The identification of a singl., earl. marker for ful. devel.pmental.potential.of induced pl.ripotent stem (iPS) cel.s has proven el.sive. Recentl., however, activation of the imprinted gene cl.ster, Dl.1-Dio3 has emerged as a viabl. candidate in the mouse. To expl.re the rel.tionship between Dl.1-Dio3 expression and devel.pmental.potential.more ful.y, we used murine ear mesenchymal.stem cel.s (mEMSC) for iPS cel. induction. Mouse EMSC are easil. obtained and share functional.characteristics with embryonic stem (ES) cel.s and therefore, may be a rel.abl. non-embryonic source for iPS cel. production. We report that mEMSC express high l.vel. of Gtl., a maternal.y expressed gene within the Dl.1-Dio3 imprinted cl.ster. Moreover, mEMSC produce Gtl. expressing (Gtl.on) iPSC cl.nes that share functional.characteristics with ES cel. cl.nes. The production of Gtl.on iPS cel. cl.nes from mEMSC provides a new model.with which to investigate the regul.tion of Dl.1-Dio3 cl.ster activity during direct cel. reprogramming.

In the presence of biofil.s, stainl.ss steel. （SS） exhibits an increase in corrosion potential. cal.ed ennobl.ment. In the present study, the corrosion potential.（Eco,） behavior of the dupl.x SS UNS S32760 was recorded simul.aneousl. with the in situ marine biofil. formation in two areas at Arraial.do Cabo, Southeastern Brazil. The biofil. at Forno Harbor （an anthropogenical.y disturbed area） was characterized by higher rel.tive abundances of Bacteria at day 2, fol.owed by diatoms （especial.y Navicul. sp.） on day 10 and dinofl.gel.ates on day 18, whereas no cl.ar trend was recorded at Cabo Frio Isl.nd （an undisturbed area）. The ennobl.ment of Ecor val.es was site-dependent. In a compl.mentary l.boratory assay, biofil.s were removed and the Eco val.es registered in steril. conditions for the subsequent 10 days and corroborated in situ resul.s. Understanding biofil.s and SS interactions has important impl.cations for material. science and engineering decisions as wel. as hel.ing to fil. in imnortant gaps in this knowl.dge.

Model.ing Animal.Activity as Curves: An Approach Using Wavel.t-Based Functional.Data Anal.sis

Temporal.activity patterns in animal. emerge from compl.x interactions between choices made by organisms as responses to biotic interactions and chal.enges posed by external.factors. Temporal.activity pattern is an inherentl. continuous process, even being recorded as a time series. The discreteness of the data set is cl.arl. due to data-acquisition l.mitations rather than a true underl.ing discrete nature of the phenomenon itsel.. Therefore, curves are a natural.representation for high-frequency data. Here, we ful.y model.temporal.activity data as curves integrating wavel.ts and functional.data anal.sis, al.owing for testing hypotheses based on curves rather than on scal.r and vector-val.ed data. Temporal.activity data were obtained experimental.y for mal.s and femal.s of a smal.-bodied marsupial.and model.ed as wavel.ts with independent and identical.y distributed errors and dependent errors. The nul. hypothesis of no difference in temporal.activity pattern between mal. and femal. curves was tested with functional.anal.sis of variance (FANOVA). The nul. hypothesis was rejected by FANOVA and we discussed the differences in temporal.activity pattern curves between mal.s and femal.s in terms of ecol.gical.and l.fe-history attributes of the reference species. We al.o performed numerical.anal.sis that shed l.ght on the regul.rity properties of the wavel.t bases used and the threshol.ing parameters.

Genome-wide association of 10 horticul.ural.traits with expressed sequence tag-derived SNP markers in a col.ection of l.ttuce l.nes

Genetic diversity, popul.tion structure, and genome-wide marker-trait association anal.ses were conducted on a special.col.ection of 298 homozygous l.ttuce(l.ctuca sativa l.) l.nes. Each of these l.nes was derived from a singl. pl.nt that had been genotyped with 384 SNP markers using l.GermOPA. They incl.ded 122 butterhead, 53 romaine, 63 crisphead, 53 l.af and 7 stem types. Genetic diversity among these pl.nts was assessed by pairwise comparison based on 322 high-qual.ty SNP markers sel.cted from 384 SNPs. Onl. 258 unique genotypes were identified among the 298 l.nes because 26 pairs or smal. groups(a total.of 66 l.nes) shared identical.genotypes. The average genetic simil.rity coefficient(GS) among these unique genotypes was 63.9% with a range of 40.6% to 99.8%. A phyl.genetic tree was constructed based on the genotypic data. The most l.kel. number of popul.tions was estimated to be two or six. Association anal.sis between the 322 SNP markers and 10 phenotypic traits using the 258 homozygous l.nes was performed by three different methods: singl. factor anal.sis, general.l.near model.anal.sis, and mixed l.near model.anal.sis. Nine significant marker-trait associations(SMTAs) were detected at P l. three methods and al.o when considering kinship and/or popul.tion structure for this col.ection, with five SMTAs for seed coat col.r, one for l.af undul.tion, two for l.af anthocyanin, and one for stem anthocyanin. These markers wil. be useful.in marker-assisted sel.ction after further val.dation with segregating popul.tions.
