From the epididymis to the egg： participation of CRISP proteins in mammalian fertilization

Mammalian fertilization is a complex process that involves different steps of interaction between the male and female gametes. In spite of its relevance, the molecular mechanisms underlying this process still remain to be elucidated. The present review describes the contribution of our laboratory to the understanding of mammalian fertilization using Cysteine-Rlch Secretory Proteins （CRISP） as model molecules. Substantial evidence obtained from in vitro assays and knockout models shows that epididymal CRISP1 associates with the sperm surface with two different affinities during maturation, and participates in the regulation of signaling pathways during capacitation as well as in both sperm-zona pellucida interaction and gamete fusion. These observations can be extended to humans as judged by our findings showing that the human homolog of the rodent protein （hCRISP1） is also involved in both stages of fertilization. Evidence supports that other members of the CRISP family secreted in the testis （CRISP2）, epididymis （CRISP3-4） or during ejaculation （CRISP3） are also involved in sperm-egg interaction, supporting the existence of a functional redundancy and cooperation between homolog proteins ensuring the success of fertilization. Together, our observations indicate that CRISP proteins accompany spermatozoa along their transit through both the male and female reproductive tracts. We believe these results not only contribute to a better mechanistic understanding of fertilization but also support CRISP proteins as excellent candidates for future research on infertility and contraception.

Background Nursery pigs undergo stressors in the post-weaning period that result in production and welfare *** challenges disproportionately impact the offspring of primiparous sows compared to those of mul-tiparous *** is known regarding potential interactions between parity and feed additives in the post-weaning period and their effects on nursery pig ***,the objective of this study was to investigate the effects of maternal parity on sow and offspring microbiomes and the influence of sow parity on pig fecal microbi-ome and performance in response to a prebiotic *** weaning,piglets were allotted into three treat-ment groups:a standard nursery diet including pharmacological doses of Zn and Cu(Con),a group fed a commercial prebiotic only(Preb)based on an Aspergillus oryzae fermentation extract,and a group fed the same prebiotic plus Zn and Cu(Preb+ZnCu).Results Although there were no differences in vaginal microbiome composition between primiparous and mul-tiparous sows,fecal microbiome composition was different(R^(2)=0.02,P=0.03).The fecal microbiomes of primiparous offspring displayed significantly higher bacterial diversity compared to multiparous offspring at d 0 and d 21 post-weaning(Pdifferences in community composition observed at d 21(R^(2)=0.03,P=0.04).When analyzing the effects of maternal parity within each treatment,only the Preb diet triggered significant microbiome distinc-tions between primiparous and multiparous offspring(d 21:R^(2)=0.13,P=0.01;d 42:R^(2)=0.19,P=0.001).Composi-tional differences in pig fecal microbiomes between treatments were observed only at d 21(R^(2)=0.12,P=0.001).Pigs in the Con group gained significantly more weight throughout the nursery period when compared to those in the Preb+ZnCu *** Nursery pig gut microbiome composition was influenced by supplementation with an Aspergillus oryzae fermentation extract,with varying effects on performance

AIM:To investigate whether we could create natural autologous tissue patches in the subcutaneous space for organ repair. METHOdS: We implanted the following three types of inert foreign bodies in the subcutaneous tissue of rats to produce autologous tissue patches of different geometries:(1) a large-sized polyvinyl tube(L = 25 mm,internal diameter = 7 mm) sealed at both ends by heat application for obtaining a large flat piece of tissue patch for organ repair;(2) a fine polyvinyl tubing(L = 25 mm,internal diameter = 3 mm) for creating cylindrically shaped grafts for vascular or nerve repair; and(3) a slurry of polydextran particle gel for inducing a bladder-like tissue. Implantation of inert materials was carried out by making a small incision on one or either side of the thoracic-lumbar region of rats. Subcutaneous pockets were created by blunt dissection around the incision into which the inert bodies were inserted(1 or 2 per rat). The incisions were closed with silk sutures,and the animals were allowed to recover. In case of the polydextran gel slurry 5 m L of the slurry was injected in the subcutaneous space using an 18 gauge needle. After implanting the foreign bodies a newly regenerated encapsulating tissue developed around the foreign bodies. The tissues were harvested after 4-42 d of implantation and studied by gross examination,histology,and histochemistry for organization,vascularity,and presence of mesenchymal stem cells(MSCs)(Cd271+Cd34+ cells). RESULTS: Implanting a large cylindrically shaped polyvinyl tube resulted in a large flat sheet of tissue that could be tailored to a specific size and shape for use as a tissue patch for repairing large organs. Implanting a smaller sized polyvinyl tube yielded a cylindrical tissue that could be useful for repairing nerves and blood vessels. BACKGROUNd Type 1 diabetes(T1d)is a complex disease with a higher incidence in Europeans than other *** Colombians Living in Medellin(CLM)is admixed with ancestry contributions from Europeans,Native Americans(NAT)and Africans(AFR).AIM Our aim was to analyze the genetic admixture component at candidate T1d loci in Colombian individuals with the *** Seventy-four ancestry informative markers(AIMs),which tagged 41 T1d candidate loci/genes,were tested by studying a cohort of 200 Northwest Colombia diseased individuals.T1d status was classified by testing for glutamic acid decarboxylase(GAd-65 kda)and protein tyrosine-like antigen-2 autoantibodies in serum *** loci/genes included HLA,INS,PTPN22,CTLA4,IL2RA,SUMO4,CLEC16A,IFIH1,EFR3B,IL7R,NRP1 and RNASEH1,amongst *** 1,000 genome database was used to analyze data from 94 individuals corresponding to the reference *** the data did not comply with a normal distribution,medians were compared between groups using the Mann-Whitney *** Both T1d patients and individuals from CLM displayed mainly European ancestry(61.58 vs 62.06)followed by Native American(27.34 vs 27.46)and to a lesser extent the AFR ancestry(10.28 vs 10.65)***,compared to CLM,ancestry of T1d patients displayed a decrease of NAT ancestry at gene EFR3B(24.30 vs 37.10)and an increase at genes IFIH1(32.07 vs 14.99)and IL7R(52.18 vs 39.18).Also,for gene NRP1(36.67 vs 0.003),we observed a non-AFR contribution(attributed to NAT).Autoimmune patients(positive for any of two auto-antibodies)displayed lower NAT ancestry than idiopathic patients at the MHC region(20.36 vs 31.88).Also,late onset patients presented with greater AFR ancestry than early onset patients at gene IL7R(19.96 vs 6.17).An association analysis showed that,even after adjusting for admixture,an association exists for at least seven such AIMs,with the strongest findings on chromosomes 5 and 10(gene IL7R,P=5.56×10-6 and gene NRP1,P=8.

El Soplao outcrop, an Early Cretaceous amber deposit recently discovered in northern Spain （Cantabria）, has been shown to be the largest site of amber with arthropod inclusions that has been found in Spain so far. Relevant data provided herein for biogeochemistry of the amber, palynology, taphonomy and arthropod bioinclusions complement those previously published. This set of data suggests at least two botanical sources for the amber of El Soplao deposit. The first （type A amber） strongly supports a source related to Cheirolepidiaceae, and the second （type B amber） shows non-specific conifer biomarkers. Comparison of molecular composition of type A amber with Frenelopsis leaves （Cheirolepidiaceae） strongly suggests a biochemical affinity and a common botanical origin. A preliminary palynological study indicates a regional high taxonomical diversity, mainly of pteridophyte spores and gymnosperm pollen grains. According to the preliminary palynological data, the region was inhabited by conifer forests adapted to a dry season under a subtropical climate. The abundant charcoalified wood associated with the amber in the same beds is evidence of paleofires that most likely promoted both the resin production and an intensive erosion of the litter, and subsequent great accumulation of amber plus plant cuticles. In addition, for the first time in the fossil record, charcoalified plant fibers as bioinclnsions in amber are reported. Other relevant taphonomic data are the exceptional presence of serpulids and bryozoans on the surfaces of some amber pieces indicating both a long exposure on marine or brackish-water and a mixed assemblage of amber. Lastly, new findings of insect bioiuclusions, some of them uncommon in the fossil record or showing remarkable adaptations, are reported. In conclusion, a documented scenario for the origin of the El Soplao amber outcrop is provided.
