crohn's disease environmental factors in the developing world: A case-control study in a statewide catchment area in Brazil

To identify environmental risk factors associated with the development of crohn’s disease (cD) in order to re-assess the hygiene hypothesis. METHODSA hospital-based, case-control study was carried out with cD patients (n = 145) and controls (n = 163) representing a socioeconomically diverse statewide catchment area in Brazil. controls were recruited from caregivers of patients seen in different outpatient clinics at the same hospital. A multi-item survey with 94 questions regarding family history of cD, perinatal and childhood circumstances, living conditions, tobacco use and familial socioeconomic status was carried out by interviewers. RESULTSOn the univariate analysis, predictive variables for cD included being male, under age of 40, a high education level, urban dweller, smaller family size, exposure to enteric pathogens and user of treated water (P cantly associated with cD were male gender (OR = 2.09), under age 40 (OR = 3.10), white (OR = 2.32), from a small family in childhood (OR = 2.34) and adulthood (OR = 3.02), absence of viral infections in childhood (OR = 2.23), exposure to enteric pathogens (OR = 2.41), having had an appendectomy (OR = 2.47) and prior or current smoker (OR = 2.83/1.12). cONcLUSIONMost variables supporting the “hygiene hypothesis” are associated with the development of cD but are not independent predictors of the diagnosis.

Dual Targeting of a Mitochondrial Protein： The case Study of cytochrome c1

As a result of the endosymbiotic gene transfer, the majority of proteins of mitochondria and chloroplasts is encoded in the nucleus and synthesized in the cytosol as precursor molecules carrying N-terminal transit peptides for the transport into the respective target organelle. In most instances, transport takes place into either mitochondria or chloroplasts, although a few examples of dual targeting into both organelles have been described. Here, we show by a combination of three different experimental strategies that also cytochrome c1 of potato, a component of the respiratory electron transport chain, is imported not only into mitochondria, but also into plastids. In organello import experiments with isolated mitochondria and chloroplasts, which were analyzed in both single and mixed organelte assays, demonstrate that the processing products accumulating after import within the two endosymbiotic organelles are different in size. Dual targeting of cytochrome c1 is observed also in vivo, after biolistic transformation of leaf epidermal cells with suitable reporter constructions. Finally, Western analyses employing cytochrome c1-specific antiserum provide evidence that the protein accumulates in significant amounts in mitochondria and chloroplasts of both pea and spinach. The possible consequences of our findings on the relevance of the dual targeting phenomenon are discussed.

Acute-on-chronic liver failure(AcLF)is a syndrome that occurs in patients with chronic liver disease and is characterized by acute decompensation,organ failure and high short-term *** due to the lack of universal diagnostic criteria,the actual AcLF prevalence remains unclear;nevertheless,it is expected to be a highly prevalent condition *** transplantation is an effective protective measure for selected AcLF *** liver transplantation,diagnosing and treating precipitant events and providing supportive treatment for organ failures are currently the cornerstone of AcLF *** new clinical specific therapies have been researched,more studies are necessary to assess safety and ***,future AcLF management strategies must consider measures to improve access to liver transplantation because the time window for this life-saving therapy is frequently ***,an urgent and global discussion about allocation and prioritization for transplantation in critically ill AcLF patients is needed because there is evidence suggesting that the current model may not portray their waitlist *** addition,while donor organ quality is meant to be a prognostic factor in the AcLF setting,recent evidence suggests that machine perfusion of the liver may be a safe tool to improve the donor organ pool and expedite liver transplantation in this scenario.

Hepatitis c virus in the new era:Perspectives in epidemiology,prevention,diagnostics and predictors of response to therapy

Despite the great successes achieved in the fields of virology and diagnostics,several difficulties affect improvements in hepatitis c virus(HcV)infection control and eradication in the new *** HcV infections still occur,especially in some of the poorest regions of the world,where HcV is endemic and long-term sequelae have a growing economic and health *** HcV vaccine is still no available,despite years of researches and discoveries about the natural history of infection and host-virus interactions:several HcV vaccine candidates have been developed in the last years,targeting different HcV antigens or using alternative delivery systems,but viral variability and adaption ability constitute major challenges for vaccine *** new antiviral drugs for HcV therapy are in preclinical or early clinical development,but different limitations affect treatment *** predictors are important tools,as they provide some guidance for the management of therapy in patients with chronic HcV infection:in particular,the role of host genomics in HcV infection outcomes in the new era of direct-acting antivirals may evolve for new therapeutic targets,representing a chance for modulated and personalized treatment management,when also very potent therapies will be *** the present review we discuss the most recent data about HcV epidemiology,the new perspectives for the prevention of HcV infection and the most recent evidence regarding HcV diagnosis,therapy and predictors of response to it.

Tumor–stromal cross-talk modulating the therapeutic response in pancreatic cancer

Background: Pancreatic ductal adenocarcinoma(PDAc) is a highly malignant solid tumor with a dismal prognosis. The stroma component makes up to 90% of the tumor mass and is thought to be one of the main reasons for the tumor’s high chemoresistance. cancer associated fibroblasts(cAFs) have previously been identified to be the key stromal players. This is the first time we provide detailed in vitro experiments investigating tumor–stromal interactions when exposed to three well-known chemotherapeutic agents. Methods: Monocultures, indirect and direct co-cultures of two PDAc cell lines(AsPc and Panc-1) and six primary patients derived cAFs were treated with gemcitabine, nab-paclitaxel and the γ-secretaseinhibitor(GSI) DAPT. The cell viability of each component was measured with XTT. Finally, IL-6 concentrations of the supernatants were analyzed. Results: On the contrary to PDAc cell lines, cAF monocultures hardly responded to any treatment which suggested that stroma(cAFs) itself is more resistant to standard chemo-treatments than the epithelial cancer cells. Moreover, only a weak chemotherapeutic response was observed in direct co-cultures of cancer cells with cAFs. A change in the morphology of direct co-cultures was accompanied with the chemoresistance. cAFs were observed to build cage-like structures around agglomerates of tumor cells. High levels of IL-6 were also associated with a reduced response to therapy. Indirect co-cultures make the tumor–stromal interaction more complex. conclusions: cAFs are highly chemoresistant. Direct cell–cell contact and high levels of IL-6 correlate with a high chemoresistance.

High-fat diets affect male reproduction and sexual ***,we evaluated the effects of prolonged resveratrol administration on the metabolic,sperm,and testicular parameters of rats fed a cafeteria *** Wistar rats were divided at weaning into control(cv n=20)and cafeteria(cAF,n=16)*** 3 months,half of them were given daily supplementations of resveratrol(c-R,n=10;cAF-R,n=8)at a dosage of 30 mg kg-1 body mass for 2 *** were killed at 5 months of age,and blood,spermatozoa,and testes were collected for further *** were analyzed by one-way ANOVA,and Pconsidered statistically *** cAF diet promoted hyperglycemia(Pcondition(PcAF diet reduced sperm viability and motility,while resveratrol improved these parameters(Fcular morphology,the height of the seminiferous epithelium was reduced in the cAF group compared with that of the c group(P=0.0007).Spermatogenic cell proliferation was also reduced in the cAF group compared with that of the c ***,the cAF-R showed an increase in cell proliferation rate compared with that of the untreated cAF group(P=0.0024).Although it did not modify body mass,the consumption of a cAF diet promoted hyperglycemia,adverse testicular morphology remodeling,and abnormal sperm,which were attenuated by treatment with resveratrol,thus suggesting a protective effect of this antioxidant on spermatogenesis.

Having a better understanding of air pollutants in railway systems is crucial to ensure a clean public *** study measured,for the first time in Brazil,nanoparticles(NPs)and black carbon(Bc)on two groundlevel platforms and inside trains of the Metropolitan Area of Porto Alegre(MAPA).An intense sampling campaign during thirteen consecutive months was carried out and the chemical composition of NPs was examined by advanced microscopy *** results showed that highest concentrations of the pollutants occur in colder seasons and influenced by variables such as frequency of the trains and passenger ***,internal and external sources of pollution at the stations were *** predominance of NPs enriched with metals that increase oxidative stress like cd,Fe,Pb,cr,Zn,Ni,V,Hg,Sn,and Ba both on the platforms and inside trains,including Fe-minerals as hematite and magnetite,represents a critical risk to the health of passengers and employees of the *** interdisciplinary and multi-analytical study aims to provide an improved understanding of reported adverse health effects induced by railway system aerosols.
