Exercise has long been known for its active role in improving physical fitness and sustaining health.Regular moderate-intensity exercise improves all aspects of human health and is widely accepted as a preventative and therapeutic strategy for various diseases.It is well-documented that exercise maintains and restores homeostasis at the organismal,tissue,cellular,and molecular levels to stimulate positive physiological adaptations that consequently protect against various pathological conditions.Here we mainly summarize how moderate-intensity exercise affects the major hallmarks of health,including the integrity of barriers,containment of local perturbations,recycling and turnover,integration of circuitries,rhythmic oscillations,homeostatic resilience,hormetic regulation,as well as repair and regeneration.Furthermore,we summarize the current understanding of the mechanisms responsible for beneficial adaptations in response to exercise.This review aimed at providing a comprehensive summary of the vital biological mechanisms through which moderate-intensity exercise maintains health and opens a window for its application in other health interventions.We hope that continuing investigation in this field will further increase our understanding of the processes involved in the positive role of moderate-intensity exercise and thus get us closer to the identification of new therapeutics that improve quality of life.

Background Tuberous sclerosis complex(TSC)is an autosomal dominant genetic disorder with a birth incidence of I:6000 in the United States that is characterized by the growth of non-cancerous tumors in multiple organ systems including the brain,kidneys,lungs,and skin.Importantly,TSCis also associated with signicant neurological manifestations including epilepsy TSC-associated neuropsychiatric disorders,intellectual disabilities,and autism spectrum disorder.Mutations in the TSCI or TSC2 genes are well-established causes of TSC,which lead to TSC1/TSC2 deficiency in organs and hyper-activation of the mammalian target of rapamycin signaling pathway.Animal models have been widely used to study the effect of TSCl/2 genes on the development and function of the brain.Despite considerable progress in understanding the molecular mechanisms underlying TSC in animal models,a human-specific model is urgently needed to investigate the effects of TSCl/2 mutations that are unique to human neurodevelopment.Data sources Literature reviews and research articles were published in PubMed-indexed journals.Results Human-induced pluripotent stem cells(iPSCs),which capture risk alleles that are identical to their donors and have the capacity to differentiate into virtually any cell type in the human body,pave the way for the empirical study of previously inaccessible biological systems such as the developing human brain.Conclusions In this review,we present an overview of the recent progress in modeling TSC with human iPSC models,the existing limitations,and potential directions for future research.

Floating plants reduced methane fluxes from wetlands by creating a habitat conducive to methane oxidation

Wetlands are one of the important natural sources of atmospheric methane (CH_4),as an important part of wetlands,floating plants can be expected to affect methane release.However,the effects of floating plants on methane release are limited.In this study,methane fluxes,physiochemical properties of the overlying water,methane oxidation potential and rhizospheric bacterial community were investigated in simulated wetlands with floating plants Eichhornia crassipes,Hydrocharis dubia,and Trapa natans.We found that E.crassipes,H.dubia,and T.natans plants could inhibit 84.31%-97.31%,4.98%-88.91%and 43.62%-92.51%of methane fluxes at interface of water-atmosphere compared to Control,respectively.Methane fluxes were negatively related to nutrients concentration in water column but positively related to the aerenchyma proportions of roots,stems,and leaves.At the same biomass,root of E.crassipes (36.44%) had the highest methane oxidation potential,followed by H.dubia (12.99%) and T.natans (11.23%).Forty-five bacterial phyla in total were identified on roots of three plants and 7 bacterial genera (2.10%-3.33%) were known methanotrophs.Type I methanotrophs accounted for 95.07%of total methanotrophs.The pmoA gene abundances ranged from 1.90×10^(16)to 2.30×10^(18)copies/g fresh weight of root biofilms.Abundances of pmoA gene was significantly positively correlated with environmental parameters.Methylotrophy (5.40%) and methanotrophy (3.75%) function were closely related to methane oxidation.This study highlights that floating plant restoration can purify water and promote carbon neutrality partially by reducing methane fluxes through methane oxidation in wetlands.

Laser-induced plasma formation in water with up to 400 mJ double-pulse LIBS

Double-pulse LIBS is a promising technique for deep-sea applications.LIBS measurements in shallow water with up to 400 mJ each pulse were done to select laser parameters which promote optimized spectral line emission from plasma even at elevated pressures,where line broadening until loss of most of the spectral information can occur.Optical emission spectroscopy,using a Czerny-Turner spectrometer,has been applied to investigate the dependence of the emitted radiation on laser parameters and hydrostatic pressure.It has been found,that higher laser pulse energies,especially with short pulse delay as required in high water pressure,can also have an adverse effect on the measured spectrum.

Melatonin is an endogenous monoamine hormone secreted by the pineal gland.Melatonergic signaling has been shown to play a role in circadian rhythm regulation,lipid and glucose metabolism,and obesity,and it has anti-inflammatory and antioxidant properties.The melatonin receptor 1B gene(MTNR1B)is expressed in,among other tissues,the brain and pancreatic beta cells,and risk variants for T2D have been reported as impairing early insulin secretion and increasing fasting glucose levels.^(1) Variants in the MTNR1B gene also have been reported in patients with depression(MDD).

An ancient conflict between hosts and pathogens has driven the innate and adaptive arms of immunity.Knowledge about this interplay can not only help us identify biological mechanisms but also reveal pathogen vulnerabilities that can be leveraged therapeutically.The humoral response to SARS-CoV-2 infection has been the focus of intense research,and the role of the innate immune system has received significantly less attention.Here,we review current knowledge of the innate immune response to SARS-CoV-2 infection and the various means SARS-CoV-2 employs to evade innate defense systems.We also consider the role of innate immunity in SARS-CoV-2 vaccines and in the phenomenon of long COVID.

Cost effectiveness of nonpharmacological prevention programs for diabetes:A systematic review of trial-based studies

Trial-based economic value of prevention programs for diabetes is inexplicit.We aimed to review the cost-effectiveness of nonpharmacological interventions to prevent type-2 diabetes mellitus(T2DM)for high-risk people.Six electronic databases were searched up to March 2022.Studies assessing both the cost and health outcomes of nonpharmacological interventions for people at high-risk of T2DM were included.The quality of the study was assessed by the Consolidated Health Economic Evaluation Reporting Standards 2022 checklist.The primary outcome for synthesis was incremental cost-effectiveness ratios(ICER)for quality-adjusted life years(QALYs),and costs were standardized in 2022 US dollars.Narrative synthesis was performed,considering different types and delivery methods of interventions.Sixteen studies included five based on the US diabetes prevention program(DPP),six on non-DPP-based lifestyle interventions,four on health education,and one on screening plus lifestyle intervention.Compared with usual care,lifestyle interventions showed higher potential of cost-effectiveness than educational interventions.Among lifestyle interventions,DPP-based programs were less cost-effective(median of ICERs:$27,077/QALY)than non-DPP-based programs(median of ICERs:$1395/QALY)from healthcare perspectives,but with larger decreases in diabetes incidence.Besides,the cost-effectiveness of interventions was more possibly realized through the combination of different delivery methods.Different interventions to prevent T2DM in high-risk populations are both cost-effective and feasible in various settings.Nevertheless,economic evidence from low-and middle-income countries is still lacking,and interventions delivered by trained laypersons and combined with peer support sessions or mobile technologies could be potentially a cost-effective solution in such settings with limited resources.

This study introduces a novel image capture and lighting techniques using a cutting-edge hybrid MEMS scanner system designed for compact microscopic imaging.The scanner comprises a tapered optical fiber waveguide and innovative aerosol-jet printed PZT(lead zirconate titanate)bimorph push-pull actuators on a stainless-steel substrate,effectively addressing issues that are commonly associated with PZT on silicon substrates such as fracture and layer separation.By leveraging nonlinear vibration,the scanner achieves a spiral scan pattern from a single signal input,in addition to the expected two-dimensional scanning and target illumination from two phase-shifted inputs.This capability is further enhanced by a novel process to taper the optical fiber,which reduces illumination scattering and tunes the fiber to the resonant frequencies of the scanner.The precisely tapered tip enables large fields of view while maintaining independent 2-axis scanning through one-degree-of-freedom actuation.Experimental validation showcases the successful generation of a spiral scan pattern with a 60μm diameter scan area and a 10 Hz frame rate,effectively reconstructing scanned images of 5μm lines,cross patterns(15μm in length with a 5μm gap),and structures of a Psychodidae wing.

Real-Time Fraud Detection Using Machine Learning

Credit card fraud remains a significant challenge, with financial losses and consumer protection at stake. This study addresses the need for practical, real-time fraud detection methodologies. Using a Kaggle credit card dataset, I tackle class imbalance using the Synthetic Minority Oversampling Technique (SMOTE) to enhance modeling efficiency. I compare several machine learning algorithms, including Logistic Regression, Linear Discriminant Analysis, K-nearest Neighbors, Classification and Regression Tree, Naive Bayes, Support Vector, Random Forest, XGBoost, and Light Gradient-Boosting Machine to classify transactions as fraud or genuine. Rigorous evaluation metrics, such as AUC, PRAUC, F1, KS, Recall, and Precision, identify the Random Forest as the best performer in detecting fraudulent activities. The Random Forest model successfully identifies approximately 92% of transactions scoring 90 and above as fraudulent, equating to a detection rate of over 70% for all fraudulent transactions in the test dataset. Moreover, the model captures more than half of the fraud in each bin of the test dataset. SHAP values provide model explainability, with the SHAP summary plot highlighting the global importance of individual features, such as “V12” and “V14”. SHAP force plots offer local interpretability, revealing the impact of specific features on individual predictions. This study demonstrates the potential of machine learning, particularly the Random Forest model, for real-time credit card fraud detection, offering a promising approach to mitigate financial losses and protect consumers.
