Carvacrol Promotes Cell Cycle Arrest and Apoptosis through PI3K/AKT Signaling Pathway in MCF-7 Breast Cancer Cells

Objective:To examine the role of carvacrol in modulating PI3 K/AKT signaling involved in human breast cancer pathogenesis using in vitro experimental model MCF-7 ***:MTT and lactate dehydrogenase assays were performed with cells treated with different doses of carvacrol(0–250μmol/L)at different time points(24 and 48 h).The nuclear morphology was assessed in MCF-7 cells with propidium iodide(PI)and acridine orange/ethidium bromide(AO/EB)staining and analyzed by fluorescence *** like cell cycle arrest and apoptosis were observed by flow cytometric analysis and expressions of p-Rb,cyclin D1,cyclin-dependent kinase 4(CDK4),CDK6,Bax,Bcl-2,PI3 K/p-AKT were analyzed by ***:Carvacrol significantly reduced cell viability with the half maximal inhibitory concentration value of 200μmol/L at 24 and 48 h(P<0.05).importantly,there was a significant increase in the accumulation of the G0/G1 phase upon treatment with carvacrol in MCF-7 cells(P<0.05 or P<0.01).A remarkable decrease in protein expressions of p-Rb,cyclin D1,CDK4 and CDK6 denoted cell cycle arrest(P<0.05 or P<0.01).In addition,carvacrol treatment significantly inhibited PI3 K/p-AKT protein expressions leading to induction of apoptosis mediated by decreased Bcl2 and increased Bax protein ***,Annexin V/PI staining by FACS analysis,dual staining by AO/EB and PI staining studies suggested induction of apoptosis by carvacrol through PI3 K/Akt signaling pathway in MCF-7 ***:Carvacrol significantly inhibited the breast cancer MCF-7 cell proliferation and induced apoptosis via suppressing PI3/AKT signaling pathway.

Genetic variants involved in gallstone formation and capsaicin metabolism,and the risk of gallbladder cancer in Chilean women

AIM:To determine the effects of genetic variants associated with gallstone formation and capsaicin (a pungent component of chili pepper) metabolism on the risk of gallbladder cancer (GBC).METHODS: A total of 57 patients with GBC, 119 patients with gallstones, and 70 controls were enrolled in this study. DNA was extracted from their blood or paraffi n block sample using standard commercial kits. The statuses of the genetic variants were assayed using Taqman SNP Genotyping Assays or Custom Taqman SNP Genotyping ***:The non-ancestral T/T genotype of apolipoprotein B rs693 polymorphism was associated with a decreased risk of GBC (OR:0.14,95% CI:0.03-0.63). The T/T genotype of cholesteryl ester transfer protein (CETP) rs708272 polymorphism was associated with an increased risk of GBC (OR:5.04,95% CI:1.43-17.8).CONCLUSION: Genetic variants involved in gallstone formation such as the apolipoprotein B rs693 and CETP rs 708272 polymorphisms may be related to the risk of developing GBC in Chilean women.

Anisotropic optical and electronic properties of two- dimensional layered germanium sulfide

Two-dimensional （2D） layered materials, transition-metal dichalcogenides, and black phosphorus have attracted considerable interest from the viewpoints of fundamental physics and device applications. The establishment of new functionalities in anisotropic layered 2D materials is a challenging but rewarding frontier, owing to the remarkable optical properties of these materials and their prospects for new devices. Herein, we report the anisotropic and thickness- dependent optical properties of a 2D layered monochalcogenide of germanium sulfide （GeS）. Three Raman-scattering peaks corresponding to the B3g,, A1g, and A2g modes with a strong polarization dependence are demonstrated in the GeS flakes, which validates polarized Raman spectroscopy as an effective method for identifying the crystal orientation of anisotropic layered GeS. Photoluminescence （PL） is observed with a peak at -1.66 eV that originates from the direct optical transition in GeS at room temperature. The polarization-dependent characteristics of the PL, which are revealed for the first time, along with the demonstration of anisotropic absorption, indicate an obvious anisotropic optical transition near the band edge of GeS, which is supported by density functional theory calculations. The significantly thickness-dependent PL is observed and discussed. This anisotropic layered GeS presents opportunities for the discovery of new physical phenomena and will find applications that exploit its anisotropic properties, such as polarization-sensitive photodetectors.

Effects of the viability of Lactobacillus rhamnosus GG on rotavirus infection in neonatal rats

Predicting factors contributing to knowledge, attitudes and practices relating to Zika virus infection among the general public in Malaysia

Objective: To identify the predicting factors that contribute to knowledge, attitude and practices relating to Zika virus infection among the general public in ***: A cross-sectional study was conducted using a validated self-administered questionnaire. Descriptive analysis was done for participants’ socio-demographic profile. Contingency table analysis was done to analyse the associations between knowledge, attitudes, and practices(KAP) scores and socio-demographic profile. A Bonferroni-corrected P-value was used to find the significance of the associations and multiple comparisons were performed in a single data set. To determine the linear relationship between each independent variable and the dependent variable, Spearman rank correlation was performed. Cohen’s correlation coefficient was evaluated to determine the strength of the effect size. Multiple correlations and regression analyses were performed to identify independent variables that predicts the dependent ***: Multiple correlation analyses were conducted between respondents’ KAP score and independent variables(Age >60 years;Female gender;Selangor state;At least 1 pregnant woman per household). The independent variables such as ‘Female gender’, ‘Selangor state’ and ‘At least 1 pregnant woman per household’ were positively and significantly correlated with KAP score whereas, age >60 years was negatively and significantly correlated with the KAP ***: There were associations between four independent factors and the KAP scores, while only three factors contributed to changes in KAP scores among the public. Among these contributing factors, respondents’ age group was the strongest predictor.

Study of Fourier-Based Velocimetry

Standard phase-domain pulsed Doppler techniques used in Colour Flow Mapping such as spectral Doppler or autocorrelation are monochromatic, focused on the analysis of the centre transmit frequency. As such all the algorithms using those approaches are limited: in terms of spatial Doppler resolution because of the long pulses typically used for transmission, in terms of frame rate because of the necessity to perform many Doppler lines repetitions and additional B-mode imaging transmissions, and in terms of accuracy which depends on the stability of the Doppler signal at the frequency considered. A velocimetry technique is presented which estimates the shifts between successive Doppler line segments using the phase information provided by the Fourier transform. Such an approach allows extraction of more information from the backscattered signal through the averaging of results from multiple frequencies inside the bandwidth, as well as the transmission of wide band-high resolution-pulses. The technique is tested on Doppler signals acquired with a research scanner in a straight latex pipe perfused with water and cellulose scatterers, and on an ultrasound contrast agent solution. The results are compared with the velocity estimates provided by standard spectral Doppler and autocorrelation methods. Results show that the proposed technique performs better than both other approaches, especially when few Doppler lines are processed. The technique is also shown to be compatible with contrast Doppler imaging. The proposed approach enables high frame rate, high resolution Doppler.

Multiferroic materials attracted a lot of attention in recent years because of their significant scientific interest and technological applications. The multiferroic core/shell powders have a better connectivity between the phases, resulting in superior dielectric and magneto electric properties. In this study, the influence of preparation condition on structure and properties of BaTiO_3/α-Fe_2 O_3 core/shell composite materials was examined. The five samples were obtained by varying synthesis conditions, such as synthesized method(co-precipitation and sonochemical method) and pH values of solution. XRD and Raman spectroscopy analyses were performed in order to determine phase composition and structural changes within samples. Morphology modifications were examined by SEM and EDS analyses. Finally, effect of structural and microstructural changes on magnetic and electrical properties was detected and explained.
