Human-specific insertions play important roles in human phenotypes and *** we reported a 446-bp insertion(Insert-446)in intron 11 of the TBC1D8B gene,located on chromosome X,and traced its origin to a portion of intron 6 of the EBF1 gene on chromosome ***,Insert-446 was present in the human Neanderthal and Denisovans genomes,and was fixed in humans after human-chimpanzee *** have demonstrated that Insert-446 acts as an enhancer through binding transcript factors that promotes a higher expression of human TBC1D8B gene as compared with orthologs in *** addition,over-expression TBC1D8B promoted cell proliferation and migration through“a dual finger”catalytic mechanism(Arg538 and Gln573)in the TBC domain in vitro and knockdown of TBC1D8B attenuated tumorigenesis in *** of Insert-446 prevented cell proliferation and migration in cancer and normal *** results reveal that the human-specific Insert-446 promotes cell proliferation and migration by upregulating the expression of TBC1D8B *** findings provide a significant insight into the effects of human-specific insertions on evolution.

High-performance flexible pressure sensors provide comprehensive tactile perception and are applied in human activity monitoring,soft robotics,medical treatment,and human-computer ***,these flexible pressure sensors require extensive nano-architectural design and complicated manufacturing and are ***,a highly sensitive,flexible piezoresistive tactile sensor is designed and fabricated,consisting of three main parts:the randomly distributed microstructure on T-ZnOw/PDMS film as a top substrate,multilayer Ti_(3)C_(2)-MXene film as an intermediate conductive filler,and the few-layer Ti_(3)C_(2)-MXene nanosheetbased interdigital electrodes as the bottom *** MXene-based piezoresistive sensor with randomly distributed microstructure exhibits a high sensitivity over a broad pressure range(less than 10 kPa for 175 kPa^(-1))and possesses an out-standing permanence of up to 5000 ***,a 16-pixel sensor array is designed,and its potential applications in visualizing pressure distribution and an example of tactile feedback are *** fully sprayed MXene-based pressure sensor,with high sensitivity and excellent durability,can be widely used in,electronic skin,intelligent robots,and many other emerging technologies.

KCNQ1 rs2237895 gene polymorphism increases susceptibility to type 2 diabetes mellitus in Asian populations

BACKGROUND The association of single nucleotide polymorphism of KCNQ1 gene rs2237895 with type 2 diabetes mellitus(T2DM)is currently *** is unknown whether this association can be gene realized across different *** To determine the association of KCNQ1 rs2237895 with T2DM and provide reliable evidence for genetic susceptibility to *** We searched PubMed,Embase,Web of Science,Cochrane Library,Medline,Baidu Academic,China National Knowledge Infrastructure,China Biomedical Literature Database,and Wanfang to investigate the association between KCNQ1 gene rs2237895 and the risk of T2DM up to January 12,*** Manager 5.4 was used to analyze the association of the KCNQ1 gene rs2237895 polymorphism with T2DM and to evaluate the publication bias of the selected *** Twelve case–control studies(including 11273 cases and 11654 controls)met our inclusion *** the full population,allelic model[odds ratio(OR):1.19;95%confidence interval(95%CI):1.09–1.29;P<0.0001],recessive model(OR:1.20;95%CI:1.11–1.29;P<0.0001),dominant model(OR:1.27.95%CI:1.14–1.42;P<0.0001),and codominant model(OR:1.36;95%CI:1.15–1.60;P=0.0003)(OR:1.22;95%CI:1.10–1.36;P=0.0002)indicated that the KCNQ1 gene rs2237895 polymorphism was significantly correlated with susceptibility to *** stratified analysis,this association was confirmed in Asian populations:allelic model(OR:1.25;95%CI:1.13–1.37;P<0.0001),recessive model(OR:1.29;95%CI:1.11–1.49;P=0.0007),dominant model(OR:1.35;95%CI:1.20–1.52;P<0.0001),codominant model(OR:1.49;95%CI:1.22–1.81;P<0.0001)(OR:1.26;95%CI:1.16–1.36;P<0.0001).In non-Asian populations,this association was not significant:Allelic model(OR:1.06,95%CI:0.98–1.14;P=0.12),recessive model(OR:1.04;95%CI:0.75–1.42;P=0.83),dominant model(OR:1.06;95%CI:0.98–1.15;P=0.15),codominant model(OR:1.08;95%CI:0.82–1.42;P=***:1.15;95%CI:0.95–1.39;P=0.14).CONCLUSION KCNQ1 gene rs2237895 was significantly associated with susceptibility to T2DM

Structural basis of negative regulation of CRISPR-Cas7-11 by TPR-CHAT

CRISPR‒Cas7-11 is a Type Ⅲ-E CRISPR-associated nuclease that functions as a potent RNA editing ***-peptide repeat fused with Cas/HEF1-associated signal transducer(TPR-CHAT)acts as a regulatory protein that interacts with CRISPR RNA(crRNA)-bound Cas7-11 to form a CRISPR-guided caspase complex(Craspase).However,the precise modulation of Cas7-11’s nuclease activity by TPR-CHAT to enhance its utility requires further ***,we report cryo-electron microscopy(cryo-EM)structures of Desulfonema ishimotonii(Di)Cas7-11-crRNA,complexed with or without the full length or the N-terminus of *** structures unveil the molecular features of the Craspase *** analysis,combined with in vitro nuclease assay and electrophoretic mobility shift assay,reveals that DiTPR-CHAT negatively regulates the activity of DiCas7-11 by preventing target RNA from binding through the N-terminal 65 amino acids of DiTPR-CHAT(DiTPR-CHAT_(NTD)).Our work demonstrates that DiTPRCHAT_(NTD) can function as a small unit of DiCas7-11 regulator,potentially enabling safe applications to prevent overcutting and offtarget effects of the CRISPR‒Cas7-11 system.

Plastic offers a new niche for microorganisms,the *** everincreasing emission of plastic waste makes it critical to understand the microbial ecology of the plastisphere and associated ***,we present a global fingerprint of the plastisphere,analyzing samples collected from freshwater,seawater,and terrestrial *** plastisphere assembles a distinct microbial community that has a clearly higher heterogeneity and a more deterministically dominated assembly compared to natural *** coexistence patterns—loose and fragile networks with mostly specialist linkages among microorganisms that are rarely found in natural habitats—are seen in the *** microbiomes generally have a great potential to metabolize organic compounds,which could accelerate carbon *** involved in the nitrogen cycle are also altered in the plastisphere,especially in freshwater plastispheres,where a high abundance of denitrifiers may increase the release of nitrite(aquatic toxicant)and nitrous oxide(greenhouse gas).Enrichment of animal,plant,and human pathogens means that the plastisphere could become an increasingly mobile reservoir of harmful *** findings highlight that if the trajectory of plastic emissions is not reversed,the expanding plastisphere could pose critical planetary health challenges.

The burgeoning growth of the new energy vehicles and aviation industry has escalated the need for energy storage capacitors capable of stable operation in harsh *** advent of metal-polyimide complexes has illuminated a novel approach for preparing tem perature-resistant ***,the general application of these metal-polyimide com plexes is impeded by the high dielectric loss and low breakdown strength,consequences of main-chain coordination and excessive metal ions ***,our study proposes a novel polyimideCu complex material(POP-Cu)predicated on side-chain-type pyridine-Cu coordination,utilizing the structural backbone PMDA-ODA of mature commercial PI(Kapton)with reliable *** to the high degree of freedom afforded by the side chain with suppressed relaxation activation energy and the long-range electron delocalization formed by d-πcoordination,the dielectric constant of this material containing merely 2.7 mol%Cu increases from 3.25(POPI)to 5.58,while maintaining a remarkably low dielectric loss of ***,this material exhibits a substantial DC breakdown strength of 436.2 MV·m^(-1)and a high energy density of 5.42 J·cm^(-3),coupled with superior mechanical and thermal *** at 150℃,it retains over 90%of its room-temperature energy density,demon strating notable dielectric stability under high *** attributes underscore its promising application for capacitors operating in harsh environments.

In prevailing p-i-n perovskite solar cells(PSCs),solution-processible fullerene molecules are widely used as electron-transporting layers(ETLs)but they typically suffer from poor uniformity and undesirable stability ***,a separate bathocuproine(BCP)layer is needed to block hole tran sfer,increasing fabrication complexity and ***,we address these limitations by develo ping a novel polymeric ETL(named PF BCP)synthesized by polymerizing C_(60)with *** innovative material achieves both efficient electron transport and hole blocking,while its excellent uniformity minimizes interface recombination and enhances ***,our blade-coated PSCs utilizing PFBCP achieve a high power conversion efficiency exceeding 22%and retain 91%of initial efficiency after 1200 h of light *** development not only paves the way for commercially viable PSCs but also opens avenues for future ETL design to realize even more efficient and stable PSCs.
