Microstructural Instability of an Experimental Nickel-Based Single-Crystal Superalloy

Microstructural instability with the precipitation of topologically close-packed(TCP)phases of an experimental nickel-based single-crystal superalloy has been investigated.A significant amount of σ phases are distinguished in the interdendritic region of the as-cast samples after thermal exposure at 900℃ for 1000 h.Theσphases are preferentially precipitated at the periphery of coarse γ/γ′eutectic,and their morphological evolution from needles to granules is observed.Microstructural analysis suggests that the local segregation of Cr and Ti at the periphery of coarse γ/γ′eutectic accounts for the formation ofσphases in the as-cast samples.After heat treatment with low solution temperature and short holding time,the dendritic segregation of alloying elements(i.e.,W,Re,Ti and Ta)and the volume fraction of γ′phase in the interdendritic region are similar to that of the as-cast samples.However,no TCP phases are present in the interdendritic region of the heat-treated samples after thermal exposure,which is primarily ascribed to the elimination of local segregation of Cr and Ti near the coarse γ/γ′eutectic.Moreover,small quantities ofμphases are precipitated in the secondary dendrite arm near the interdendritic region after thermal exposure,due to the increased volume fraction ofγ′phase and the concomitant enrichment of W and Re in theγmatrix.

Background:To investigate the mental health status and its influencing factors of medical staff in Karamay City who assisted Ili during the outbreak of COVID-19,so as to provide a reliable basis for timely and effective psychological intervention.Methods:Applied questionnaire survey and used the“questionnaire star”platform to investigate the mental health status of medical staff assisting Ili by the General information,the PHQ-9,the GAD-7 and the SCL-90.And the relevant influencing factors were analyzed by univariate analysis and multiple linear regression analysis.Results:Factors such as whether they will support again,whether they need to provide psychological staff,and whether they have a sense of guilt for their families have a significant impact on the PHQ-9 of medical staff(p<0.05);Whether they will support again,whether they need psychological staff,and whether they feel bored and lonely during the closed loop management have a significant impact on the GAD-7 of medical staff(p<0.05);The SCL-90 was compared with the norm,and the scores of somatization,interpersonal sensitivity and terror factors of medical personnel were statistically significant(p<0.05),scores in other aspects(diet sleep)are obvious(1.63±0.72).Conclusion:During the period of COVID-19,the medical staff in Karamay City who assisted Ili had anxiety and depression,and their mental health was not optimistic,more attention and the necessary intervention should be given.

Enterovirus 71(EV71), a major causative agent of hand, foot, and mouth disease, has caused periodic infection outbreaks in children in the Asia–Pacific region. In order to describe the largely unknown life cycle of EV71, the molecular basis of its virus-host interactions must first be determined. The 50 untranslated region of EV71 contains a cloverleaf-like structure and internal ribosomal entry site(IRES), which play an important role in transcription and translation of viral protein. We found that polypyrimidine tract-binding protein 1(PTB) bound to the IRES of EV71. RNA recognition motifs 1 and 2 of PTB were responsible for its binding to the EV71 IRES. Moreover, PTB protein was shuttled from nucleus to cytoplasm after EV71 infection. Additionally, IRES activity and viral protein production were inhibited by PTB knockdown. These results suggest that PTB interacts with the EV71 IRES, and positively regulates viral protein translation.

Cytoplasmic poly（A） binding protein 4 （PABPC4） is an RNA-processing protein that plays an important role in the regulation of gene expression. The aim of this study was to investigate the expression pattern and identify the potential clinical significance of PABPC4 in colorectal cancer. Immunohistochemical analysis revealed that 26.7% （27/101 patients） of primary colorectal tumors and 60.5% （23/38 patients） of corresponding adjacent, normal tissues showed high cytoplasmic expression of PABPC4, whereas expression was absent in 98% （43/44 patients） of distant, normal tissues. Using Kaplan--Meier analysis, we observed that the expression of PABPC4 was significantly correlated with disease-free survival and overall survival in patients with stage II and stage III colorectal cancer （P = 0.022 and P = 0.020, respectively）. PABPC4 expression was positively associated with survival outcome, and may have predictive value in the prognosis of patients with colorectal cancer. Taken together, our findings indicate that PABPC4 may play a role in the pathogenesis of colorectal cancer.

Genes encoding proteins with PHD (plant homeodomain) finger motif (C4HC3) are highly conserved from Arabidopsis to Homo sapiens. One of the major functions of these genes is regulating the expression of homeotic genes during the stage of embryonic development. They play a role in cell-cycle and cell differentiation and seem to be related with some human malignant diseases, such as leukemia. A human placenta cDNA library has been screened with cDNA probe amplified by PCR. The PGR primers have been designed according to the M96 (a mouse gene encoding a protein with PHD domain) homologous data in dbEST.A 2. 1 kb insert fragment in a positive cDNA clone has been isolated and sequenced. This new full-length cDNA is named PHF2 (GenBank Acc: AF052205). The putative protein composed of 567 aa has two typical PHD fingers at its N-terminus. 粗糙单裂隙及其周围岩石基质是组成复杂岩体裂隙网络的基本单元,了解其中发生的流体流动行为具有重要意义。通过直接求解Navier-Stokes(N-S)方程开展三维粗糙单裂隙和裂隙-孔隙双重介质模型的渗流模拟工作,研究了裂隙表面粗糙度、匹配性和基质渗透性对粗糙单裂隙中发生的非达西流动的影响规律。结果表明:Forchheimer方程可以准确描述双重介质模型中粗糙单裂隙流体流量与压力梯度间的非线性关系;表面粗糙度和不匹配性的增大会加剧裂隙内部开度分布的不均匀性,增大裂隙的流动阻力而促进非达西流动的发生;可渗透基质会使得裂隙过流能力增强,通过粗糙单裂隙的流量最多可增大约14%,这会抑制非达西流动的发生;基质渗透性对裂隙过流能力增长的影响程度与表面粗糙度和不匹配性呈正相关。

RNA-binding Protein UNR Promotes Glioma Cell Migration and Regulates the Expression of Ribosomal Protein L9

Objective To investigate the role of RNA binding protein—upstream-of-N-Ras(UNR)in the development of glioma and its molecular mechanism.Methods First,bioinformatics analysis of CGGA database was performed to detect UNR expression level and prognosis of patients with glioma.Western blot and real-time PCR were used to detect UNR expression level in glioma cell lines and tissues.Next,UNR siRNAs were transfected in glioma cells,and MTS assay and scratch wound-healing assay were used to detect changes in cell proliferation and migration.Then,the candidate UNR target mRNAs were identified by analyzing the sequencing data of UNR iCLIP-seq,RNA sequencing and ribosome profiling databases of human melanoma.RNA immunoprecipitation and biotin pull-down assays were used to identify the UNR target mRNAs in glioma cells.Finally,western blot was used to detect the effect of UNR knockdown on ribosomal protein L9(RPL9)and RPL9 protein expression level in glioma cell lines.RPL9 siRNA was transfected in A172 and T98G and the expression of vimentin in the cells was detected with western blot.Results Bioinformatics analysis showed that UNR mRNA expression level was significantly higher in highgrade glioma[GradeⅡ(n=126),GradeⅢ(n=51),GradeⅣ(n=128),Pbinded the 3’untranslated region(UTR)of PTEN and RPL9 mRNAs.RPL9 protein was significantly highly expressed in most glioma cell lines(n=9)and knockdown of UNR resulted in a downregulation of RPL9 protein expression.

Based on the similarities in the conformation of VS008（N-（4-methylphenyl）-3-（tert-butyl）-1-（phenylmethyl）-1H-pyrazole-5-carboxamide） and BYIO6830（N＇-（3,5-dimethylbenzoyl）-N＇-tert-butyl-5-methyl-2,3-dihydro-l,4-benzodioxine-6-carbohydrazide） bound to the active site of the EcR subunit of the ecdysone receptor（EcR）-ultraspiracle protein（USP） heterodimeric receptor,a series of new pyrazole amide derivatives were designed and synthesized.Their structures were confirmed by IR,~1H NMR,^（13）C NMR and elemental analysis.Results from a preliminary bioassay revealed that two of the pyrazole derivatives exhibited promising insecticidal activity.Specifically,compounds 6e and 6i exhibited good activity against Helicoverpa armigera（cotton bollworm） at low concentration.Symptoms displayed by tebufenozide-treated H.armigera were identical with those displayed by its treated counterpart.6i showed the same poisoning symptoms as those of tebufenozide.In addition,results from molecular docking result indicated that the binding modes of 6e and 6i at the active site of the EcR subunit of the heterodimeric receptor were similar to that of the bound tebufenozide.

Cloning and expression analysis of MBLL cDNA

The mbl (musdeblind) gene of Drosophila encodes a nuclear protein which contains two Cys3His motifs. The mutation of mbl gene will disturb the differentiation of ali the Drosophila’s photoreceptors. Primers have been designed according to human EST086139, which is highly homologous to mbl gene. Human fetal brain cDNA library has been screened and a novel cDNA clone has been obtained. The 2595 bp cDNA, designated MBLL (muscleblind-like), contains an open reading frame which encodes 255 amino acids and has 4 Cys3His motifs (GenBank Acc. AF061261). The amino acids seguence shares high homology to Drosophila’s mbl. The Northern blot and RNA dot blot hybridization of 43 human adult tissues and 7 fetal tissues show that MBLL is a widely expressed gene, but the expression amounts differ in these tissues.
