Web3,also known as Web 3.0,has recently been attracting increasing attention from industry and academia.Leveraging the potential of blockchain technologies,Web3 has emerged as a pivotal foundation in the realm of metaverse development,which is considered by many as the next-generation Internet.Specifically,Web3 technologies such as smart contracts and protocols like non-fungible tokens(NFTs)have supported the immersive and content-rich experience of current Web3 metaverse projects.

Plastic offers a new niche for microorganisms,the plastisphere.The everincreasing emission of plastic waste makes it critical to understand the microbial ecology of the plastisphere and associated effects.Here,we present a global fingerprint of the plastisphere,analyzing samples collected from freshwater,seawater,and terrestrial ecosystems.The plastisphere assembles a distinct microbial community that has a clearly higher heterogeneity and a more deterministically dominated assembly compared to natural habitats.New coexistence patterns—loose and fragile networks with mostly specialist linkages among microorganisms that are rarely found in natural habitats—are seen in the plastisphere.Plastisphere microbiomes generally have a great potential to metabolize organic compounds,which could accelerate carbon turnover.Microorganisms involved in the nitrogen cycle are also altered in the plastisphere,especially in freshwater plastispheres,where a high abundance of denitrifiers may increase the release of nitrite(aquatic toxicant)and nitrous oxide(greenhouse gas).Enrichment of animal,plant,and human pathogens means that the plastisphere could become an increasingly mobile reservoir of harmful microorganisms.Our findings highlight that if the trajectory of plastic emissions is not reversed,the expanding plastisphere could pose critical planetary health challenges.

The SARS-coV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)expression.This study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOc)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOcD614G regardless of host gender,age and presence of pneumonia.Remarkably,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and non-VOc-D614G.The presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUc¼0.91,95%cI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual patient.Further potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Objective To investigate the in vivo immunomodulatory and anti-tumor mechanisms of the combined treatment of novel Four-Herb formula(4HF)and doxorubicin in triple-negative breast cancer(TNBc).Methods Murine-derived triple-negative mammary carcinoma cell line,4T1 cells,was cultured and inoculated into mouse mammary glands.Sixty-six mice were randomly assigned into 6 groups(n=11 in ench):naive,control,LD 4HF(low dose 4HF),HD 4HF(high dose 4HF),LD 4HF+D(low dose and doxorubicin),and D(doxorubicin).Apart from the naive group,each mouse received subcutaneous inoculation with 5×10^(5)4T1 cells resuspended in 100µL of normal saline in the mammary fat pads.Starting from the day of tumor cell inoculation,tumors were grown for 6 days.The LD and HD groups received daily oral gavage of 658 and 2,630 mg/kg 4HF,respectively.The LD 4HF+D group received daily oral gavage of 658 mg/kg 4HF and weekly intraperitoneal injection of doxorubicin(5 mg/kg).The D group received weekly intraperitoneal injections of doxorubicin(5 mg/kg).The treatment naive mice received daily oral gavage of 0.2 mL double distilled water and 0.1 mL normal saline via intraperitoneal injection once a week.The control group received daily oral gavage of 0.2 mL double-distilled water.The treatment period was 30 days.At the end of treatment,mice organs were harvested to analyze immunological activities via immunophenotyping,gene and multiplex analysis,histological staining,and gut microbiota analysis.Results Mice treated with the combination of 4HF and doxorubicin resulted in significantly reduced tumor and spleen burdens(Pcyte landscape,and manipulated gut microbiota to favor the anti-tumor immunological activities.Moreover,immunosuppressive genes,cytokines,and chemokines such as c-c motif chemokine 2 and interleukin-10 of tumors were significantly downregulated(Pcin combination treatment demonstrated synergetic activities and was most effective in activating the anti-tumor immune response(P<

Background Optimising the immunogenicity of cOVID-19 vaccines to improve their protection against disease is necessary.Fractional dosing by intradermal(ID)administration has been shown to be equally immunogenic as intramuscular(IM)administration for several vaccines,but the immunogenicity of ID inactivated whole severe acute respiratory syndrome coronavirus 2(SARS-coV-2)at the full dose is unknown.This study(NcT04800133)investigated the superiority of antibody and T-cell responses of full-dose coronaVac by ID over IM administration in adolescents.Methods Participants aged 11–17 years received two doses of IM or ID vaccine,followed by the 3rd dose 13–42 days later.Humoral and cellular immunogenicity outcomes were measured post-dose 2(IM-cc versus ID-cc)and post-dose 3(IMccc versus ID-ccc).Doses 2 and 3 were administered to 173 and 104 adolescents,respectively.Results Spike protein(S)immunoglobulin G(IgG),S-receptor-binding domain(RBD)IgG,S IgG Fcγreceptor IIIa(FcγRIIIa)-binding,SNM[sum of individual(S),nucleocapsid protein(N),and membrane protein(M)peptide pool]-specific interleukin-2(IL-2)+cD4+,SNM-specific IL-2+cD8+,S-specific IL-2+cD8+,N-specific IL-2+cD4+,N-specific IL-2+cD8+and M-specific IL-2+cD4+responses fulfilled the superior and non-inferior criteria for ID-cc compared to IM-cc,whereas IgG avidity was inferior.For ID-ccc,S-RBD IgG,surrogate virus neutralisation test,90%plaque reduction neutralisation titre(PRNT90),PRNT50,S IgG avidity,S IgG FcγRIIIa-binding,M-specific IL-2+cD4+,interferon-γ+cD8+and IL-2+cD8+responses were superior and non-inferior to IM-ccc.The estimated vaccine efficacies were 49%,52%,66%and 79%for IM-cc,ID-cc,IM-ccc and ID-ccc,respectively.The ID groups reported more local,mild adverse reactions.conclusion This is the first study to demonstrate superior antibody and M-specific T-cell responses by ID inactivated SARScoV-2 vaccination and serves as the basis for future research to improve the immunogenicity of inactivated vaccines.

A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATcc 16872.Heterologous production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 VrcB.Intriguingly,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic window.Our study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

As the most common internal modification of mRNA,Ne-methyladenosine(m^(6)A)and its regulators modulate gene expression and play critical roles in various biological and patholog-ical processes including tumorigenesis.It was reported previously that m^(6)A methyltransferase(writer),methyltransferase-like 3(METTL3)adds m^(6)A in primary microRNAs(pri-miRNAs)and fa-cilitates its processing into precursor miRNAs(pre-miRNAs).However,it is unknown whether m^(6)A modification also plays a role in the maturation process of pre-miRNAs and(if so)whether such a function contributes to tumorigenesis.Here,we found that YTHDF2 is aberrantly overexpressed in acute myeloid leukemia(AML)patients,especially in relapsed patients,and plays an onco-genic role in AML.Moreover,YTHDF2 promotes expression of miR-126-3p(also known as miR-126,as it is the main product of precursor miR-126(pre-miR-126)),a miRNA that was reported as an oncomiRNA in AML,through facilitating the processing of pre-miR-126 into mature miR-126.Mechanistically,YTHDF2 recognizes m^(6)A modification in pre-miR-126 and recruits AGO2,a regulator of pre-miRNA processing,to promote the maturation of pre-miR-126.YTHDF2 posi-tively and negatively correlates with miR-126 and miR-126's downstream target genes,respec-tively,in AML patients,and forced expression of miR-126 could largely rescue YTHDF2/Ythdf2 depletion-mediated suppression on AML cell growth/proliferation and leukemogenesis,indi-cating that miR-126 is a functionally important target of YTHDF2 in AML.Overall,our studies not only reveal a previously unappreciated YTHDF2/miR-126 axis in AML and highlight the ther-apeutic potential of targeting this axis for AML treatment,but also suggest that m^(6)A plays a role in pre-miRNA processing that contributes to tumorigenesis.

Mechanistic study of vacuum UV catalytic oxidation for toluene degradation over ceO_(2) nanorods

The present study specifically investigates vacuum ultraviolet (VUV) catalytic oxidation for toluene degradation over ceO_(2) nanorods.Synergetic effects of ultraviolet photocatalytic oxidation (UV-PcO) and ozone catalytic oxidation (OZcO) were manifested in the results of toluene removal and cOx generation,while the combination of UV-PcO andOZcO (UV-OZcO) did not lead to improvement of mineralization.All the processes contribute to ozone decomposition,but no obvious synergetic effects of the different processes can be observed.Intermediate analysis results indicated that more toluene was oxidized into by-products,such as benzyl alcohol and benzaldehyde,by UV-OZcO rather than forming cOx.Both hydroxyl radical (·OH) and superoxide radical (·O_(2)^(-)) were found in all the processes of the VUV-PcO-OZcO system(combination of VUV photolysis,UV-PcO,OZcO and UV-OZcO processes).In the UV-OZcO process,the formation of hydroxyl radical was promoted,while that of superoxide radical was impeded,resulting in lower mineralization level of toluene.The mechanistic study of toluene degradation over ceO_(2) nanorods in the VUV-PcO-OZcO system revealed that with the formation of·O_(2)^(-)and·OH,toluene is first oxidized to intermediates,followed by further ring–opening reaction and,finally,degradation into cO_(2) and H_(2)O.ceO_(2) nanorods function as both ozonation catalyst and photocatalyst,and the redox pair of ce^(3+)and ce^(4+)are interconvertible and can keep a balance.

colloidal solution combustion synthesis(cScS)is a simple and easy method for mass-production of crystalline nanomaterials with tunable pore structure.In this work,mesoporous Mn/ceO_(2) catalysts were fabricated via cScS method coupled with a dip-coating process and used for photocatalytic oxidation(PcO)of toluene.Under vacuum ultraviolet(VUV)irradiation,a high toluene removal efficiency of about 92%was achieved with a toluene reaction rate of about 118μmol/g/h in a continuous flow reactor.A possible degradation pathway was proposed based on the analysis of intermediates by Fourier transform infrared photoluminescence spectra(FTIR)and Gc-Mass.Hydrogen temperature-programmed reduction(H_(2)-TPR),Brunauer-Emmett-Teller(BET)surface areas,photoluminescence spectra(PL)spectra and X-ray photoelectron spectroscopy(XPS)were carried out to analyze physical and chemical properties of the catalysts.compared with Mn_(x)ce_(1-x)O_(2) catalysts synthesized by one step cScS method,Mn/ceO_(2) has a higher photocatalytic activity,which is attributed to the presence of higher contents of ce^(3+),Mn^(2+)and Mn^(3+)species.The presence of higher contents of these species plays a key role in the activity enhancement of toluene oxidation and ozone decomposition.This method is facile,efficient and scalable,and it may become a promising industrial application technology for catalyst synthesis in the near future.
