Structural control of magnetic nanoparticles for positive nuclear magnetic resonance imaging

In addition to the tens of millions of medical doses consumed annually around the world, a vast number of nuclear magnetic resonance imaging (MRI) contrast agents are being deployed in MRI research and development, offering precise diagnostic information, targeting capabilities, and analyte sensing. Superparamagnetic iron oxide nanoparticles (SPIONs) are notable among these agents, providing effective and versatile MRI applications while also being heavy-metal-free, bioconjugatable, and theranostic. We designed and implemented a novel two-pronged computational and experimental strategy to meet the demand for the efficient and rigorous development of SPION-based MRI agents. Our MATLAB-based modeling simulation and magnetic characterization revealed that extremely small maghemite SPIONs in the 1-3 nm range possess significantly reduced transversal relaxation rates (R-2) and are therefore preferred for positive (T-1-weighted) MRI. Moreover, X-ray diffraction and X-ray absorption fine structure analyses demonstrated that the diffraction pattern and radial distribution function of our SPIONs matched those of the targeted maghemite crystals. In addition, simulations of the X-ray near-edge structure spectra indicated that our synthesized SPIONs, even at 1 nm, maintained a spherical structure. Furthermore, in vitro and in vivo MRI investigations showed that our 1-nm SPIONs effectively highlighted whole-body blood vessels and major organs in mice and could be cleared through the kidney route to minimize potential post-imaging side effects. Overall, our innovative approach enabled a swift discovery of the desired SPION structure, followed by targeted synthesis, synchrotron radiation spectroscopic studies, and MRI evaluations. The efficient and rigorous development of our high-performance SPIONs can set the stage for a computational and experimental platform for the development of future MRI agents.

Vaccinations in kidney transplant recipients: Clearing the muddy waters

Vaccine preventable diseases account for a significant proportion of morbidity and mortality in transplant recipients and cause adverse outcomes to the patient and allograft. Patients should be screened for vaccination history at the time of pre-transplant evaluation and vaccinated at least four weeks prior to transplantation. For non-immune patients, dead-vaccines can be administered starting at six months post-transplant. Live attenuated vaccines are contraindicated after transplant due to concern for infectious complications from the vaccine and every effort should be made to vaccinate prior to *** transplant recipients are on life-long immunosuppression, these patients may have lower rates of serological conversion, lower mean antibody titers and waning of protective immunity over shorter period as compared to general population. Recommendations regarding booster dose in kidney transplant recipients with sub-optimal serological response are lacking. Travel plans should be part of routine post-transplant assessment and pre-travel vaccines and counseling should be provided. More studies are needed on vaccination schedules, serological response, need for booster doses and safety of live attenuated vaccines in this special population.

A Rapid and Sensitive One Step-SYBR Green Based Semi Quantitative Real Time RT-PCR for the Detection of peste des petits ruminants Virus in the Clinical Samples

A sensitive and rapid single step real time （rt） RT-PCR was standardized using one-step Brilliant SYBR Green kit for detection and semi-quantitation ofpeste des petitis ruminants virus （PPRV） using the virus RNA and matrix （M） protein gene-specific primers and compared with established conventional RT-PCR and TaqMan RT-PCR. The assay amplifies a 124 bp fragment of the PPRV M gene with Tm of 78.28 to 78.50. The assay was linear within a range of 50 ng to 0.5 fg total virus RNA with a detection limit （sensitivity） of 0.5 fg. Based on the serial dilution of the live-attenuated PPR vaccine virus, the detection limit was -0.0001 cell culture infectious dose 50% units （TCID50）. Additionally, swab materials spiked with known titre of vaccine virus were equally well detected in the assay. The standardized rt RT-PCR was easily employed for the detection of PPRV nucleic acid directly in the field and experimental clinical samples. The assay detected the PPRV nucleic acid as early as 3 day post infection （dpi） and up to 20 dpi in swab materials from the experimental samples. The assay was rapid and more sensitive than TaqMan and conventional RT-PCR in the detection of PPRV nucleic acid from the PPR suspected clinical samples of sheep and goats. Therefore, the established, simplified SYBR green rt RT-PCR is an alternative test to the already existing various diagnostic assays and could be useful for rapid clinical diagnosis with advantage in reducing risk of contamination.

Polarization Multiplexed Interrogation Technique for FBG Sensor Array

This paper proposes a polarization multiplexed interrogation technique for fiber Bragg grating （FBG） sensor array. The novelty of the proposed model is its ability to reduce interference and cross talk, thus allowing larger number of FBG sensors to be interrogated in an array. The calibration technique has been illustrated in this work for the FBG sensor array, where data from each sensor are linearly polarized and multiplexed before co-propagation, to find out the tapping points that enable identification of each sensor data uniquely. Simulation has been carried out for odd number and even number of sensors in an array. Even with interfering input, this proposed scheme can interrogate and distinctively identify each sensor data using appropriate tuning of polarization-splitter, polarization-rotator, and polarization-attenuator at the detector end during the calibration process. The significance of the proposed method is its compact size, which makes this calibration system ready to be deployed in real-time sensing applications and data acquisition from the FBG sensor array.
