A method for microbial decontamination of Acanthamoeba cultures using the peritoneal cavity of mice

Objective: To evaluate whether the inoculation of contaminated cultures in the peritoneal cavity of mice could implement decontamination of Acanthamoeba ***: Suspensions of Acanthamoeba, Acanthamoeba polyphaga ATCC 30461, or Acanthamoeba spp. isolated from soil(Un B13 strain) were inoculated in the peritoneal cavity of Swiss mice(n = 24). After 1, 6, 12, or 24 h of exposure the peritoneal cavity was washed and assessed for the presence of bacteria, fungi, and ***: After 1 h of intraperitoneal inoculation at least 97% of the bacteria and 96% of the fungi(P < 0.05) and 99% of the bacteria(P < 0.05) were successfully eliminated from the ATCC 30461 strain and from the soil isolate Un B13 strain, respectively. This method also allowed the recovery of most trophozoites and cysts from both Acanthamoeba cultures at the end of 24 ***: Our data demonstrated that this technique has great potential for decontamination of Acanthamoeba cultures in a short period of time.

Inter- and intraobserver agreement in computed tomography enterography in inflammatory bowel disease

AIM To evaluate intra- and interobserver agreement in imaging features in inflammatory bowel disease and comparison with fecal calprotectin(FC) *** Our institutional computed tomography enterography(CTE) database was retrospectively queried to identify patients who underwent CTE from January 2014 to June 2015. Patient inclusion criteria were confirmed inflammatory bowel disease(IBD) and FC collected 1 pattern, or normal); and signs of active disease(present or absent). In 42 of 44 patients evaluated, routine CTE reports were made by one of the readers who reevaluated the CTEs ≥ 6 mo later, to determine the intraobserver agreement. FC was considered a sign of disease activity when it was higher than 250 μg/*** Forty-four patients with IBD(38 with Crohn's disease and 6 with ulcerative colitis) were included. There was a moderate interobserver agreement regarding localization of IBD(k = 0.540), type of disease(k = 0.410) and the presence of active signs in CTE(k = 0.419). There was almost perfect intraobserver agreement regarding localization, type and signs of active disease in IBD. The k values were 0.902, 0.937 and 0.830, respectively. After a consensus between both radiologists regarding inflammatory activity in CTE, we found that 24(85.7%) of 28 patients who were classified with active disease had elevated FC, and six(37.5%) of 16 patients without inflammatory activity in CTE had elevated FC(P = 0.003). The correlation between elevated FC and the presence of active disease in CTE was significant(k = 0.495, P = 0.001).CONCLUSION We found almost perfect intraobserver and moderate interobserve

Preparation of Fame by Microwave Irradiation Using Boron Trifluoride as a Catalyst

Fatty acid methyl ester (FAME) preparation is an important step to determine fatty acid composition of oils and fats. Transesterification with boron trifluoride (BF3) in methanol has been widely used;however, the transesterification of triacylglycerols usually involves a long heating time, whereas much shorter reaction times may be sufficient when microwave irradiation is used. The purpose of this paper was to optimize FAME preparation with BF3 by microwave irradiation. A three-factor central composite routable (CCRD) design was used to study the effect of the heating time (A), irradiation power (B), and volume of sodium hydroxide (C) on FAME concentration. A second-order polynomial model was employed to generate a surface response. Optimum conditions obtained for FAME preparation were time of 60 s, irradiation power of 220 W, and base volume of 1.5 mL. Compared to conventional methods, the advantages of this method are the quick result and the accuracy.

This study aimed to show that the physicochemical proprieties obtained by Fourier transform infrared spectroscopy （FTIR）, thermogravimetry （TG）, and scanning electronic microscopy （SEM） can be useful tools for evaluating the quality of active pharmaceutical ingredients （APIs） and pharmaceutical products. In addition, a simple, sensitive, and efficient method employing HPLC-DAD was developed for simulta- neous determination of lidocaine （LID）, ciprofloxacin （CFX） and enrofloxacin （EFX） in raw materials and in veterinary pharmaceutical formulations. Compounds were separated using a Gemini Cm （250 mm×4.6 mm, 5μm） Phenomenex column, at a temperature of 25℃, with a mobile phase containing 10 mM of phosphoric acid （pH 3.29）： acetonitrile （85.7：14.3, v/v） and a flow rate of 1.5 mL/min. Physicochemical characterization by TG, FTIR, and SEM of raw materials of LID, CFX, and EFX provided information useful for the evaluation, differentiation, and qualification of raw materials. Finally, the HPLC method was proved to be useful for evaluation of raw material and finished products, besides satisfying the need for an analytical method that allows simultaneous determination of EFX, CFX, and LID, which can also be extended to other matrices and applications.

Use of Intragastral Balloon and Increase in Fertility： Case Report

The IGB （intragastric balloon） is a silicone device, inflated by endoscopy in the gastric cavity, with physiological saline and a blue dye, up to the volume of 700 mL, and used for a maximum of 6 months, It is indicated for patients with BMI〉27 kg/m^2 that have not undergone previous gastric surgery. In obese women there is a decrease in peripheral resistance fertility, and the weight loss achieved in bariatric surgery improves fertility due to reduced subcutaneous fat, with lower insulin resistance. Similar results could be achieved through the use of IGB, a less invasive method. The objective of the article is to report the case of a patient who became pregnant using IGB, considering the hypothesis that the increase in fertility was due to weight loss and/or due to the improvement in the self-esteem （and sexual life） of patient. We selected 13 articles from PubMed, UpToDate, JAMA and a case report. Results： Balloon with drawal occurred on 05/02/16 after positive pregnancy test with final weight 65.5 kg and BMI 25.91 kg/m2. Conclusions： Weight loss actually favors fertility, but self-esteem may have been instrumental in improving the sex life, enabling conception.

Enteroparasitoses in Patients with Rheumatoid Arthritis

Background: Patients with rheumatic diseases have profound alterations in the immune system as a result of underlying diseases and the treatments used, which increases the risk of occurrence and severity of infections, including enteroparasitosis. The current treatment for rheumatoid arthritis involves immunosuppressive therapies powerfully needed for screening infectious processes. The prevalence of parasitic infections in rheumatoid arthritis patients is not currently documented in literature. The objective of the current study was to determine the prevalence of parasitic diseases in a population with RA. Methods: We collected demographic and socioeconomic data from 67 patients at the Hospital Universitário de Brasília from July 2015 to April 2016. All patients underwent a parasitological examination of their stool and multiple variables were analyzed using Poisson regression method. Results: The mean age of patients was 53.9 years. They were predominantly in women (94%) and caucasian (47.8%). The mean disease duration was 9.2 years and most patients had the disease in remission or light activity. The prevalence of parasitic infections in these patients was 11.9%, all cases being that of protozoa of the following species: Endolimax nana, Entamoeba histolytica and Entamoeba coli. The final multivariate analysis indicated that the presence of disease had a significant statistical relationship between the presence of enteroparasitosis and rheumatoid arthritis with no fatigue by VAS (p = 0.0488) and best current health index by VAS (p = 0.0012). Conclusion: This study indicates that the prevalence of enteroparasitoses in a population with Rheumatoid Arthritis was 11.9% and all cases were found of protozoa.

Synthesis, Characterization, and Study of PLGA Copolymer <i>in Vitro</i>Degradation

The poly(lactic-co-glycolic acid), known as PLGA, is one of the main bioreabsorbable polymers used in the field of medicine today. This copolymer is widely applied in sutures, devices geared toward the controlled release of medication, and the guided regeneration of bone tissue as it presents a short degradation time. This work aimed to synthesize the 82/18 PLGA (expressed by the mass ratio of D,L-lactide and glycolide, respectively), to characterize and study the in Vitro degradation in the form of rods in phosphate buffer solution (PBS). The copolymer was synthesized by opening the cyclic dimer rings of the monomers D,L-lactide and glycolide, in the presence of the tin octanoate initiator and of the lauryl alcohol co-initiator. The characterization of the copolymer and the follow-up of its in vitro degradation were studied using: Differential Scanning Calorimetry (DSC), Thermogravimetry (TG), Infrared Molecular Absorption Spectroscopy with Fourier Transform (FTIR), Rheometry, and Scanning Electron Microscopy (SEM). Through these characterization techniques, it was possible to obtain the glass transition temperature, thermal stability, chemical composition, morphology, and molar mass of both the synthesized and the degraded copolymer. The molar mass of the synthesized copolymer was, approximately, 106 g·mol-1. The degradation rate of PLGA significantly increased from the 19th to the 28th day in PBS. After 28 days in PBS, the glass transition temperature and the molar mass reduced from 45°C to 17°C and from 1.5 × 106 g·mol-1?to 7.5 × 104?g·mol-1, respectively. The pH of the medium has a significant influence on the copolymer degradation profile. When it diminishes, it accelerates the degradation process, resulting in smaller PLGA polymer chains. This pH dependent degradation can be useful for drug release systems.
