Elevated alanine aminotransferase activity is not associated with dyslipidemias,but related to insulin resistance and higher disease grades in non-diabetic non-alcoholic fatty liver disease

Objective:To explore demographic and metabolic factors associated with increased alanine aminotransferase(alT)activity in non-diabetic non-alcoholic fatty liver disease(NAFLD)***:Overall 372 patients who consecutively attended to Gastroenterology Clinic of Baqiyatallah University of Medical Sciences,Tehran,Iran awere diagnosed as NAFLD entered into *** criteria were having diabetes mellitus and fasting blood glucose over126 mg/dL,active hepatitis B virus infection,having hepatitis C virus positive serology,and to be under corticosteroid *** levels were considered pathologically high when it was over30 IU/L for men and over 19 IU/L for ***:Bivariate analyses using t test and chisquare test showed that patients with pathologically augmented alT levels had significantly higher NAFLD grades in their ultrasonographic evaluations(P=0.003).Moreover,these patients represented significantly higher homeostatic model assessment levels(P=0.003),levels of serum insulin(P=0.002),fasting blood glucose(Palence of insulin resistance was also higher in patients with increased serum alT *** logistic regression models showed that ultrasonographic grading of NAFLD(P=0.027)and insulin resistance(P=0.013)were the only variables significantly associated with abnormal alT ***:This study shows that the associations of increased alT serum levels in NAFLD patients are different from what are supposed *** excluding diabetic patients from our population,we find that increased alT levels are not associated with dyslipidemias but are independently associated with insulin resistance and NAFLD grading on ultrasonographic *** studies are needed to confirm our results.

Square mesh codend improves size selectivity and catch pattern for Trichiurus lepturus in bottom trawl used along Northwest coast of India

The size selectivity and catch pattern of Trichiurus lepturus in 40 mm diamond and square mesh codends were investigated in this study.A 34 m high opening trawl was used,which is a common design used by fishermen operating along the northwest coast of India,and the cover codend method was employed to determine codend *** with the changes in the length at 50%retention rate for Trichiurus lepturus with the usage of the square and diamond mesh codends,indicators to understand the exploitation pattern of this species in the fishery were assessed based on the total number of individuals and on total weight of the *** mean selection length increased and discard fraction is reduced when 40 mm square mesh codends are used in place of diamond meshes,however,it is observed that individuals at commercial length are also *** results demonstrate that mandatory use of legal mesh sizes alone will be insufficient to aid in the sustainable harvest of this species,given its estimated length at first sexual maturity of 61.2 *** is the first study to compare the size selection of this species in 40 mm diamond and square mesh codends along Northwest coast of *** of the study will help as a baseline for gear-based regulations in the region.

Coordinated regulation of plant immunity by poly(ADP-ribosyl)ation and K63-linked ubiquitination

Poly(ADP-ribosyl)ation(PARylation)is a posttranslational modification reversibly catalyzed by poly(ADP-ribose)polymerases(PARPs)and poly(ADP-ribose)glycohydrolases(PARGs)and plays a key role in multi-ple cellular *** molecular mechanisms by which PARylation regulates innate immunity remain largely unknown in *** we show that Arabidopsis UBC13A and UBC13B,the major drivers of lysine 63(K63)-linked polyubiquitination,directly interact with PARPs/*** of pathogen-associated molecular pattern(PAMP)-triggered immunity promotes these interactions and enhances PARylation of *** parp1 parp2 and ubc13a ubc13b mutants are compromised in immune responses with increased accumulation of total pathogenesis-related(PR)proteins but decreased accu-mulation of secreted PR *** disulfide-isomerases(PDIs),essential components of endo-plasmic reticulum quality control(ERQC)that ensure proper folding and maturation of proteins destined for secretion,complex with PARPs/PARGs and are PARylated upon PAMP ***,PARylation of UBC13 regulates K63-linked ubiquitination of PDIs,which may further promote their disulfide isomerase activities for correct protein folding and subsequent *** together,these results indicate that plant immunity is coordinately regulated by PARylation and K63-linked ubiquitination.

Mycological tools to estimate the effects of diverse land-use practices on fungal diversity are scarce,because of poor knowledge of the taxonomic diversity of tropical fungi and their response to anthropogenic habitat ***,we investigate assemblages of fungal spores,recently deposited in the bottom sediments of 24 small crater lakes in western Uganda,to assess the relationship between the local richness of fungi and environmental variation in the crater basin along regional gradients of natural vegetation and land *** recovered~9500 fungal spore specimens,which could be attributed to 216 *** an information-theoretic approach based on the corrected Akaike Information Criterion(AICc),we determined the environmental factors which best explained variation in the diversity of fungal spores among three datasets:(i)the full set of 24 crater basins,(ii)the subset of 22 basins with freshwater lakes,and(iii)the subset of 17 basins partly or completely in agricultural use(cropland,fallow land,pasture and plantation).In these 17 human-impacted crater basins our results revealed a negative relationship between fungal spore richness and the areal fraction of basins in agricultural ***,this detrimental effect of land use on fungal spore richness was not apparent across the full set of both disturbed and(presently)undisturbed *** was due to large variation in fungal spore richness among the undisturbed basins covered either with forest or savannah vegetation,probably resulting from site-specific controls on fungal habitat diversity,such as climatic moisture balance and the composition of natural and/or secondary *** land-use effects on fungal spore diversity,as documented in this study,suggest that communities of tropical fungi progressively exposed to land-use practices are threatened by species ***,our study demonstrates the need to develop conservation strategies mitigating the impacts of agriculture on the biodiversity o

Effect of Prometheus liver assist system on systemic hemodynamics in patients with cirrhosis: A randomized controlled study

AIM： To evaluate treatment safety and hemodynamic changes during a single 6-h treatment with the PrometheusTM liver assist system in a randomized, controlled study. METHODS： Twenty-four patients were randomized to either the study group or to one of two control groups： Fractionated Plasma Separation Adsorption and Dialysis, PrometheusTM system （Study group; n = 8）; Molecular Adsorbent Recirculation System （MARS）TM （Control group 1, n = 8）; or hemodialysis （Control group 2; n = 8）. all patients included in the study had decompensated cirrhosis at the time of the inclusion into the study. Circulatory changes were monitored with a Swan-Ganz catheter and bilirubin and creatinine were monitored as measures of protein-bound and water-soluble toxins. RESULTS： Systemic hemodynamics did not differ between treatment and control groups apart from an increase in arterial pressure in the MARS group （P = 0.008）. No adverse effects were observed in any of the groups. Creatinine levels significantly decreased in the MARS group （P = 0.03） and hemodialysis group （P = 0.04）. Platelet count deceased in the Prometheus group （P = 0.04）.CONCLUSION： Extra-corporal liver support with Prometheus is proven to be safe in patients with endstage liver disease but does not exert the beneficial effects on arterial pressure as seen in the MARS group,

Sildenafil does not influence hepatic venous pressure gradient in patients with cirrhosis

AIM： To investigate if sildenafil increases splanchnic blood flow and changes the hepatic venous pressure gradient （HVPG） in patients with cirrhosis. Phosphodiesterase type-5 inhibitors are valuable in the treatment of erectile dysfunction and pulmonary hypertension in patients with end-stage liver disease. However, the effect of phosphodiesterase type-5 inhibitors on splanchnic blood flow and portal hypertension remains essentially unknown. METHODS： Ten patients with biopsy proven cirrhosis （five females/five males, mean age 54：1：8 years） and an HVPG above 12 mmHg were studied after informed consent. Measurement of splanchnic blood flow and the HVPG during liver vein catheterization were done before and 80 min after oral administration of 50 mg sildenafil. Blood flow was estimated by use of indocyanine green clearance technique and Fick＇s principle, with correction for non-steady state. RESULTS： The plasma concentration of sildenafil was 222 ± 136 ng/mL 80 min after administration. Mean arterial blood pressure decreased from 77 ±7 mmHg to 66 ± 12 mmHg, P = 0.003, while the splanchnicblood flow and oxygen consumption remained unchanged at 1.14 ± 0.71 L/min and 2.3 ± 0.6 mmol/ min, respectively. also the HVPG remained unchanged （18 ± 2 mmHg vs 16 ± 2 mmHg） with individual changes ranging from -8 mmHg to ±2 mmHg. In seven patients, HVPG decreased and in three it increased. CONCLUSION： In spite of arterial blood pressure decreases 80 min after administration of the phosphodiesterase type-5 inhibitor sildenafil, the present study could not demonstrate any clinical relevant influence on splanichnic blood flow, oxygen consumption or the HVPG.

Polymorphisms in NF-κB,PXR,LXR,PPARγ and risk of inflammatory bowel disease

AIM:To investigate the contribution of polymorphisms in nuclear receptors to risk of inflammatory bowel disease(IBD) . METHODS:Genotypes of nuclear factor(NF) -κB(NFKB1) NFκB-94ins/del(rs28362491) ;peroxisome proliferatoractivated receptor(PPAR) -γ(PPARγ) PPARγPro12ala(rs1801282) and C1431T(rs 3856806) ;pregnane X receptor(PXR) (NR1I2) PXR A-24381C(rs1523127) ,C8055T(2276707) ,and A7635G(rs 6785049) ;and liver X receptor(LXR) (NR1H2) LXR T-rs1405655-C and T-rs2695121-C were assessed in a Danish case-control study of 327 Crohn's disease patients,495 ulcerative colitis(UC) patients,and 779 healthy *** ratio(OR) and 95%CI were estimated by logistic regression models. RESULTS:The PXR A7635G variant,the PPARγPro12ala and LXR T-rs2695121-C homozygous variant genotypes were associated with risk of UC(OR:1.31,95% CI:1.03-1.66,P=0.03,OR:2.30,95%CI:1.04-5.08,P=0.04,and OR:1.41,95%CI:1.00-1.98,P=0.05,respectively) compared to the corresponding homozygous wild-type *** never smokers,PXR A7635G and the LXR T-rs1405655-C and T-rs2695121-C variant genotypes were associated with risk of IBD(OR:1.41,95%CI:1.05-1.91,P=0.02,OR:1.63,95%CI:1.21-2.20,P=0.001,and OR:2.02,95%CI:1.36-2.99,P=0.0005,respectively) compared to the respective homozygous variant *** A7635G(rs6785049) variant genotype was associated with a higher risk of UC diagnosis before the age of 40 years and with a higher risk of extensive disease(OR:1.34,95%CI:1.03-1.75 and OR:2.49,95%CI:1.24-5.03,respectively) . CONCLUSION:Common PXR and LXR polymorphisms may contribute to risk of IBD,especially among never smokers.
