Characterization of carotenoid biosynthetic pathway genes in the pea aphid(Acyrthosiphon pisum)revealed by heterologous complementation and RNA interference assays

Carotenoids are involved in many essential physiological functions and are produced from geranylgeranyl pyrophosphate through synthase,desaturase,and cyclase activities.In the pea aphid(Acyrthosiphon pisum),the duplication of carotenoid biosynthetic genes,including carotenoid synthases/cyclases(ApCscA-C)and desaturases(ApCdeA-D),through horizontal gene transfer from fungi has been detected,and ApCdeB has known dehydrogenation functions.However,whether other genes contribute to aphid carotenoid biosynthesis,and its specific regulatory pathway,remains unclear.In the current study,functional analyses of seven genes were performed using heterologous complementation and RNA interference assays.The bifunctional enzymes ApCscA-C were responsible for the synthase of phytoene,and ApCscC may also have a cyclase activity.ApCdeA,ApCdeC,and ApCdeD had diverse dehydrogenation functions.ApCdeA catalyzed the enzymatic conversion of phytoene to neurosporene(three-step product),ApCdeC catalyzed the enzymatic conversion of phytoene to ζ-carotene(two-step product),and ApCdeD catalyzed the enzymatic conversion of phytoene to lycopene(four-step product).Silencing of ApCscs reduced the expression levels of ApCdes,and silencing these carotenoid biosynthetic genes reduced the α-,β-,and γ-carotene levels,as well as the total carotenoid level.The results suggest that these genes were activated and led to carotenoid biosynthesis in the pea aphid.

The involvement of systemic RNA interference deficient-1-like (SIL1) in cellular dsRNA uptake in Acyrthosiphon pisum

Systemic RNA interference deficient-1-like(SIL1)is considered a core component in dsRNA uptake in some insect species.Investigation related to the potential function of SIL1 in dsRNA uptake can contribute to a further understanding of RNA interference(RNAi)mechanisms in insects and agricultural pest control.However,the role of SIL1 in dsRNA uptake in insects such as aphids remains controversial.We have thoroughly analyzed the role of SIL1 from the model aphid Acyrthosiphon pisum(ApSIL1)in cellular dsRNA to clarify its function.First,the induced expression of ApSIL1 upon dsRNA oral exposure provided a vital clue for the possible involvement of ApSIL1 in cellular dsRNA uptake.Subsequent in vivo experiments using the RNAi-of-RNAi approach for ApSIL1 supported our hypothesis that the silencing efficiencies of reporter genes were reduced after inhibition of ApSIL1 expression.The impaired biological phenotypes of aphids,including cumulative average offspring,deformities of the nymph,and mortality upon pathogen infection,were then observed in the treatment group.Thereafter,in vitro dual-luciferase reporter assay showed compelling evidence that the luciferin signal was significantly attenuated when dsluciferase or dsGFP was transferred into ApSIL1-transfected Drosophila S2 cells.These observations further confirmed that the signal of Cy3-labeled dsRNA was rapidly attenuated with time in ApSIL1-transfected Drosophila S2 cells.Overall,these findings conclusively establish that ApSIL1 is involved in dsRNA uptake in A.pisum.

A Liutex-based subgrid stress model for large-eddy simulation

The concept of vortex is crucial in both understanding and modeling of turbulence.For large eddy simulation(LES),the effect of small-scale eddies onto the large scales or the resolved flow field is modeled by subgrid stress models.Even though the rotating motions of fluids,i.e.,vortices or eddies are central in developing turbulent models,vortex identification methods are seldom used in these models.In this study,we develop a new subgrid model based on the Liutex vector,a new quantity introduced to decompose fluid motions into rigid rotation,pure shear and stretching,and thus identify vortices.The new model is then applied in a decaying homogeneous isotropic turbulence(DHIT)and a turbulent channel flow at Reynolds number Reτ=180.It is shown that the new model can predict accurate energy spectra compared with experiments in DHIT and give a well-matched velocity profile in turbulent channel flow without changing the form of the model.Future directions include improvement of the Liutex based model,for example developing anisotropic subgrid models,and its applications in various turbulent flows.

Impact of parameter fluctuations on the performance of ethanol precipitation in production of Re Du Ning Injections, based on HPLC fingerprints and principal component analysis

The present study was designed to determine the relationships between the performance of ethanol precipitation and seven process parameters in the ethanol precipitation process of Re Du Ning Injections,including concentrate density,concentrate temperature,ethanol content,flow rate and stir rate in the addition of ethanol,precipitation time,and precipitation temperature.Under the experimental and simulated production conditions,a series of precipitated resultants were prepared by changing these variables one by one,and then examined by HPLC fingerprint analyses.Different from the traditional evaluation model based on single or a few constituents,the fingerprint data of every parameter fluctuation test was processed with Principal Component Analysis(PCA)to comprehensively assess the performance of ethanol precipitation.Our results showed that concentrate density,ethanol content,and precipitation time were the most important parameters that influence the recovery of active compounds in precipitation resultants.The present study would provide some reference for pharmaceutical scientists engaged in research on pharmaceutical process optimization and help pharmaceutical enterprises adapt a scientific and reasonable cost-effective approach to ensure the batch-to-batch quality consistency of the final products.

Large-scale computational screening of metal–organic frameworks for D_(2)/H_(2) separation

Deuterium(D_(2)) is one of the important fuel sources that power nuclear fusion reactors. The existing D_(2)/H_(2) separation technologies that obtain high-purity D_(2) are cost-intensive. Recent research has shown that metal-organic frameworks(MOFs) are of good potential for D_(2)/H_(2) separation application. In this work, a high-throughput computational screening of 12020 computation-ready experimental MOFs is carried out to determine the best MOFs for hydrogen isotope separation application. Meanwhile, the detailed structure-performance correlation is systematically investigated with the aid of machine learning. The results indicate that the ideal D_(2)/H_(2) adsorption selectivity calculated based on Henry coefficient is strongly correlated with the 1/ΔAD feature descriptor;that is, inverse of the adsorbility difference of the two adsorbates. Meanwhile, the machine learning(ML) results show that the prediction accuracy of all the four ML methods is significantly improved after the addition of this feature descriptor. In addition, the ML results based on extreme gradient boosting model also revealed that the 1/ΔAD descriptor has the highest relative importance compared to other commonly-used descriptors. To further explore the effect of hydrogen isotope separation in binary mixture, 1548 MOFs with ideal adsorption selectivity greater than 1.5 are simulated at equimolar conditions. The structure-performance relationship shows that high adsorption selectivity MOFs generally have smaller pore size(0.3-0.5 nm) and lower surface area. Among the top 200 performers, the materials mainly have the sql, pcu, cds, hxl, and ins topologies.Finally, three MOFs with high D_(2)/H_(2) selectivity and good D_(2) uptake are identified as the best candidates,of all which had one-dimensional channel pore. The findings obtained in this work may be helpful for the identification of potentially promising candidates for hydrogen isotope separation.

针对单一检验质量的无拖曳卫星姿控系统传感器故障，提出一种基于扩展卡尔曼滤波器组的诊断方法。首先，建立了单一检验质量无拖曳卫星的系统模型，并进一步给出了无拖曳卫星中特有的重力参考传感器模型；基于所建立的系统模型设计了扩展卡尔曼滤波器组以估计系统状态，包括了一个总滤波器和四个并行的子滤波器，这些滤波器分别接收不同类型的传感数据；然后，通过多样本残差卡方检验的方法分析滤波器生成的估计残差，实现了传感器故障检测和初级故障隔离；最终，在估计残差的基础上，通过计算并比较广义似然函数值，能够进一步隔离出故障传感器的具体故障位置。仿真结果验证了所提方法的有效性。

Is the transsacral axial interbody fusion a candidate surgical approach for fusing both L5/S1 and L4/5？

Background Previous clinical and basic research of axial lumbar interbody fusion （AxiaLIF） all focused on the L5/S1.However,there is no data on the feasibility of this approach for the fusion of both L4/5 and L5/S1.This study aimed to explore whether transsacral axial interbody fusion is a candidate for the fusion of both L4/5 and L5/S1.Methods The subjects （n=40） underwent lumbosacral magnetic resonance imaging （MRI）.The median sagittal MRI images were analyzed and five measurement markers were defined as follows：the center of the L4/5 disc （A）,the center of the L5/S1 disc （B）,the anterior margin of the S1/2 space （C）,the sacrococcygeal junction （D）,and the coccygeal tip （E）.The measurement markers were connected each other to produce nine lines （AB,AC,AD,AE,BC,BD,BE,CD and CE） as the reference lines for surgical approaches.The distance between each reference line and the anterior and posterior margins of the L4,L5 and S1 vertebral bodies were measured to determine the safety of the respective approaches.Results Twenty subjects were capable of finding one reference line to fuse both L4/5 and L5/S1 via transsacral axial interbody fusion approach.The surgical approach reference line was AE or CE line.In the other 20 subjects,it was failed to find a reference line which met the safety criteria for fusing both L4/5 and L5/S1.Conclusions About half of subjects were capable of finding a suitable AxiaLIF reference line to fuse both L4/5 and L5/S1.In some subjects,it was difficult to find a suitable AxiaLIF reference line to fuse both L4/5 and L5/S1.

Multi-stage igneous rocks developed in the recently discovered Huoluotai Cu-(Mo)deposit provide new insights into the controversial late Mesozoic geodynamic evolution of the northern segment of the Great Xing’an Range(NSGXR).Zircon U-Pb dating suggests that the monzogranite,ore-bearing granodiorite porphyry,diorite porphyry,and granite porphyry in the deposit were emplaced at 179.5±1.6,148.9±0.9,146.1±1.3,and 142.2±1.5 Ma,respectively.The Re-Os dating of molybdenite yielded an isochron age of 146.9±2.3 Ma(MSWD=0.27).The Jurassic adakitic monzogranite and granodiorite porphyry are characterized by high SiO_(2)and Na_(2)O contents,low K_(2)O/Na_(2)O ratios,low Mg O,Cr,and Ni contents,low zirconεHf(t)values relative to depleted mantle,and relatively high Th contents.They were produced by partial melting of a subducted oceanic slab,with involvement of marine sediments in the magma source and limited interaction with mantle peridotites during magma ascent.The Late Jurassic diorite porphyry is characterized by moderate SiO_(2) contents,high Mg O,Cr,and Ni contents,and positive dominatedεHf(t)values,indicating it was produced by partial melting of a subduction-modified lithospheric mantle wedge and underwent limited crustal contamination during magma ascent.The early Early Cretaceous adakitic granite porphyry shows high SiO_(2) and K_(2)O contents and K_(2)O/Na_(2)O ratios,low Mg O,Cr,and Ni contents,enriched Sr-Nd isotopic compositions,and slightly positive zirconεHf(t)values,suggesting it was produced by partial melting of thickened mafic lower crust.The NSGXR experienced a tectonic history that involved flat-slab subduction(200-160 Ma),and tearing and collapse(150-145 Ma)of the Mongol-Okhotsk oceanic lithosphere.The period of magmatic quiescence from ca.160 to 150 Ma was a response to flat-slab subduction of the Mongol-Okhotsk oceanic lithosphere.Crustal thickening in the NSGXR(145-133 Ma)was due to the collision between the Amuria Block and the Siberian Craton.

Coronary artery perforation(CAP)remains an infrequent but potentially lifethreatening complication during percutaneous coronary intervention(PCI).Although the prevalence of CAP is about 0.2%−0.9%in relatively low risk PCI,the incidence could up to 9%in complex clinical scenarios such as severe coronary calcification,chronic total occlusions and rotational atherectomy.[1]Despite the new techniques and equipment had lower morbidity and mortality,up to 17%of acute CAP cases would evolve to tamponade and subsequent death.
