Brazilian vaccinia virus strains show a classical orthopoxvirus infection course and cross-protection

Objectives:The purpose of this work was to study the infection course and cross-protection in mice after intradermal injection of Vaccinia virus(VACV) strain Western Reserve and three Brazilian VACV strains: Aracatuba,Muriae and BeAn58058 isolated from cow,human and rodent,respectively.Methods:Balb/c mice were inoculated by footpad and back scarification and daily monitored regarding lesion development and weight loss.To check cross protection after intradermal VACV inoculation,mice were subsequendy infected with different VACV strains and monitored to check lesion development.Serum neutralization assays were performed to check for the presence of antibodies against Orthopoxvirus.Results:After VACV intradermal inoculation the lesion development pattern was similar in mice infected with the different virus strains.By using the footpad scarification model,cross-protection among VACV strains was observed.Moreover,neutralizing antibodies against Orthopoxvirus were detected in sera from mice infected with all VACV strains.Conclusion:Although it was not possible to observe virulence differences among VACV strains isolated from cow,rodent and human using the murine model,this inoculation route showed to be an appropriated model to study lesions development since it mimics natural infections by VACV in nature.

A remote sensing approach to estimate the load bearing capacity of soil

Preconsolidation pressure(σp)of soil can be considered as an indicator of the Load Bearing Capacity(LBC),which is the tolerated surface pressure before compaction,often caused by the traffic of agricultural machinery.In this pioneering study,a remote sensing approach was introduced to estimate LBC throughσp from soils of the“Rio Preto”Hydrographic Basin,Bahia State,Brazil,in a monthly time lapse from 2016 to 2019.Traditionally,σp is measured by a laborious and time demanding laboratory analysis,making it unfeasible to map large areas.The innovative methodology of this work consists of combining active–passive satellite data on soil moisture and pedotransfer functions of clay content and water matric potential to obtain geo-located estimates ofσp.Estimates were analysed under different classes of soil use,land cover and slope;95%confidence intervals were built for the time series of mean values of LBC for each class.The overall seasonal variation in LBC estimates is similar in areas with annual crops,grasslands and natural vegetation,and flat areas are less affected by soil moisture variations over the year(between seasons).LBC decreased,in general,at about 0.5%a year in flat areas.Therefore,these areas demand attention,since they occupy 86%of the Basin and are mostly subjected to agricultural soil management and surface pressure by heavy machinery.

RPLP0/TBP are the most stable reference genes for human dental pulp stem cells under osteogenic differentiation

BACKGROUND Validation of the reference gene(RG)stability during experimental analyses is essential for correct quantitative real-time polymerase chain reaction(RT-qPCR)data normalisation.Commonly,in an unreliable way,several studies use genes involved in essential cellular functions[glyceraldehyde-3-phosphate dehydro-genase(GAPDH),18S rRNA,andβ-actin]without paying attention to whether they are suitable for such experimental conditions or the reason for choosing such genes.Furthermore,such studies use only one gene when Minimum Information for Publication of Quantitative Real-Time PCR Experiments guidelines recom-mend two or more genes.It impacts the credibility of these studies and causes dis-tortions in the gene expression findings.For tissue engineering,the accuracy of gene expression drives the best experimental or therapeutical approaches.We cultivated DPSCs under two conditions:Undifferentiated and osteogenic dif-ferentiation,both for 35 d.We evaluated the gene expression of 10 candidates for RGs[ribosomal protein,large,P0(RPLP0),TATA-binding protein(TBP),GAPDH,actin beta(ACTB),tubulin(TUB),aminolevulinic acid synthase 1(ALAS1),tyro-sine 3-monooxygenase/tryptophan 5-monooxygenase activation protein,zeta(YWHAZ),eukaryotic translational elongation factor 1 alpha(EF1a),succinate dehydrogenase complex,subunit A,flavoprotein(SDHA),and beta-2-micro-globulin(B2M)]every 7 d(1,7,14,21,28,and 35 d)by RT-qPCR.The data were analysed by the four main algorithms,ΔCt method,geNorm,NormFinder,and BestKeeper and ranked by the RefFinder method.We subdivided the samples into eight subgroups.RESULTS All of the data sets from clonogenic and osteogenic samples were analysed using the RefFinder algorithm.The final ranking showed RPLP0/TBP as the two most stable RGs and TUB/B2M as the two least stable RGs.Either theΔCt method or NormFinder analysis showed TBP/RPLP0 as the two most stable genes.However,geNorm analysis showed RPLP0/EF1αin the first place.These algorithms’two least stable RGs were B2M/GAPDH.For BestKeeper,ALAS1 was r

Land use change and occupation have led to modifications in the environment causing loss of biodiversity and ecosystem services throughout the planet.Some environments with high economic relevance,such as the ferruginous campo rupestre(rupestrian grassland known as Canga in Brazil),are even more susceptible to severe impacts due to their extreme habitat conditions and low resilience.The determination of reference ecosystems based on the intrinsic characteristics of the ecosystem is essential for conservation as well as to the implementation of ecological restoration.We proposed the reference ecosystem of the three main types of habitats of the ferruginous campo rupestre based on their floristic composition.We described the floristic composition of each habitat and evaluated the physicochemical properties of the soils and the relationship between plants and soils.All three habitats showed high diversity of plant species and many endemic species,such as Chamaecrista choriophylla,Cuphea pseudovaccinium,Lychnophora pinaster,and Vellozia subalata.The distribution of vegetation was strongly related with the edaphic characteristics,with a set of species more adapted to high concentration of base saturation,fine sand,organic carbon,and iron,while another set of species succeeded in more acidic soils with higher S and silt concentration.We provide support for the contention that the ferruginous campo rupestre is a mosaic of different habitats shaped by intrinsic local conditions.Failure to recognize the floristic composition of each particular habitat can lead to inappropriate restoration,increased habitat homogenization and increased loss of biodiversity and ecosystem services.This study also advances the knowledge base for building the reference ecosystem for the different types of ferruginous campo rupestre habitats,as well as a key database for highlighting those species contribute most to community assembly in this diverse and threatened tropical mountain ecosystem.

Background:The AbSeS-classification defines specific phenotypes of patients with intra-abdominal infection based on the(1)setting of infection onset(community-acquired,early onset,or late-onset hospital-acquired),(2)presence or absence of either localized or diffuse peritonitis,and(3)severity of disease expression(infection,sepsis,or septic shock).This classification system demonstrated reliable risk stratification in intensive care unit(ICU)patients with intra-abdominal infection.This study aimed to describe the epidemiology of ICU patients with pancreatic infection and assess the relationship between the components of the AbSeS-classification and mortality.Methods:This was a secondary analysis of an international observational study(“AbSeS”)investigating ICU patients with intra-abdominal infection.Only patients with pancreatic infection were included in this analysis(n=165).Mortality was defined as ICU mortality within 28 days of observation for patients discharged earlier from the ICU.Relationships with mortality were assessed using logistic regression analysis and reported as odds ratio(OR)and 95%confidence interval(CI).Results:The overall mortality was 35.2%(n=58).The independent risk factors for mortality included older age(OR=1.03,95%CI:1.0 to 1.1 P=0.023),localized peritonitis(OR=4.4,95%CI:1.4 to 13.9 P=0.011),and persistent signs of inflammation at day 7(OR=9.5,95%CI:3.8 to 23.9,Pbetween survivors and non-survivors.Conclusions:In pancreatic infection,a challenging source/damage control and ongoing pancreatic inflammation appear to be the strongest contributors to an unfavorable short-term outcome.In this limited series,essentials of the AbSeS-classification,such as the setting of infection onset,diffuse peritonitis,and severity of disease expression,

As the causative agent of coronavirus disease 2019(COVID-19),severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is one of the seven coronaviruses pathogenic to human and one of the four human coronaviruses that cause acute respiratory disease syndrome.Although it affects people of all age groups,the risk of mortality is age-dependent.As in many other diseases,elderly patients have a higher risk of infection,mortality,and morbidity[1].While it is a common belief that children fare better than adult and elderly,recent evidence merged that neonates and infants are associated with a higher risk of mortality and morbidity,compared to children and adolescents[2,3].For instance,a cross-country study found that the infection–fatality ratio of those agedbetween 5 and 14 years of 0.001 despite lower than those aged between 20 and 59 that vary between 0.006 and 0.323[4].However,whether this holds true in the risk of infection is not yet verified.Against this background,the present work aimed to verify whether neonates are more susceptible to COVID-19 using publicly available data of all polymerase chain reaction(PCR)tests performed in the state of São Paulo of Brazil.

Evolution of the 3.65-2.58 Ga Mairi Gneiss Complex,Brazil:Implications for growth of the continental crust in the São Francisco Craton

The composition and formation of the Earth’s primitive continental crust and mantle differentiation are key issues to understand and reconstruct the geodynamic terrestrial evolution,especially during the Archean.However,the scarcity of exposure to these rocks,the complexity of lithological relationships,and the high degree of superimposed deformation,especially with long-lived magmatism,make it difficult to study ancient rocks.Despite this complexity,exposures of the Archean Mairi Gneiss Complex basement unit in the São Francisco Craton offer important information about the evolution of South America’s primitive crust.Therefore,here we present field relationships,LA-ICP-SFMS zircon U-Pb ages,and LA-ICP-MCMS Lu-Hf isotope data for the recently identified Eoarchean to Neoarchean gneisses of the Mairi Complex.The Complex is composed of massive and banded gneisses with mafic members ranging from dioritic to tonalitic,and felsic members ranging from TTG(Tonalite-Trondhjemite-Granodiorite)to granitic composition.Our new data point to several magmatic episodes in the formation of the Mairi Gneiss Complex:Eoarchean(ca.3.65–3.60 Ga),early Paleoarchean(ca.3.55–3.52 Ga),middle-late Paleoarchean(ca.3.49–3.33 Ga)and Neoarchean(ca.2.74–2.58 Ga),with no records of Mesoarchean rocks.Lu-Hf data unveiled a progressive evolution of mantle differentiation and crustal recycling over time.In the Eoarchean,rocks are probably formed by the interaction between the pre-existing crust and juvenile contribution from chondritic to weakly depleted mantle sources,whereas mantle depletion played a role in the Paleoarchean,followed by greater differentiation of the crust with thickening and recycling in the middle–late Paleoarchean.A different stage of crustal growth and recycling dominated the Neoarchean,probably owing to the thickening of the continental crust by collision,continental arc growth,and mantle differentiation.

Old climatically-buffered infertile landscapes(OCBILs):more than harsh habitats,Atlantic Forest inselbergs can be drivers of evolutionary diversity

Granite-gneiss rock outcrop inselbergs are ancient stable ecosystems with old,climaticallybuffered infertile landscapes(OCBILs).Although inselbergs provide key ecosystem services,little is done for their conservation and,so far,a lot of their unknown evolutionary history has already been lost by human activities.Using a fine-scale approach,here we tested if habitat and environmental filtering(the inselberg’s harshness)affect the evolutionary diversity of an Atlantic Forest inselberg in Brazil.We recorded all trees with a diameter at breast height≥5cm in 20 plots in four habitat types(total sampled area of 0.8 hectares),from highest to lowest:island,hillside,foothill,and semideciduous forest(matrix).We also collected soil samples for chemical,textural and physical soil characterization.We fitted linear models to test the effects of soil and habitat on plotlevel metrics of phylogenetic diversity and structure,lineage diversity,phylogeneticβ-diversity,and evolutionary distinctiveness.We found that the upper inselberg habitats contain a distinct set of ancient,closely related,harsh-tolerant lineages,as well as a subset of lineages that persist under harsh conditions with a certain degree of water availability.The inferior inselberg habitats harbor higher lineage diversity than expected by chance.Soil strongly predicted evolutionary diversity.We concluded that soil depth,slope,nutrients and texture(environmental filtering)and habitat types and topography(habitat filtering)shape the evolutionary history contained in fine-scale inselberg habitats,which should encourage the conservation of these ancient ecosystems.

The main purpose of this paper is to demonstrate the relevance of the aesthetic understanding of the Western culture’s development in Nietzsche’s thought based on his artistic conception of interpretation.In order to accomplish this,his considerations on the figure of Homer in the production of the classical imaginary of Greek culture are taken as a discursive motif in the comparison between three distinct nuclei of his oeuvres,namely,his methodological discussions on philology,his theoretical analyses on the rhetorical nature of language-both from the time of his academic activity in Basel-and his philosophical reflections on creative interpretation in the 1880s.The fundamental premises raised by such comparison are the following:(a)interpretation as an art is a consequence of language as a pragmatically produced system of references;and(b)language is a practical aspect of the individual’s synthetic organic condition as a body in the world.As such,interpretation would be an aesthetic synthesis in Nietzsche’s philosophy.As a concluding remark,it should be demonstrated how the interpreter conceived as a creator is as worthwhile for understanding the historical processes of cultural transmission as it is for launching prospects for the future of cultural horizons.
