo[d]imidazo[2,1-b]thiazole)with N-phenylglycines in dimethyl carbonate(DMC)has been developed.The toxic solvents,external oxidants,and restricted reaction conditions could be effectively avoided in this powerful and sustainable protocol.Remarkably,NM-g-C3N4 could be straightforwardly recovered by simple centrifugation and recycled and reused at least 7 times without an obvious decrease in catalytic activity.

While drug chirality remains one of the most important themes in medicinal chemistry and fluorine-containing drugs have comprised large portion of global pharmaceutical market,the synthesis of fluorine-containing compounds featuring a trifluoromethylated stereogenic carbon center is still underdeveloped.Recent strategy of enantioconvergent cross-coupling of trifluoromethylated alkyl halide has significantly updated previous toolbox that is dependent on additional functional group to achieve desired bond formation and highly enantioselective control.However,evident limitations were present in specific coupling candidate(sp?carbon)and/or prerequisite heteroatom in trifluoroalkyl source for inducing effective stereoconvergent differentiation.Leveraging novel design of sterically hindered bisoxazoline ligand and drastic increase of molecular complexity of in situ generated sp3 coupling partner via metal-hydride atom transfer,herein,we report a nickel-catalyzed,highly enantioselective hydrotrifluoroalkylation of unactivated alkenes for diverse synthesis of enantioenriched aliphatic alkanes featuring trifluoromehylated stereogenic carbon.Excellent stereochemical control(mostly>95:5 er),high catalytic reactivity,mild conditions,and broad substrate scope(40+examples),enable convenient late-stage asymmetric trifluoroalkylation of various biologically active complexmolecules.

Stromal cell-derived factor-1 and its receptor C-X-C chemokine receptor 4(CXCR4) have been shown to regulate neural regeneration after stroke.Howeve r,whether stromal cell-derived factor-1 receptor CXCR7,which is widely distributed in the develo ping and adult central nervous system,participates in neural regeneration remains poorly unde rstood.In this study,we established rat models of focal cerebral ischemia by injecting endothelin-1 into the cerebral co rtex and striatum.Starting on day 7 after injury,CXCR7-neutralizing antibody was injected into the lateral ventricle using a micro drug delivery system for 6 consecutive days.Our results showed that CXCR7-neutralizing antibody increased the total length and number of sprouting co rticospinal tra ct fibers in rats with cerebral ischemia,increased the expression of vesicular glutamate transporter 1 and growth-related protein 43,marke rs of the denervated spinal cord synapses,and promoted the differentiation and maturation of oligodendrocyte progenitor cells in the striatum.In addition,CXCR7 antibody increased the expression of CXCR4 in the striatum,increased the protein expression of RAS and ERK1/2 associated with the RAS/ERK signaling pathway,and im proved rat motor function.These findings suggest that CXCR7 improved neural functional recovery after ischemic stroke by promoting axonal regeneration,synaptogenesis,and myelin regeneration,which may be achieved by activation of CXCR4 and the RAS/ERK1/2 signaling pathway.

Reported herein is an example of highly regio-, diastereo-and enantioselective Cu（I）-catalyzed intermolecular [3＋2] cycloaddition reaction of a-substituted iminoesters with α-trifluoromethyl α,β-unsaturated esters. This novel strategy provided a facile access to pyrrolidines with two skipped （aza）quaternary stereocenters including a CF3 all-carbon quaternary stereocenter. A broad substrate scope was observed and high yields （up to 94%） with excellent diastereoselectivity （up to 〉20 ： 1 d.r.） and enantioselectivity （up to 98% ee） were obtained.

Quantitative analysis of the effect of the PAY1 gene on rice canopy structure during different reproductive stages

The leaf and stem types are core structural characteristics of the rice phenotype that determine the light interception ability of the canopy and directly affect crop yield.The PLANT ARCHITECTURE AND YIELD 1(PAY1)gene has been shown to alter the prostrate growth habit of wild rice and to inhibit the wild rice prostrate growth gene PROSTRATE GROWTH 1(PROG1).In this paper,the wild rice introgression line YIL55,which contains the PROG1 gene;its mutant,PAY1;and its parent,TQ,were used as test varieties to construct three-dimensional(3D)canopy structure models based on 3D digital assay technology.On this basis,quantitative analyses of the PAY1 gene and the plant leaf and stem types at the jointing,heading and filling stages were performed.Under the influence of the PAY1 gene,the plant stem and leaf angles from vertical decreased significantly;the plants were upright,with larger leaves;the culm angle changed from loose to compact;and the average tiller angle during the three key reproductive stages decreased from 44.9,28.5 and 21.3°to 17.6,8.4 and 10.5°,respectively.Moreover,the PAY1 mutant retained the PROG1 gene characteristic of exhibiting dynamic changes in the tiller angle throughout the growth period,and its culm angle changed from loose during the jointing stage to compact during the heading stage.The measurements of photosynthetically active radiation(PAR)in the canopy also showed that the mutant PAY1 allowed more PAR to reach the bottom of the canopy than the other varieties.The light-extinction coefficients for PAY1 at the jointing,heading and filling stages were 0.535,0.312 and 0.586,respectively,which were lower than those of the other two varieties.In this study,the influence of the PAY1 gene on rice canopy structural characteristics was quantitatively analyzed to provide effective canopy structure parameters for breeding the ideal plant type.

Thalassemia is a group of genetically heterogeneous diseases characterized by hemolytic anemia.To investigate molecular characteristics ofα-andβ-thalassemia among young individuals of marriageable age in Guangdong Province,24,788 subjects with suspected thalassemia were genetically tested forα-andβ-thalassemia by GapPCR and reverse dot blot during 2018–2019.For suspected rare thalassemia cases,DNA sequencing was performed to identify rare and unknown thalassemia gene mutations.A total of 14,346 thalassemia carriers were detected,including 7,556 cases ofα-thalassemia with 25 genotypes and 8α-gene mutations identified,5,860 cases ofβ-thalassemia with 18 genotypes and 18β-gene mutations identified,and 930 cases of compoundα/β-thalassemia.Among them,the frequency of--SEA deletion was the highest inα-thalassemia(66.01%),followed by-α3.7(17.98%)and-α.24(8.22%),and the frequency of CD41-42(-TCTT)mutation was the highest inβ-thalassemia(38.38%),followed by IVS-II-654(C>T)(25.67%),-28(A>G)(15.76%),and CD17(10.01%).In addition,5 rare mutations(--THAI and HKαα,CD113,-90,and CD56)were found in the study population.Our results revealed molecular epidemiological background ofα-andβ-thalassemia in Guangdong Province,which can support optimization of thalassemia prevention and control strategies.We demonstrated that thalassemia is heterogeneous with significant geographical differences and population specificity.

Epigenetic information carried by histone modifications not only reflects the state of gene expression,but also participates in the maintenance of chromatin states and the regulation of gene expression.Recycling of parental histones to daughter chromatin after DNA replication is vital to mitotic inheritance of epigenetic information and the maintenance of cell identity,because the locus-specific modifications of the parental histones need to be maintained.To assess the precision of parental histone recycling,we developed a synthetic local label-chasing system in budding yeast Saccharomyces cerevisiae.Using this system,we observed that parental histone H3 can be recycled to their original position,thereby recovering their position information after DNA replication at all tested loci,including heterochromatin boundary,non-transcribed region,and actively transcribed regions.Moreover,the recycling rate appears to be affected by local chromatin environment.We surveyed a number of potential regulatory factors and observed that histone H3-H4 chaperon Asf1 contributed to parental histone recycling,while the eukaryotic replisome-associated components Mcm2 and Dpb3 displayed compounding effects in this process.In addition,the FACT complex also plays a role in the recycling of parental histones and helps to stabilize the nucleosomes.

Glucagon-like peptide-1 receptor has anti-apoptotic,anti-inflammatory,and neuroprotective effects.It is now recognized that the occurrence and development of chronic pain are strongly associated with anti-inflammatory responses;however,it is not clear whether glucagon-like peptide-1 receptor regulates chronic pain via anti-inflammatory mechanisms.We explored the effects of glucagon-like peptide-1 receptor on nociception,cognition,and neuroinflammation in chronic pain.A rat model of chronic pain was established using left L5 spinal nerve ligation.The glucagon-like peptide-1 receptor agonist exendin-4 was intrathecally injected into rats from 10 to 21 days after spinal nerve ligation.Electrophysiological examinations showed that,after treatment with exendin-4,paw withdrawal frequency of the left limb was significantly reduced,and pain was relieved.In addition,in the Morris water maze test,escape latency increased and the time to reach the platform decreased following exendin-4 treatment.Immunohistochemical staining and western blot assays revealed an increase in the numbers of activated microglia and astrocytes in the dentate gyrus of rat hippocampus,as well as an increase in the expression of tumor necrosis factor alpha,interleukin 1 beta,and interleukin 6.All of these effects could be reversed by exendin-4 treatment.These findings suggest that exendin-4 can alleviate pain-induced neuroinflammatory responses and promote the recovery of cognitive function via the glucagon-like peptide-1 receptor pathway.All experimental procedures and protocols were approved by the Experimental Animal Ethics Committee of Renmin Hospital of Wuhan University of China(approval No.WDRM 20171214)on September 22,2017.
