Although nowadays there are methods for determining the Minimal Bactericidal Concentration and Minimal Fungicidal Concentration, it is indispensable that?the establishment of innovative methodologies?could be more practical and cheaper. The new methodology Flash microbiocide is an assay in which one aliquot from 96 well plate of Minimal Inhibitory Concentration test is transferred to another plate containing different culture medium. The correspondence with the reference methods described in the National Committee for Clinical Laboratory Standards (NCCLS-CLSI) document M26-A was achieved, denoting the efficiency of this fast and simple method.

Roots of Zeyheriamontana, a species native to the savanna (Cerrado) region of central Brazil, produce lapachol, a naphthoquinone with anticancer activity. Lapachol is also the precursor of β-lapachone, a novel drug candidate for preventive and adjuvant cancer therapies. The leaves of Z. montana are a renewable source of ursolic acid and oleanoic acid, compounds known for their anticancer, antioxidant and antimicrobial properties. The potential prophylactic use of β-lapachone, as well as the medicinal properties of ursolic acid, highlights the importance of this study on ***’s germplasm conservation. Multiple shoots were induced on Woody Plant media with supplemented 0.1 mg·L-1 of thidiazuron (TDZ). Rooting was promoted on half strength WP (Woody Plant media containing 1.0 mg·L-1 of Indolbutiric acid-IBA). Plantlet acclimatization to ex-vitro condition was done at a 70% success rate using different substrates. It was possible to store ***’s elite germplasm using in vitro cultures of media containing 2% sucrose plus 4% sorbitol for six months without subcultures.

Screening Test for Antibiotics in Medicinal Plants(STAMP):Using Powdered Plant Materials Instead of Extracts

Plants are a rich source of antibiotics, but screening all the existing plant species for biological activity using current methods can be time and resource consuming. The present study is to investigate whether powdered plant materials would perform as well as plant extracts in the screening of plants with antimicrobial activity. In the new method proposed (STAMP), we compared in vitro antimicrobial activity of powdered plant materials from 12 species against bacteria and fungi. We confirmed these results with their corresponding aqueous (wet) and hydro-alcoholic extracts and one species testing the antimicrobial activity of two isolated compounds. Compared with hydro-alcoholic extracts, screening using the powdered plant materials correctly identified the majority of the species with antimicrobial activity against Candida albicans (sensitivity 91%, specificity 86%), C. parapsilosis (sensitivity 100%, specificity 67%), and Staphylococcus aureus (sensitivity 64%, specificity 86%). For bacteria, the method performed better in a pH of 9.0. The antimicrobial activity of two compounds isolated from one species (maytenin and netzahualcoyone) confirmed the results. In conclusion, the use of powdered plant materials for screening plants with antimicrobial properties is a cheap, widely available, technically easy, time sparing, reproducible, and sensitive method and can significantly shorten the time and money spent during drug development.

Species of the genus Erythrina are frequently used as ornamental plants and in projects to restore degraded areas. Also, extracts from its shoots and stems are popularly used as a herbal medicine with validated activity on the central nervous system. The objective of this study was to evaluate E. mulungu and E. velutina seed germination and seedling development potential aiming commercial scale production of those species. Seeds stored for one or twelve months at 10°C ± 2°C were sown in sand, soil or Plantmax? substrates and evaluated for germination and seedling development. Subsequently, seedlings sprouted in Plantmax? were transferred to polyethylene bags and kept in greenhouse, under direct sunlight for plant development (plant height, stem diameter and root length pivoting) evaluations. Four-month-old plantlets were transplanted to the field and after a period of one year the collar diameter and shoot height (of each plant were measured. Seed germination rates of both species grown in soil and in Plantmax? were significantly high (over 80%). Storing seeds for 1 month did not inhibit germination. However, seeds stored for twelve months had germination reduced by more than a quarter. The development of plants in greenhouse and in the field was satisfactory, indicating the viability of producing E. mulungu and E. velutina on a commercial scale, in order to meet the expanding market demand for herbal medicines.

Phenolic Compounds Influence Seed Dormancy of Palicourea rigida H.B.K.(Rubiaceae),a Medicinal Plant of the Brazilian Savannah

Palicourea rigida H.B.K. (Rubiaceae), a medicinal species commonly known as douradinha, has wide distribution across ecosystems in Central and South America. This species exhibits seed dormancy delaying germination until optimal conditions for seedling growth and development are in place. While dormancy ensures species survival, it also presents a technical problem for developing P. rigida’s plant production program. Thus, the objective of this study was to investigate if secondary metabolites present in seeds influence the seed dormancy of P. rigida. Mature fruits were harvested from the native habitat, in the savanna region of the State of Minas Gerais during February 2009, 2010 and 2011. The content of phenolic compounds in the seed of P. rigida was measured, and the allelopathic effects were assessed using the germination of lettuces as model to detect phytotoxicity. The P. rigida seeds geminated at rates varying between 7% and 31% with a Seed Germination Index (SGI) of 0.09. Data suggest that the phenolic compounds present in the seeds may be responsible for seed dormancy.
