The incidence o. breast cancer is still increasing, and with impro.ed cancer treatment, mo.e wo.en live lo.ger with the side effects o. their treatment. The respo.se o. no.mal tissue to.radiatio. co.tinues fo. years after the treatment is co.pleted. The influence o. radio.herapy o. the o.tco.e o. breast reco.structtive surgery remains unpredictable. The co.binatio. o. two.surgical sites o. which o.e is previo.sly irradiated, is rarely enco.ntered in humans and thus co.piles a unique o.po.tunity to.study the implicatio.s o. irradiatio. fo.lo.ed by surgery. The aim o. this study was to.examine the lo.g-term effect o. radiatio. therapy o. the pro.eins expressed in the wo.nd tissue after a breast reco.structio.. Ten patients were included in the study, all treated with radio.herapy after a mastecto.y and breast reco.structio. with a co.tralateral pedicled TRAM flap. Expanded po.y-tetrafluo.etylene po.ymer tubes were implanted fo. 10 days, subcutaneo.sly, belo. the inframammary fo.d and belo. the do.o. site. The pro.ein fro. the newly synthesized granulatio. tissue in the tubes was extracted and analyzed fo. differences in pro.ein expressio. with 2D gel electro.ho.esis and mass spectro.etry. A to.al o. 676 pro.eins were detected;o. these, 4 pro.eins changed significantly and were successfully identified. TPM4 and APo.4 fro. the radiatio. treated tissue were sho.n to.be significantly decreased, whereas IGKC and VDAC1 were fo.nd to.be significantly increased. The pro.eo.ic technique co.bined with the ePTFE tube wo.nd mo.el can elucidate so.e o. the mo.ecular alteratio.s in the wo.nd healing induced by radiatio. therapy. The pro.ein mo.ificatio.s o. TPM4, APo.4, IGKC and VDAC1 may influence the cell pro.iferatio., apo.to.is and the inflammatio. o. the tissue repair pro.ess.
