High mobility group box-1 protein as a therapeutic target in perinatal hypoxic-ischemic brain injury

Perinatal hypoxic-ischemic(HI)brain injury is a leading cause of morbidity and longstanding disability in newborns(Millar et al.,2017).Improved neonatal intensive care has increased survival in infants with pregnancy and birth related ***,many surviving neonates exhibit neurological abnormalities that can persist throughout life(Millar et al.,2017).Early neuroprotective strategies have the potential to improve neurological outcomes and attenuate developmental delay in ***,hypothermia is the only currently approved intervention for HI encephalopathy in fullterm infants,which is only partially protective(Millar et al.,2017).Findings in preterm and full-term infants suggest that elevations in proinflammatory cytokines are important in the pathogenesis of HI-related brain injury(Millar et al.,2017).The high mobility group box-1(HMGB1),a representative damage associatedmolecular pattern(DAMP)protein,has been reported to be implicated in a variety of brain related inflammatory diseases including traumatic brain injury,epilepsy,and stroke(Nishibori et al.,2019).Anti-HMGB1 therapies have gained increasing interest to treat inflammation related disorders in the brain(Nishibori et al.,2019).However,there is a paucity of information regarding the pathology of HMGB1 in HI-related brain injury during the perinatal *** current perspective discusses the potential contributions of HMGB1 to HI-related brain injury during the perinatal period and also addresses the potential of HMGB1 as a therapeutic target of the brain ***,this perspective emphasizes the potential for combinational therapeutics for hypothermia with anti-HMGB1 monoclonal antibodies(mAb)in perinatal HI brain injury.

Rbfox1 controls alternative splicing of focal adhesion genes in cardiac muscle cells

Alternative splicing is one of the major cellular processes that determine the tissue-specific expression of protein ***,it remains challenging to identify physiologically relevant and tissue-selective proteins that are generated by alternative ***,we investigated the target spectrum of the splicing factor Rbfox1 in the cardiac muscle context in more *** using a combination of in silico target prediction and in-cell validation,we identified several focal adhesion proteins as alternative splicing targets of *** focused on the alternative splicing patterns of vinculin(metavinculin isoform)and paxillin(extended paxillin isoform)and identified both as potential Rbfox1 *** analyses suggested that both isoforms are promoted by Rbfox1 due to binding in the *** adhesions play an important role in the cardiac muscle context,since they mainly influence cell shape,cytoskeletal organization,and cell–matrix *** data confirmed that depletion of Rbfox1 changed cardiomyoblast morphology,cytoskeletal organization,and multinuclearity after differentiation,which might be due to changes in alternative splicing of focal adhesion ***,our results indicate that Rbfox1 promotes alternative splicing of focal adhesion genes in cardiac muscle cells,which might contribute to heart disease progression,where downregulation of Rbfox1 is frequently observed.

Dear Editor,Afferent synapses of cochlear inner hair cells(IHCs)employ a unique molecular machinery(see extended background in supplementary Materials).Otoferlin is a key player in this machinery and its defects cause human auditory synaptopathy(Moser and starr,2016).Otoferlin,a tail-anchored(Vogl et al.,2016)multi-C_(2)-domain protein(Fig.1Ai)specific to hair cells(Roux et al.,2006),is a member of the ferlin protein family involved in membrane trafficking and repair that are of major disease relevance(Pangršičet al.,2012),also see supplementary *** is distributed broadly within IHCs(Fig.2Ai-Aiii;Pangrsic et al.,2010;Roux et al.,2006).

DFT studies of Electronic Properties and Effect of He and Xe Incorporation in selected Ceramics

The electronic properties of several prospective nuclear fuels are not yet well known. We used Quantum Espresso and EPW codes to evaluate the electron density of states, the electronic heat capacity coefficient, the electron-phonon coupling strength, the number of mobility electrons, and the electronic heat conductivity. The electronic properties for ThN, ThC and UN using a slightly different approach that were previously evaluated are discussed and the results are compared. We confirmed that while the electronic heat capacity coefficient is linearly dependent on the electron density of states at Fermi energy, such a simple relation could not be used to determine the difference in the electronic heat conductivity of investigated materials. The highest heat conductivity was registered in ThN. These metallic fuels also have high U/Th density, therefore are more economical since enrichment is expensive. Furthermore, it is important to examine swelling in these high-density fuels. We evaluated that UN had 42% more U atoms per unit volume than UO2 and a 55% higher volume increase when accommodating one Xe atom in one interstitial of a (2 × 2 × 2) supercell. However, for He, the volume increase was 27% lower in UN. Interestingly, even though the Th atom’s density in ThN and ThC was lower than that of U atoms in the UN compound, a similar trend of volume changes was found. We concluded, therefore, that when we consider swelling, the local structural symmetry (tetrahedral versus octahedral sites) is more important than the density of atoms. The 37 % greater of absolute value of the total energy increase due to incorporation of Xe in ThC versus ThN cannot be explained by the crystal structure since a ThC-Xe supercell has a higher lattice constant than a ThN-Xe corresponding supercell. such results can only be explained by investigating electronic structure.
