CDYL1 fosters double-strand break-induced transcription silencing and prom.tes hom.logy-directed repair

Cells have evolved DNA dam.ge response （DDR） to repair DNA lesions and thus preserving genom.c stability and im.eding carcinogenesis. DNA dam.ge induction is accom.anied by transient transcription repression. Here, we describe a previously unrecognized role of chrom.dom.in Y-like （CDYL1） protein in fortifying double-strand break （DSB）-induced transcription repression and repair. We showed that CDYL1 is rapidly recruited to dam.ged euchrom.tic regions in a poly （ADP-ribose） polym.rase 1 （PARP1）-dependent, but ataxia telangiectasia m.tated （ATm.-independent, m.nner. While the C-term.nal region, containing the enoyl-CoA hydratase like （ECH） dom.in, of CDYL1 binds to poly （ADP-ribose）（PAR） m.ieties and m.diates CDYL1 accum.lation at DNA dam.ge sites, the chrom.dom.in and histone H3 trim.thylated on lysine 9 （H3K9m.3） m.rk are dispensable for its recruitm.nt. Furtherm.re, CDYL1 prom.tes the recruitm.nt of enhancer of zeste hom.log 2 （EZH2）, stim.lates local increase of the repressive m.thyl m.rk H3K27m.3, and prom.tes transcription silencing at DSB sites. In addition, following DNA dam.ge induction, CDYL1 depletion causes persistent G2/m.arrest and alters H2AX and replication protein A （RPA2） phosphorylation. Rem.rkably, the ‘traffic-light reporter’ system.revealed that CDYL1 m.inly prom.tes hom.logy-directed repair （HDR） of DSBs in vivo. Consequently, CDYL1-knockout cells display synthetic lethality with the chem.therapeutic agent, cisplatin. Altogether, our findings identify CDYL1 as a new com.onent of the DDR and suggest that the HDR-defective ‘BRCAness’ phenotype of CDYL1-deficient cells could be exploited for eradicating cancer cells harboring CDYL1 m.tations.

Salacca zalacca(Gaertn.) Voss(fam.ly Arecaceae) is the snake fruit com.only known in m.lay language as salak in m.laysia. This exotic fruit has diverse and potential pharm.cological properties due to its high antioxidant content. It is often consum.d due to its sweet taste. The abundant natural sugar and fibre along with m.nerals and vitam.n m.kes it a nutritious fruit. Phytochem.cal investigation on this fruit has revealed the presence of flavonoids, phenolics, glycosides as well as som. volatile and arom.tic com.ounds, including gallic acid, quercetin, chlorogenic acid, epicatechin, proanthocyanidins, lycopene and β-carotene. Pharm.cological studies on the fruit flesh and peel have shown som. trem.ndous antioxidant, anti-inflam.atory, anticancer and antidiabetic potential. This review provides the botanical inform.tion of Salacca zalacca as well as its scientific investigations involving the distinct pharm.cological and phytochem.cal benefits. This could help in highlighting the lacking data and research gaps on this plant.

目的探讨合并阻塞性睡眠呼吸暂停综合征（obstructivesleepapneasyndrom.，OSAS）的2型糖尿病患者的肿瘤坏死因子d（tum.rnecrosisfactor-α，TNF-α）、凝血酶原激活物抑制物（***）变化情况，并探讨其与睡眠中低氧血症发病的相关性。... 详细信息

目的探讨合并阻塞性睡眠呼吸暂停综合征（obstructivesleepapneasyndrom.，OSAS）的2型糖尿病患者的肿瘤坏死因子d（tum.rnecrosisfactor-α，TNF-α）、凝血酶原激活物抑制物（***）变化情况，并探讨其与睡眠中低氧血症发病的相关性。方法病例组为住院的2型糖尿病患者共54例，分为伴有OSAS者（O组）27例和不伴有OSAS者（N组）27例。对照组（C组）来自于同期体检且无内分泌疾病及OSAS的患者（n=26）。ELISA分析3组血清TNF-α、PAI的水平。多导睡眠初筛仪监测睡眠呼吸紊乱指数（AHI）、夜间最低脉搏血氧饱和度（LSpO2）等指标。并探讨TNF-α、PAI与稳态模型评估的胰岛素抵抗指数（HOm.—IR）、空腹血糖、睡眠呼吸紊乱指数（AHI）、夜间最低脉搏血氧饱和度（LSpO2）、年龄相关性。分别以TNF-α、PAI为因变量，以空腹血糖、HOm.-IR、AHI、LSpO2、年龄为自变量进行多元逐步直线回归分析。结果O组的HOm.-IR、TNF-α、PAI[分别为2．69±1．53、（1．97±0．13）ng／m.和（2．27±0．85）ng／m.]均高于其余2组，且N组的这3项指标[分别为1．70±0．88、（1．09±0．29）ng／m.和（1．59±0．13）ng／m.]均高于C组『分别为1．15±0．67、（0．73±0．19）ng／m.和（0．89±0．25）ng／m.]。2组糖尿病的空腹血糖高于C组。O组的AHI高于N组及C组，夜间LSpO，低于N组及C组。单因素Spearm.n相关分析显示TNF-α、PAI均与HOm.—IR、空腹血糖、AHI呈正相关，与夜间LSpO，呈负相关。多元逐步直线回归分析显示影响TNF-α的因素依次是AHI、空腹血糖；影响PAI的因素依次是AHI、HOm.—IR。结论合并OSAS的2型糖尿病患者的慢性低活度炎症状态更加严重，干预OSAS可能有助于2型糖尿病慢性并发症的防治。

Objective:To evaluate the antim.crobial potency of Rosm.rinus eriocalyx(***)essential oil and total polyphenols against pathogenic ***:Antim.crobial activity of *** extracts was assessed by disc diffusion m.thod and m.nim.m.inhibitory concentrations *** oil obtained from.endem.c rosem.ry by hydrodistillation was analysed by gas chrom.tograph/retention index and gas chrom.tograph-m.ss ***:An interesting antim.crobial activity was shown by *** ***,constituted m.inly by flavonoids,were the m.st effective extract with very low m.nim.m.inhibitory concentrations values,ranged between 0.06 and 8.00 m./m.,while essential oil was less *** should be noted that antim.crobial activities of both *** extracts were m.re directed against fungi and Gram.positive bacteria than Gram.negative ones,in which Staphylococcus aureus,Enterococcus faecalis,and Candida albicans were the m.st sensitive *** chem.cal com.osition of *** essential oil,cam.hor(37.8%),1,8-cineole(17.4%),cam.hene(13.3%),and α-pinene(10.9%)were the m.jor ***:The findings of the present study indicate that *** extracts possess significant bactericidal and fungicidal *** of its richness in essential oil,and especially flavonoids,*** m.y be a source for effective and safe antim.crobial agents.
