Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

Fairness of Extra-Gain Guilty in Performance of Supply Chain and Contract Design

Considering a one-supplier and two-retailer supply chain setting in which the supplier makes contacts with retailers in sequence based on FCFS(first–come–first-service) principle, the authors investigate the marginal effect of extra-gain guilty fairness concerns of supply chain members on the contract design, supply chain's profit and profit distribution. When the supplier considers distributional fairness of loss aversion and extra-gain guilty, extra-gain guilty concern reduces the optimal wholesale and retail price, and improves supply chain performance. When the first or the second retailer considers distributional fairness and peer-induced fairness, extra-gain guilty concerns decrease the optimal wholesale and retail price but increase the profit of all supply chain members only when the level of loss aversion is large enough and the level of extra-gain guilty is less than 1/2. However, when the level of loss aversion is small and the level of extra-gain guilty is less than 1/2, extra-gain guilty has no influence on supply chain performance. The analysis reveals that the supply chain performance can be further improved by considering fairness of extra-gain guilty.

The welding arc,as a carrier for the conversion of electrical energy to thermal energy,has a direct impact on the quality of welding by its properties and *** the tungsten inert gas(TIG)welding process under the condition of Ar-He alternating gas supply,the arc is alternately converted between Ar arc and He arc with an alternating gas supply cycle,which has obvious arc change *** FLUENT software was used to numerically simulate the characteristics of the TIG arc under the condition of alternating gas supply,and the arc temperature field,arc pressure,electric potential and current density distribution under the condition of alternating gas supply were *** with the real-time data of arc pressure measured by the water-cooled copper plate with holes,it is proved that the TIG arc has obvious dynamic characteristics under the condition of Ar-He alternating gas *** unique dynamic TIG arc acts on the 5A06 aluminum alloy weld,causing the molten pool to stir,resulting in uniform microstructure and grain refinement at the weld,and thereby improving the mechanical properties of the welded joint.

Process monitoring of the Au-S bond conversion in acetylene hydrochlorination

The continuous expansion of vinyl chloride production increases environmental pollution caused by mercury catalysts,which is an issue that urgently needs to be *** and stable catalysts should be researched to alleviate this *** this research,Thiolactic acid acts as a ligand where sulfhydryl groups form a stable complex with Au on the surface of a spherical activated carbon(SAC).An Au-thiolactic acid/SAC catalyst was designed with a Au theoretical loading of 0.5%(mass)to overcome the disadvantages of traditional Au-based catalysts,such as a low conversion rate and poor life *** ratio of Au to ligand was screened,and the activity was best when Au/S=1:*** formation of the Au-S bond was proven by FT-IR and *** longevity test of the Au1 S8/SAC catalyst was carried out at 1200 h^(-1) for 50 *** with reaction times of 0 h,5 h,10 h,20 h,and 50 h were taken to monitor the catalyst *** XPS and TPR tests proved that the Au-S bond broke as the acetylene hydrochlorination reaction proceeded,The DFT calculation proved that the Au-S bond is the active site,and the sulfur atom promotes the adsorption of C_(2)H_(2) by the catalyst.

Optimal Sales Promotion in a Supply Chain Using Consignment Contract under Stochastic Demand

Sales promotion is getting more and more prosperous in Chinese cross-border e-commerce platforms where the demand is ***,most existing literature on promotion strategies is focusing on deterministic *** this paper,we propose a game-theoretical model under multiplicative stochastic demand to investigate the pricing,inventory quantity and sales promotion strategies for a supply chain which is consisted of one cross-border distributor and one capital-constrained retailer under a consignment *** obtain the equilibrium outcomes under stochastic demand,and find that the optimal price and promotion investment depend on demand uncertainty under endogenous inventory *** exogenous unlimited inventory,the retailer prefers owing promotion right when the elasticity of price and promotion is small enough and its capital is sufficient,while the distributor always prefers to control sales *** endogenous inventory quantity,the sensitivity of demand to price is influence by the demand *** retailer prefers to decide the promotion when the price-elasticity is small,while the distributor prefers to decide the promotion under large *** the intensity of optimal sales promotion made by retailers may be stronger than that when the distributor owns the promotion right,which depends on the elasticity of price and *** importantly,it is always better for consumers when the distributor reserves the promotion right as a lower optimal retailing price is offered.

Full-frame Data Reduction Method:A Data Mining Tool to Detect the Potential Variations in Optical Photometry

A Synchronous Photometry Data Extraction(SPDE)program,performing indiscriminate monitoring of all stars appearing in the same field of view of an astronomical image,is developed by integrating several Astropy affiliated packages to make full use of time series observed by traditional small/medium aperture ground-based *** complete full-frame stellar photometry data reductions implemented for the two time series of cataclysmic variables:RX J2102.0+3359 and Paloma J0524+4244 produce 363 and 641 optimal light curves,respectively.A cross-identification with SIMBAD finds 23 known stars,of which 16 are red giant-/horizontal-branch stars,2 W UMa-type eclipsing variables,2 program stars,an X-ray source and 2 Asteroid Terrestrial-impact Last Alert System *** on the data products from the SPDE program,a follow-up light curve analysis program identifies 32 potential variable light curves,of which 18 are from the time series of RX J2102.0+3359,and 14 are from that of Paloma J0524+*** are preliminarily separated into periodic,transient,and peculiar *** querying for the 58 VizieR online data catalogs,their physical parameters and multi-band brightness spanning X-ray to radio are compiled for future analysis.

PRICING STRATEGIES IN A SUPPLY CHAIN WITH MULTI-MANUFACTURER AND A COMMON RETAILER UNDER ONLINE REVIEWS

Online reviews play an important role in consumer purchasing behavior when shopping online, and in turn affect pricing strategies of sellers. We consider a supply chain consisting of three competitive manufacturers and an e-tailer, which sells multiple substitutable products procured from different manufacturers. Considering reviews in terms of quality information and fit information, we study the effect of online reviews on consumers＇ purchasing decisions as well as pricing strategies of manufacturers and the e-retailer. Through modelling customer choice with online reviews based on neo-Hoteling model and solving Stackelberg game composed by manufacturers and e-tailer, we obtain the optimal equilibrium prices of manufacturers and the e-tailer. And we distinguish two kinds of effect from online reviews： effect of the opinion bias in product quality and effect of the match informativeness. Furthermore, compared to the case without online reviews, we find online reviews improve a manufacturer＇s optimal wholesale price, profit as well as the retailer＇s optimal retailing price, when the adjusted quality revealed by reviews is high; however, online reviews can improve the e-tailer＇s optimal profit, only when the standard deviation of products＇ quality revealed by reviews is large enough. Last, we get that the increase of competing manufacturers＇ quantity would change the dominance between two kinds of effect from reviews on supply chain members.

Lanthanum incorporated in MCM-41 and its application as a support for a stable Ni-based methanation catalyst

Herein, lanthanum was incorporated via hydrothermal synthesis into a MCM-41 framework structure with La/Si molar ratios from 0.01 to 0.1. Samples of NiO supported on LaMCM-41 were prepared using the impregnation method. The catalyst performance was evaluated using a fixed bed CO methanation reactor. A Ni/LaMCM-41 catalyst with La/Si = 0.1 shows the best catalytic performance with a CO conversion of almost 100% and a CH4 selectivity of 89.5% at 250 ℃ under a pressure of 1.5 MPa and at an airspeed of 36,000 mL/（g·h）. Compared with Ni-La/MCM-41（La/Si = 0.1） and Ni/MCM-41 prepared via the impregnation method, Ni/LaMCM-41（La/Si = 0.1） shows a higher CO conversion and CH4 *** a 100 h stability test, the Ni/LaMCM-41（La/Si = 0.1） catalyst shows excellent stability; furthermore, the CO conversion is always greater than 98.0%, which is significantly better than the results for Ni/*** experimentally demonstrate that elemental La enters the framework of MCM-41. The Ni/LaMCM-41 catalyst performs well because the La reduces the average particle size of the NiO particles and enhances the interaction between NiO and MCM-41; moreover, the introduction of La significantly inhibits the sintering of the catalyst and the formation of carbon deposits.
