Paternal mixed exposure to nicotine/ethanol/caffeine damaged cartilage quality in paternal/offspring rats and its differential glucocorticoid regulation mechanisms

Nicotine， ethanol， and caffeine are the most common exogenous substances in the men’s living environment， but their effects on the cartilage quality in the father and offspring have not been reported. According to the average daily intake of adult men， we constructed a male rat model of paternal mixed exposure (PME) to low-dose nicotine (0.1 mg/kg.d)， ethanol (0.5 g/kg.d)， and caffeine (7.5 mg/kg.d) for 8 weeks. Then， the male rats mated with normal female rats to obtain offspring. The results showed that PME reduced the cartilage quality of paternal and offspring rats. Among them， the paternal cartilage was damaged by enhancing matrix degradation， while the offspring cartilage was damaged by reducing matrix synthesis. The cartilage damage in male offspring rats was more evident than in female offspring. It was further confirmed that differential GC regulation mechanisms were the main reasons for the intergenerational differential damage of paternal/offspring cartilage quality caused by PME. In addition， the sex hormone receptor AR and ERβ mediated the sex difference of PME-induced fetal cartilage dysplasia by affecting the binding degree of GR/P300. This study provided a theoretical and experimental basis for guiding male healthy lifestyle and exploring early prevention and treatment strategies for paternal diseases.

using data from the China Education Panel Survey,this study examines the impact of homework time on academic performance among Chinese junior high school *** findings show that seventh and ninth graders spend 2.28 and 2.67 hours daily on homework,respectively,exceeding the 90-minute limit stipulated by the"double reduction"*** time significantly varies by gender,grade,and *** relationship between homework time and the academic performance of junior high school students follows an inverted u-shaped *** Least Squares analysis indicates that the optimal homework time for maximizing academic performance is less than 0.96 hours per *** the threshold,the positive effect diminishes,and when homework time exceeds 4.07 hours,it negatively impacts academic *** effect analysis shows that excessive homework time leads to negative emotions,such as boredom,unhappiness,frustration,and sadness while depriving students of necessary sleep for healthy *** is concluded that optimal daily homework time is about 1 hour,aligning with the"double reduction"policy guidelines.

The crystallization behavior of silica-filled polydimethylsiloxane （PDMS） was investigated in detail by1H solid-state nuclear magnetic resonance （1H SS-NMR） in combination with synchrotron radiation wide-angle X-ray scattering （WAXS）,and temperature-modulated differential scanning calorimetry （TMDSC） *** neat PDMS,no apparent difference is observed for the crystallinity characterized by1H SS-NMR and WAXS at low-temperature ***,upon filler addition,a 15%-35%lower difference in crystallinity is observed measured by1H SS-NMR compared to *** origin of such mismatch was explored through multi-component structural,dynamics,and chain-order analysis of PDMS samples with different filler *** 1D integrated WAXS results of PDMS with different filler fractions at different temperatures show that the packing structure as well as crystal size basically remain unchanged,but as the filler fraction increases from 0 phr to 60 phr,the rigid component’s dynamics order parameter Srobtained by1H SS-NMR decreases from 0.70 to *** filler fraction-dependent crystallinity calculated based on Srwas compared with experimental values,revealing a behavior of decreasing order in the crystalline *** with the results of accelerated chain dynamics in crystalline region as reflected by T2values,the molecular origin is attributed to the formation of CONDIS crystals,whose conformational order is lost but the position and orientation orders are *** hypothesis is further supported by the TMDSC results,where,as the filler fraction increases from 0 phr to 60 phr,the melting range widens from 8.77 K to 14.56 K,representing a growth of166%.In addition to previous reports related to the condition for forming CONDIS mesophase,i.e.,temperature,pressure,and stretching,the nano-sized filler could also introduce the local conformational disorder for chain packing.

●AIM:To evaluate the role of semaphorin 7A(Sema7A)and its associated regulatory mechanisms in modulating the barrier function of cultured human corneal epithelial cells(HCEs).●METHODS:Barrier models of HCEs were treated with recombinant human Sema7A at concentrations of 0,125,250,or 500 ng/mL for 24,48,or 72h in *** electrical resistance(TEER)as well as Dextran-fluorescein isothiocyanate(FITC)permeability assays were conducted to assess barrier *** quantify tight junctions(TJs)such as occludin and zonula occludens-1(ZO-1)at the mRNA level,reverse transcriptionpolymerase chain reaction(RT-PCR)analysis was *** was used to examine the activity of the nuclear factor-kappa B(NF-κB)signaling pathway and the production of TJs *** analyses were employed to localize the ***-linked immunosorbent assay(ELISA)and RT-PCR were utilized to observe changes in interleukin(IL)-1β*** investigate the role of NF-κB signaling activation and IL^(-1)βin Sema7A’s anti-barrier mechanism,we employed 0.1μmol/L IκB kinase 2(IKK2)inhibitor IV or 500 ng/mL IL^(-1)receptor(IL-1R)antagonist.●RESuLTS:Treatment with Sema7A resulted in decreased TEER and increased permeability of Dextran-FITC in HCEs through down-regulating mRNA and protein levels of TJs in a time-and dose-dependent manner,as well as altering the localization of ***,Sema7A stimulated the activation of inhibitor of kappa B alpha(IκBα)and expression of IL-1β.The anti-barrier function of Sema7A was significantly suppressed by treatment with IKK2 inhibitor IV or IL-1R antagonists.●CONCLuSION:Sema7A disrupts barrier function through its influence on NF-κB-mediated expression of TJ proteins,as well as the expression of IL-1β.These findings suggest that Sema7A could be a potential therapeutic target for the diseases in corneal epithelium.

The China Spallation Neutron Source(CSNS) upgrade project(CSNS-Ⅱ) aims to enhance the beam power from 100 to 500 kW. A dual-harmonic accelerating method has been adopted to alleviate the stronger space-charge effect in rapid-cycling synchrotrons owing to the increased beam intensity. To satisfy the requirements of dual-harmonic acceleration, a new radiofrequency(RF) system based on a magnetic alloy-loaded cavity is proposed. This paper presents design considerations and experimental results regarding the performance evaluation of the proposed RF system through high-power tests and beam commissioning. The test results demonstrate that the RF system satisfies the desired specifications and affords significant benefits for CSNS-Ⅱ.

Tropical cloud clusters (TCCs) can potentially develop into tropical cyclones (TCs),leading to significant casualties and economic *** prediction of tropical cyclogenesis (TCG) is crucial for early *** traditional deep learning methods applied to TCG prediction rely on predictors from a single time point,neglect the ocean-atmosphere interactions,and exhibit low model *** study proposes the Tropical Cyclogenesis Prediction-Net (TCGP-Net) based on the Swin Transformer,which leverages convolutional operations and attention mechanisms to encode spatiotemporal features and capture the temporal evolution of *** model incorporates the coupled ocean-atmosphere interactions,including multiple variables such as sea surface ***,causal inference and integrated gradients are employed to validate the effectiveness of the predictors and provide an interpretability analysis of the model’s decision-making *** model is trained using GridSat satellite data and ERA5 reanalysis *** results demonstrate that TCGP-Net achieves high accuracy and stability,with a detection rate of 97.9%and a false alarm rate of 2.2%for predicting TCG 24 hours in advance,significantly outperforming existing *** indicates that TCGP-Net is a reliable tool for tropical cyclogenesis prediction.
